    Comprehensive Evaluation of Corticospinal Tract Metabolites in Amyotrophic Lateral Sclerosis Using Whole-Brain 1H MR Spectroscopy

    Changes in the distribution of the proton magnetic resonance spectroscopy (MRS) observed metabolites N-acetyl aspartate (NAA), total-choline (Cho), and total-creatine (Cre) in the entire intracranial corticospinal tract (CST) including the primary motor cortex were evaluated in patients with amyotrophic lateral sclerosis (ALS). The study included 38 sporadic definite-ALS subjects and 70 age-matched control subjects. All received whole-brain MR imaging and spectroscopic imaging scans at 3T and clinical neurological assessments including percentage maximum forced vital capacity (FVC) and upper motor neuron (UMN) function. Differences in each individual metabolite and its ratio distributions were evaluated in the entire intracranial CST and in five segments along the length of the CST (at the levels of precentral gyrus (PCG), centrum semiovale (CS), corona radiata (CR), posterior limb of internal capsule (PLIC) and cerebral peduncle (CP)). Major findings included significantly decreased NAA and increased Cho and Cho/NAA in the entire intracranial CST, with the largest differences for Cho/NAA in all the groups. Significant correlations between Cho/NAA in the entire intracranial CST and the right finger tap rate were noted. Of the ten bilateral CST segments, significantly decreased NAA in 4 segments, increased Cho in 5 segments and increased Cho/NAA in all the segments were found. Significant left versus right CST asymmetries were found only in ALS for Cho/NAA in the CS. Among the significant correlations found between Cho/NAA and the clinical assessments included the left-PCG versus FVC and right finger tap rate, left -CR versus FVC and right finger tap rate, and left PLIC versus FVC and right foot tap rate. These results demonstrate that a significant and bilaterally asymmetric alteration of metabolites occurs along the length of the entire intracranial CST in ALS, and the MRS metrics in the segments correlate with measures of disease severity and UMN function

    Dopamine Agonists and their risk to induce psychotic episodes in Parkinson's disease: a case-control study

    <p>Abstract</p> <p>Background</p> <p>Psychosis is rare in untreated patients with Parkinson's disease (PD) but the prevalence rises to 40% during dopaminergic treatment. So far, no systematic comparison of the psychogenic potential of different dopaminergic drugs had been performed.</p> <p>Methods</p> <p>Eighty PD patients with psychotic episodes were compared to an age-matched control group of PD patients without psychotic episodes (n = 120) in a cross-sectional retrospective study.</p> <p>Results</p> <p>We found a positive correlation between psychotic episodes and dementia, number of concomitant medication, and pergolide intake. Odds ratio calculation confirmed the association with dementia. With respect to dopaminergic treatment, pergolide showed the highest odds ratio, levodopa the lowest. An adjusted logistic regression model confirmed the strong association with psychotic episodes and pergolide and no association with levodopa (adjusted odds ratio 2.01 and 0.11, respectively).</p> <p>Conclusion</p> <p>The analysis indicates that dementia and concomitant medication are factors in PD associated with psychotic symptoms. Furthermore, different dopaminergic drugs showed markedly different associations with psychotic symptoms</p

    Old stones' song: Use-wear experiments and analysis of the Oldowan quartz and quartzite assemblage from Kanjera South (Kenya)

    Evidence of Oldowan tools by w2.6 million years ago (Ma) may signal a major adaptive shift in hominin evolution. While tool-dependent butchery of large mammals was important by at least 2.0 Ma, the use of artifacts for tasks other than faunal processing has been difficult to diagnose. Here we report on use-wear analysis ofw2.0 Ma quartz and quartzite artifacts from Kanjera South, Kenya. A use-wear framework that links processing of specific materials and tool motions to their resultant use-wear patterns was developed. A blind test was then carried out to assess and improve the efficacy of this experimental use-wear framework, which was then applied to the analysis of 62 Oldowan artifacts from Kanjera South. Usewear on a total of 23 artifact edges was attributed to the processing of specific materials. Use-wear on seven edges (30%) was attributed to animal tissue processing,corroborating zooarchaeological evidence for butchery at the site. Use-wear on 16 edges (70%)was attributed to the processing of plant tissues, including wood, grit-covered plant tissues that we interpret asunderground storage organs (USOs), and stems of grass or sedges. These results expand our knowledge of the suite of behaviours carried out in the vicinity of Kanjera South to include the processing of materials that would be ‘invisible’ using standard archaeological methods. Wood cutting and scraping may represent the production and/or maintenance of wooden tools. Use-wear related to USO processing extends the archaeological evidence for hominin acquisition and consumption of this resource by over 1.5 Ma. Cutting of grasses, sedges or reeds may be related to a subsistence task (e.g., grass seed harvesting, cutting out papyrus culm for consumption) and/or a non-subsistence related task (e.g., production of ‘twine,’ simple carrying devices, or bedding).     ASPECTOS MORFOANATÔMICOS DE FRUTOS DE TOMATEIRO CULTIVAR ÂNGELA GIGANTE, SUBMETIDOS A TRATAMENTOS COM REGULADORES VEGETAIS

