    Design of an Information System for optimizing the Programming of nursing work shifts

    Health institutions operate twenty-four hours a day, seven days a week. They face a demand that fluctuates daily. Unlike jobs with fixed hours and obligatory days off, in health, operational continuity is required. The allocation for nursing shifts generates a rotation of people for health services according to legal and casuistic guidelines. Assigning and planning shifts results in a workload that takes an average of five to six extra hours. Existing applications offer a partial solution because they do not consider the news and contingencies of a health service. A web application is presented that, given a list of nurses, historical shifts and restrictions, a work shift planning is generated. This application comes to support the current shift allocation method based on electronic spreadsheets. The development consists of two modules. The first module has a shift allocation algorithm developed in C ++ and the second module has a graphical interface. As a case study, a set of health services from Chile and Colombia was used. The services have a defined number of nurses, who work different shifts according to the role and need of the institution. The results obtained are similar to a historical one. La participación de psiquiatras en programas clínicos específicos como las unidades de trasplantes de órganos sólidos tiene fundamentaciones técnicas precisas dada la correlación entre síntomas psíquicos y las características clínicas específicas de las enfermedades médicas que requieren de este procedimiento. Cada vez hay una mayor preocupación en los clínicos y cirujanos en relación a los factores psíquicos que obstaculizan la adherencia a los tratamientos e indicaciones médicas tanto antes como después de efectuado el trasplante. La población de pacientes pre y post-trasplantados exhiben elevadas tasas de cuadros psiquiátricos específicos que ameritan un tratamiento coordinado multidisciplinario.     Immunotherapies have shown remarkable, albeit tumor-selective, therapeutic benefits in the clinic. Most patients respond transiently at best, highlighting the importance of understanding mechanisms underlying resistance. Herein, we evaluated the effects of the engineered immunocytokine PD1-IL2v in a mouse model of de novo pancreatic neuroendocrine cancer that is resistant to checkpoint and other immunotherapies. PD1-IL2v utilizes anti-PD-1 as a targeting moiety fused to an immuno-stimulatory IL-2 cytokine variant (IL2v) to precisely deliver IL2v to PD-1 &lt;sup&gt;+&lt;/sup&gt; T cells in the tumor microenvironment. PD1-IL2v elicited substantial infiltration by stem-like CD8 &lt;sup&gt;+&lt;/sup&gt; T cells, resulting in tumor regression and enhanced survival in mice. Combining anti-PD-L1 with PD1-IL2v sustained the response phase, improving therapeutic efficacy both by reprogramming immunosuppressive tumor-associated macrophages and enhancing T cell receptor (TCR) immune repertoire diversity. These data provide a rationale for clinical trials to evaluate the combination therapy of PD1-IL2v and anti-PD-L1, particularly in immunotherapy-resistant tumors infiltrated with PD-1 &lt;sup&gt;+&lt;/sup&gt; stem-like T cells

    Centralized Modularity of N-Linked Glycosylation Pathways in Mammalian Cells

    Glycosylation is a highly complex process to produce a diverse repertoire of cellular glycans that are attached to proteins and lipids. Glycans are involved in fundamental biological processes, including protein folding and clearance, cell proliferation and apoptosis, development, immune responses, and pathogenesis. One of the major types of glycans, N-linked glycans, is formed by sequential attachments of monosaccharides to proteins by a limited number of enzymes. Many of these enzymes can accept multiple N-linked glycans as substrates, thereby generating a large number of glycan intermediates and their intermingled pathways. Motivated by the quantitative methods developed in complex network research, we investigated the large-scale organization of such N-linked glycosylation pathways in mammalian cells. The N-linked glycosylation pathways are extremely modular, and are composed of cohesive topological modules that directly branch from a common upstream pathway of glycan synthesis. This unique structural property allows the glycan production between modules to be controlled by the upstream region. Although the enzymes act on multiple glycan substrates, indicating cross-talk between modules, the impact of the cross-talk on the module-specific enhancement of glycan synthesis may be confined within a moderate range by transcription-level control. The findings of the present study provide experimentally-testable predictions for glycosylation processes, and may be applicable to therapeutic glycoprotein engineering

