    Proton-Induced and Electron-Induced X-Ray Microanalysis of Insulin-Secreting Cells

    Elemental redistribution induced by insulin secretion, was investigated by electron and proton probe X-ray microanalysis. In particular, ion fluxes following immediately upon stimulation were studied. As the sensitivity of the electron probe was insufficient, the proton microprobe was employed. In order to see whether the cell is asymmetric with respect to Ca2+ influx, the cells were stimulated in the presence of Sr2+ (as a Ca2+ analog). Insulin-secreting cells (RINm5F cells and isolated mouse β-cells) were cultured on grids and shock-frozen at 2-30 seconds after stimulation. In a large number of cells, the major elements and and large fluxes were analyzed by the electron microprobe. In the proton microprobe, selected cells were analyzed and elemental maps were compared with electron micrographs of the same cells. The proton microprobe, but not the electron microprobe, could detect an influx of Sr in response to K+-stimulation for 2 seconds, in RINm5F-cells. No polarization of Sr2+ uptake in RINm5F-cells could be detected, and the β-cells did not respond to high K+ by uptake of Sr. Momentary stimulation of β-cells also resulted in a significant increase in Na, detected by the electron probe. Spreading of the β-cells on the substrate appears to influence the subcellular elemental distribution. Thus, the proton probe has potential to detect small changes in elements such as those occurring after short-time stimulation

    Is There a Need for Congruent Treatment Goals Between Alcohol-Dependent Patients and Caregivers?

    Background: Alcohol-dependent patients have different treatment goals when entering treatment. Furthermore, different treatment settings advocate different treatment goals. Earlier studies have pointed out that treatment goal is important for treatment outcome, both in the treatment setting as well as in the patients themselves. However, to our knowledge, no study has so far investigated the interaction between patient's goal and the goal of the treatment setting. The aim of the study was therefore to study the interaction between these 2 factors on treatment outcome. Methods: Patients' (n = 201) goals from 2 treatment settings one that had an abstinence-oriented goal and one with a low-risk drinking goal—were investigated. The patients were followed up 2.5 years after treatment entry and effectiveness of congruent treatment goals on treatment outcome was investigated. Results: There was no significant association between congruent goals and treatment outcomes (p = 0.060). However, when comparing the effectiveness of congruent treatment goal between the 2 treatment settings, the abstinence-oriented treatment setting was significantly more effective (p < 0.01). Conclusions: The major finding was that there appeared to be no association between congruence itself and treatment outcome. On the other hand, we found that the treatment outcome was more successful if the patient as well as the treatment setting had abstinence as a goal (i.e., congruent goals of abstinence)

    Spruce forest afforestation leading to increased Fe mobilization from soils

    Increasing exports of Fe and DOC from soils, causing browning of freshwaters, have been reported in recent decades in many regions of the northern hemisphere. Afforestation, and in particular an increase of Norway spruce forest in certain regions, is suggested as a driver behind these trends in water chemistry. In this study, we tested the hypothesis that the gradual accumulation of organic soil layers in spruce forests, and subsequent increase in organic acid concentrations and acidity enhances mobilization of Fe. First generation Norway spruce stands of different ages (35, 61, 90 years) and adjacent arable control plots were selected to represent the effects of aging forest. Soil solutions were sampled from suction lysimeters at two depths (below organic soil layer and in mineral soil) during two years, and analyzed for Fe concentration, Fe speciation (XAS analysis), DOC, metals, major anions and cations. Solution Fe concentrations were significantly higher in shallow soils under older spruce stands (by 5- and 6-fold) than in control plots and the youngest forest. Variation in Fe concentration was best explained by variation in DOC concentration and pH. Moreover, Fe in all soil solutions was present as mononuclear Fe(III)-OM complexes, showing that this phase is dominating Fe translocation. Fe speciation in the soil was also analyzed, and found to be dominated by Fe oxides with minor differences between plots. These results confirmed that Fe mobilization, by Fe(III)-OM complexes, was higher from mature spruce stands, which supports that afforestation with spruce may contribute to rising concentrations of Fe in surface waters

    Differences in soil properties in adjacent stands of Scots pine, Norway spruce and silver birch in SW Sweden

    Soil properties were compared in adjacent 50-year-old Norway spruce, Scots pine and silver birch stands growing on similar soils in south-west Sweden. The effects of tree species were most apparent in the humus layer and decreased with soil depth. At 20–30 cm depth in the mineral soil, species differences in soil properties were small and mostly not significant. Soil C, N, K, Ca, Mg, and Na content, pH, base saturation and fine root biomass all significantly differed between humus layers of different species. Since the climate, parent material, land use history and soil type were similar, the differences can be ascribed to tree species. Spruce stands had the largest amounts of carbon stored down to 30 cm depth in mineral soil (7.3 kg C m−2), whereas birch stands, with the lowest production, smallest amount of litterfall and lowest C:N ratio in litter and humus, had the smallest carbon pool (4.1 kg C m−2), with pine intermediate (4.9 kg C m−2). Similarly, soil nitrogen pools amounted to 349, 269, and 240 g N m−2 for spruce, pine, and birch stands, respectively. The humus layer in birch stands was thin and mixed with mineral soil, and soil pH was highest in the birch stands. Spruce had the thickest humus layer with the lowest pH

    Long-term social skills group training for children and adolescents with autism spectrum disorder: a randomized controlled trial

    Social skills group training (SSGT) is widely used for intellectually able children and adolescents with autism spectrum disorder (ASD). Previous studies indicate small to moderate effects on social communication capacities. The duration of most available programs is relatively short, and extended training might lead to further improvement. This randomized controlled trial compared an extended 24-week version of the SSGT program KONTAKT with standard care. The weekly sessions gradually shifted in content from acquisition of new skills to real-world application of the acquired skills. A total of 50 participants with ASD (15 females; 35 males) aged 8–17 years were included. The study was conducted at two child and adolescent psychiatry outpatient units in Sweden. The primary outcome was the Social Responsiveness Scale–Second Edition (SRS-2) rated by parents and blinded teachers. Secondary outcomes included parent- and teacher-rated adaptive behaviors, trainer-rated global functioning and clinical severity, and self-reported child and caregiver stress. Assessments were made at baseline, posttreatment, and at 3-months follow-up. Parent-rated SRS-2 scores indicated large effects posttreatment [- 19.2; 95% CI - 29.9 to - 8.5; p &lt; .001, effect size (ES) = 0.76], which were maintained at follow-up (- 20.7; 95% CI - 31.7 to - 9.7; p &lt; .0001, ES = 0.82). These estimates indicate substantially larger improvement than previously reported for shorter SSGT. However, the effects on teacher-rated SRS-2 and most secondary outcomes did not reach statistical significance. Our results suggest added benefits of extended SSGT training, implying that service providers might reach better results by optimizing the delivery of SSGT