    O presente trabalho teve como finalidade obter dados morfológicos de frutos de tomateiro, Lycopersicon esculentum Mill. `Ângela Gigante', submetidos à ação de reguladores vegetais, em dois ensaios que ocorreram em épocas distintas, em casa de vegetação. As mudas foram selecionadas e transplantadas para vasos com capacidade de 12 L de terra, contendo uma mistura de solo argiloso, areia, matéria orgânica e uma adubação mineral complementar de N, P, K. No segundo ensaio, após o transplante das mudas, além da adubação mineral complementar de N, P, K, efetuaram-se adubações adicionais (fertirrigação). Em ambos os ensaios, quando as plantas atingiram quatro folhas definitivas, realizaram-se as pulverizações com giberelina, GA3 50 mg/L; ácido naftalenacético, NAA 100 mg/L; cloreto (2-cloroetil) trimetilamônio, CCC 1.500 mg/L e ácido succínico -2,2 dimetil-hidrazida, SADH 3.000 mg/L. Em relação aos estudos anatômicos, observou-se que os tratamentos com retardadores vegetais (CCC e SADH) produziram frutos firmes, com formato tipo barril e ombros salientes; entretanto, em seção transversal, notou-se perda de viscosidade e atrofia de sementes, principalmente nos frutos de plantas tratadas com SADH. Os tratamentos com NAA e GA3 causaram eventual formação de frutos geminados. O tratamento com GA3 apresentou o parênquima do pericarpo com grãos de amido em processo de fragmentação, provavelmente em virtude de o GA3 acelerar a atividade da amilase, afetando o processo de maturação dos frutos e transformando o amido em açúcares. Notaram-se no mesocarpo células com grande quantidade de cristais de oxalato de cálcio sob a forma de areia cristalina. Do tratamento com CCC resultaram frutos suculentos com células da placenta degeneradas, deixando livre grande quantidade de mucilagem. O pericarpo apresentou grande quantidade de grãos de amido composto em toda a extensão, provavelmente por haver um atraso no processo de maturação dos frutos.<br>The objective of the present work was to observe anatomical and morphological aspects of fruits of Lycopersicon esculentum Mill.`Ângela Gigante', treated with plant growth regulators. Two assays were carried out in the greenhouse: (1) in the first, the seedlings were selected and transferred to 12 L capacity pots, containing a mixture of clayey soil, sand, organic matter and a complementary N, P, K mineral fertilizing; (2) in the second, after the seedling transfer, besides the N, P, K fertilizing, a supplementary fertirrigation was carried out. In both assays after the plants had developed four mature leaves, they were pulverized with GA3 50 mg/L; NAA 100 mg/L; CCC 1,500 mg/L and SADH 3,000 mg/L. The treatments with growth retardants (CCC and SADH) produced firm fruits, although in cross sections, viscosity loss and seed atrophy were observed in fruits of plants treated with SADH. Plants treated with NAA and GA3 showed the formation of fused fruits. The treatment with GA3 revealed starch grain in process of fragmentation in the parenchyma of the pericarp. The treatment with GA3 probably accelerated the process of ripening, transforming starch to sugars. Cells with crystal calcium oxalate in the mesocarp as a crystalline sand were observed. The treatment of CCC formed juicy fruits with degenerated placenta cells, liberating great amounts of mucilage. The pericarp showed great amounts of compound starch grain, probably due to retardation in fruit ripening
