    Sexuated Topology and the Suspension of Meaning: A Non-Hermeneutical Phenomenological Approach to Textual Analysis

    This study assumes the subject's pursuit of meaning is generally incapacitating and should be suspended. It aims to demonstrate how such a suspension is theoretically accomplished by utilizing Lacan's formulae of sexuation integrated with his work in discourse theory and topology. Part I places this study into context by examining scholarship from the established fields of hermeneutics, phenomenology, (post)structuralism, aesthetic theory and psychoanalysis in order to extract out their respective theory of meaning. These theories reveal that an historical struggle with meaning has been underway since the Reformation and reaches near crisis proportions in the 20th century. On the one hand this crisis is mollified by the rise of Heideggerian-Gadamerian hermeneutical phenomenology which questions traditional epistemological approaches to the text using a new ontological conceptualization of meaning and a conscious rejection of methodology. On the other hand this crisis is exacerbated when the ubiquitous nature of meaning is itself challenged by (post)structuralism's discovery of the signifier which inscribes a limit to meaning, and by the domains of sense and nonsense newly opened up by aesthetic theory. These historical developments culminate in the field of psychoanalysis which most consequentially delimits a cause of meaning said to be closely linked to the core of subjectivity. Part II extends these findings by rigorously constructing out of the Lacanian sexuated formulae a decidedly non-hermeneutical phenomenological approach useful in demonstrating the sexual nature of meaning. Explicated in their static state by way of an account of their original derivation from the Aristotelian logical square, it is argued that these four formulae are relevant to basic concerns of textual theory inclusive of the hermeneutical circle of meaning. These formulae are then set into motion by integrating them with Lacan's four discourses to demonstrate the breakdown of meaning. Finally, the cuts and sutures of two-dimensional space that is topology as set down in L'Ă©tourdit are performed to confirm how the very field of meaning is ultimately suspended from a nonsensical singular point known in Lacanian psychoanalysis as objet a. The contention is that by occupying this point the subject frees himself from the debilitating grip of meaning

    Machine Learning-Based Estimation and Goodness-of-Fit for Large-Scale Confirmatory Item Factor Analysis

    I investigate novel parameter estimation and goodness-of-fit (GOF) assessment methods for large-scale confirmatory item factor analysis (IFA) with many respondents, items, and latent factors. For parameter estimation, I extend Urban and Bauer's (2021) deep learning algorithm for exploratory IFA to the confirmatory setting by showing how to handle user-defined constraints on loadings and factor correlations. For GOF assessment, I explore new simulation-based tests and indices. In particular, I consider extensions of the classifier two-sample test (C2ST), a method that tests whether a machine learning classifier can distinguish between observed data and synthetic data sampled from a fitted IFA model. The C2ST provides a flexible framework that integrates overall model fit, piece-wise fit, and person fit. Proposed extensions include a C2ST-based test of approximate fit in which the user specifies what percentage of observed data can be distinguished from synthetic data as well as a C2ST-based relative fit index that is similar in spirit to the relative fit indices used in structural equation modeling. Via simulation studies, I first show that the confirmatory extension of Urban and Bauer's (2021) algorithm produces more accurate parameter estimates as the sample size increases and obtains comparable estimates to a state-of-the-art confirmatory IFA estimation procedure in less time. I next show that the C2ST-based test of approximate fit controls the empirical type I error rate and detects when the number of latent factors is misspecified. Finally, I empirically investigate how the sampling distribution of the C2ST-based relative fit index depends on the sample size.Master of Art

    Developmental Acquisition of Regulomes Underlies Innate Lymphoid Cell Functionality

    Innate lymphoid cells (ILCs) play key roles in host defense, barrier integrity, and homeostasis and mirror adaptive CD4(+) T helper (Th) cell subtypes in both usage of effector molecules and transcription factors. To better understand the relationship between ILC subsets and their Th cell counterparts, we measured genome-wide chromatin accessibility. We find that chromatin in proximity to effector genes is selectively accessible in ILCs prior to high-level transcription upon activation. Accessibility of these regions is acquired in a stepwise manner during development and changes little after in vitro or in vivo activation. Conversely, dramatic chromatin remodeling occurs in naive CD4(+) T cells during Th cell differentiation using a type-2-infection model. This alteration results in a substantial convergence of Th2 cells toward ILC2 regulomes. Our data indicate extensive sharing of regulatory circuitry across the innate and adaptive compartments of the immune system, in spite of their divergent developing pathways

    A composite reference section for terminal Proterozoic strata of southern Namibia

    Integrated sequence stratigraphic and chemostratigraphic data yield a framework for correlations of stratigraphic units in the terminal Proterozoic to Cambrian Witvlei and Nama Groups of Namibia. Coupled with precise U-Pb zircon age constraints, these correlations make it possible to construct a composite reference section for use in calibrating terminal Proterozoic chronostratigraphy. The Namibian reference section starts with two distinct glacial horizons and extends up to within 1 million years of the Proterozoic-Cambrian boundary. The two glacial horizons may represent each of two distinct Varanger-age glaciations better known from the North Atlantic region. From the higher of the two glacial horizons up, the composite stratigraphy preserves one of the thickest and most complete available records of carbon-isotope variability in post-Varanger terminal Proterozoic seawater. Four carbon-isotope chemostratigraphic intervals are recognized: (1) a postglacial negative δ^(13)C excursion (Npg interval); (2) a rising interval (Pr interval) of increasing positive delta 13 C values; (3) a falling interval (Pf interval) characterized by decreasing positive δ^(13)C and culminating in near zero or negative values; and (4) an interval of moderately positive, relatively invariant δ^(13)C values (I interval) that extends up to the unconformity that contains the Proterozoic-Cambrian boundary. Each of these chemostratigraphic intervals can be recognized in widely separated correlative sections around the world. By comparing sediment accumulation rate in the radiometrically calibrated Namibian stratigraphy with sediment accumulation rates in correlative sections in Arctic Canada and Oman, a maximum age of 564 Ma is estimated for the end of the younger Varanger glaciation, 25 m.y. younger than previous estimates
