© 2017 Ilinskaya, Ulyanova, Yarullina and Gataullin. Current studies of human gut microbiome usually do not consider the special functional role of transient microbiota, although some of its members remain in the host for a long time and produce broad spectrum of biologically active substances. Getting into the gastrointestinal tract (GIT) with food, water and probiotic preparations, two representatives of Bacilli class, genera Bacillus and Lactobacillus, colonize epithelium blurring the boundaries between resident and transient microbiota. Despite their minor proportion in the microbiome composition, these bacteria can significantly affect both the intestinal microbiota and the entire body thanks to a wide range of secreted compounds. Recently, insufficiency and limitations of pure genome-based analysis of gut microbiota became known. Thus, the need for intense functional studies is evident. This review aims to characterize the Bacillus and Lactobacillus in GIT, as well as the functional roles of the components released by these members of microbial intestinal community. Complex of their secreted compounds is referred by us as the "bacillary secretome." The composition of the bacillary secretome, its biological effects in GIT and role in counteraction to infectious diseases and oncological pathologies in human organism is the subject of the review

The article presents the analysis of disastrous mud loss achieved during casing at Dulisminsk oil and gas condensate field (DOGCF) and comparative evaluation of geological structure of DOGCF and Naryksko-Ostashkinsk area of Kemerovo oblast, where the same problem was solved successfully. On the basis of the analysis, an engineering solution is proposed to prevent mud loss in the conditions of DOGCF

Species identification of spider mites (Tetranychidae: Tetranychinae): a review of methods

    Get PDF
Spider mites (Acari: Tetranychidae) are dangerous pests of agricultural and ornamental crops, the most economically significant of them belonging to the genera Tetranychus, Eutetranychus, Oligonychus and Panonychus. The expansion of the distribution areas, the increased harmfulness and dangerous status of certain species in the family Tetranychidae and their invasion of new regions pose a serious threat to the phytosanitary status of agroand biocenoses. Various approaches to acarofauna species diagnosis determine a rather diverse range of currently existing methods generally described in this review. Identification of spider mites by morphological traits, which is currently considered the main method, is complicated due to the complexity of preparing biomaterials for diagnosis and a limited number of diagnostic signs. In this regard, biochemical and molecular genetic methods such as allozyme analysis, DNA barcoding, restriction fragment length polymorphism (PCR-RFLP), selection of species-specific primers and real-time PCR are becoming important. In the review, close attention is paid to the successful use of these methods for species discrimination in the mites of the subfamily Tetranychinae. For some species, e. g., the two-spotted spider mite (Tetranychus urticae), a range of identification methods has been developed – from allozyme analysis to loop isothermal amplification (LAMP), while for many other species a much smaller variety of approaches is available. The greatest accuracy in the identification of spider mites can be achieved using a combination of several methods, e. g., examination of morphological features and one of the molecular approaches (DNA barcoding, PCR-RFLP, etc.). This review may be useful to specialists who are in search of an effective system for spider mite species identification as well as when developing new test systems relevant to specific plant crops or a specific region

Development of mathematical models to control the technological properties of cement slurries

    Get PDF
Oil and gas producing enterprises are making increasingly high demands on well casing quality, including the actual process of injection and displacement of cement slurry, taking into account requirements for the annular cement level, eliminating possible hydraulic fracturing, with developing a hydraulic cementing program. It is necessary to prevent deep invasion of cement slurry filtrate into the formation to exclude bridging of productive layers. It is impossible to fulfill all these requirements at the same time without application of modifying additives; complex cement compositions are being developed and applied more often. Furthermore, need to adjust cement slurries recipes appears for almost every particular well. In order to select and justify cement slurries recipes and their prompt adjustment, taking into account requirements of well construction project, as well as geological and technical conditions for cementing casing strings, mathematical models of the main technological properties of cement slurries for cementing production casing strings in the Perm Region were developed. Analysis of the effect of polycarboxylic plasticizer (Pl) and a filtration reducer (fluid loss additive) based on hydroxyethyl cellulose (FR) on plastic viscosity (V), spreadability (S) and filtration (F) of cement slurries is conducted. Development of mathematical models is performed according to more than 90 measurements

Direct inhibition of oncogenic KRAS by Bacillus pumilus ribonuclease (binase)

© 2016 Elsevier B.V.RAS proteins function as molecular switches that transmit signals from cell surface receptors into specific cellular responses via activation of defined signaling pathways (Fang, 2015). Aberrant constitutive RAS activation occurs with high incidence in different types of cancer (Bos, 1989). Thus, inhibition of RAS-mediated signaling is extremely important for therapeutic approaches against cancer. Here we showed that the ribonuclease (RNase) binase, directly interacts with endogenous KRAS. Further, molecular structure models suggested an inhibitory nature of binase-RAS interaction involving regions of RAS that are important for different aspects of its function. Consistent with these models, phosphorylation analysis of effectors of RAS-mediated signaling revealed that binase inhibits the MAPK/ERK signaling pathway. Interestingly, RAS activation assays using a non-hydrolysable GTP analog (GTPγS) demonstrated that binase interferes with the exchange of GDP by GTP. Furthermore, we showed that binase reduced the interaction of RAS with the guanine nucleotide exchange factor (GEF), SOS1. Our data support a model in which binase-KRAS interaction interferes with the function of GEFs and stabilizes the inactive GDP-bound conformation of RAS thereby inhibiting MAPK/ERK signaling. This model plausibly explains the previously reported, antitumor-effect of binase specific towards RAS-transformed cells and suggests the development of anticancer therapies based on this ribonuclease

Background. Low-grade inflammation is currently one of the main risk factors for the development of adverse events in the cardiovascular system, including death from cardiovascular diseases or their complications that cause mortality in the postCOVID period. The aim of the study. To assess the impact of health resort treatment in the conditions of the Southern Coast of Crimea on clinical and functional parameters, as well as on the level of systemic inflammation in patients after a new coronavirus infection.Materials and methods. The study included 67 patients (54.9 ± 9.05 years) in the post-COVID period who underwent health resort treatment at the I.M. Sechenov Academic Research Institute of Physical Treatment Methods, Medical Climatology and Rehabilitation. Methods of health resort treatment included climatic treatment on the Southern Coast of Crimea, therapeutic breathing exercises, terrainkur, and various methods of respiratory therapy. In all patients, the study of the C-reactive protein (CRP) in peripheral blood upon admission and at discharge was carried out.Results. Despite a statistically significant improvement in most clinical and functional parameters, the level of CRP and the number of leukocytes and their subpopulations in peripheral blood in patients who underwent health resort treatment did not differ significantly (p > 0.05) from the baseline values obtained upon admission at the I.M. Sechenov Academic Research Institute of Physical Treatment Methods, Medical Climatology and Rehabilitation. The CRP index upon admission and at discharge corresponded to the lower limit of the interval specific for low-grade inflammation (from 3 to 10 mg/l).Conclusion. Our results indicate the lack of effectiveness of the presented plan of health resort treatment in the correction of low-intensity inflammation, as well as the necessity for deeper scientific research in the direction of studying the mechanisms of low-grade inflammation development and the methods of its management