    How do youth choose activities? Assessing the relative importance of the micro-selection mechanisms behind adolescent extracurricular activity participation

    We investigate the network micro-selection mechanisms responsible for patterns of high school student extracurricular activity (ECA) participation, with a particular focus on those that can lead to ethnoracial segregation. We identify six types of mechanisms by which students select into activities (e.g., peer influence, homophily), which we test using a unique longitudinal dataset that combines student surveys with yearbook data on ECA involvement. These contexts represent two ethnoracially diverse U.S. high schools involving 2403 students and over 200 different activities spanning two school years. Using a stochastic actor-oriented model for two-mode networks, we find support for the hypothesized activity selection mechanisms. Follow-up analyses convey the relative importance of different mechanisms and inform our discussion of how ECA participation patterns develop and possible sources of segregation

    Epigenetic Engineering of Ribosomal RNA Genes Enhances Protein Production

    Selection of mammalian high-producer cell lines remains a major challenge for the biopharmaceutical manufacturing industry. Ribosomal RNA (rRNA) genes encode the major component of the ribosome but many rRNA gene copies are not transcribed [1]–[5] due to epigenetic silencing by the nucleolar remodelling complex (NoRC) [6], which may limit the cell's full production capacity. Here we show that the knockdown of TIP5, a subunit of NoRC, decreases the number of silent rRNA genes, upregulates rRNA transcription, enhances ribosome synthesis and increases production of recombinant proteins. However, general enhancement of rRNA transcription rate did not stimulate protein synthesis. Our data demonstrates that the number of transcriptionally competent rRNA genes limits efficient ribosome synthesis. Epigenetic engineering of ribosomal RNA genes offers new possibilities for improving biopharmaceutical manufacturing and provides novel insights into the complex regulatory network which governs the translation machinery in normal cellular processes as well as in pathological conditions like cancer

    Occurrence of Squash yellow mild mottle virus and Pepper golden mosaic virus in Potential New Hosts in Costa Rica

    Leaf samples of Solanum lycopersicum, Capsicum annuum, Cucurbita moschata, Cucurbita pepo, Sechium edule and Erythrina spp. were collected. All samples were positive for begomoviruses using polymerase chain reaction and degenerate primers. A sequence of ~1,100 bp was obtained from the genomic component DNA-A of 14 samples. In addition, one sequence of ~580 bp corresponding to the coat protein (AV1) was obtained from a chayote (S. edule) leaf sample. The presence of Squash yellow mild mottle virus (SYMMoV) and Pepper golden mosaic virus (PepGMV) were confirmed. The host range reported for SYMMoV includes species of the Cucurbitaceae, Caricaceae and Fabaceae families.     New and noteworthy bird records from the east slope of the andes of colombia

    We present significant new information on the distribution and status of 138 speciesof birds from the Andean East Slope of Colombia, based upon fieldwork between1990 and 2000 at 28 sites from central Dpto. Boyacá south to the Ecuador border.The first Colombian specimens of two species (Campylopterus villaviscensio, NapoSabrewing; Myrmotherula spodionota, Foothill Antwren) are reported. Three othertaxa (Ocreatus underwoodii addae, Piculus leucolaemus leucolaemus andMyiophobus p. phoenicomitra), the first two sometimes considered species distinctfrom known Colombian forms, represent first reports from Colombia based uponsightings or photographs; we add several more sightings of two species (Pipreolachlorolepidota, Iridisornis analis) previously known from single sight records. Inall, we report 35 species from the Andean East Slope of Colombia for the first time,southward range extensions on this slope for 47 species, northward extensions for21, upward or downward altitudinal extensions for 19, filling in of major discontinuitiesin distribution for 22; for ten of the latter, known from very few reports, new informationindicates a continuous distribution and far greater abundance than previouslysupposed. Range extensions and previously undiscovered populations of severalrestricted-range, Vulnerable, Near-threatened and Threatened species help toemphasize the importance of implementing conservation measures in the face of the increasing colonization pressures and insecurity. Based on our observations, wepresent several recommendations for setting conservation priorities in this rich butstill relatively poorly known region
