    Chromatin regulation and the histone code in HIV latency

    The formation of a latent reservoir of Human Immunodeficiency Virus (HIV†) infection hidden from immune clearance remains a significant obstacle to approaches to eradicate HIV infection. Towards an understanding of the mechanisms of HIV persistence, there is a growing body of work implicating epigenetic regulation of chromatin in establishment and maintenance of this latent reservoir. Here we discuss recent advances in the field of chromatin regulation, specifically in our understanding of the histone code, and how these discoveries relate to our current knowledge of the chromatin mechanisms linked to HIV transcriptional repression and the reversal of latency. We also examine mechanisms unexplored in the context of HIV latency and briefly discuss current therapies aimed at the induction of proviral expression within latently infected cells. We aim to emphasize that a greater understanding of the epigenetic mechanisms which govern HIV latency could lead to new therapeutic targets for latency reversal and clearance cure strategies

    Validity and Reliability of Firefighting Simulation Test Performance

    To assess the validity and reliability of a firefighting simulation test (FFST). Sixty-nine operational firefighters completed a best-effort FFST on one occasion and twenty-two participants completed a further FFST. All participants completed a maximal treadmill test to determine cardiorespiratory fitness (VO2max). Time to complete the FFST demonstrated a strong inverse relationship with VO2max (r = -0.73), although the prediction error was high. Reliability of the FFST was high (r = 0.84, p = 0.01), demonstrating a coefficient of variation of 4.5%. The FFST demonstrated reasonable validity as a surrogate assessment of cardiorespiratory fitness for firefighting. The FFST also demonstrated good reliability. Given the apparent magnitude of the prediction error, the FFST would be best used as a training tool, rather than as a primary means of assessing cardiorespiratory fitness for firefighting

    A web-based dietary intervention in early pregnancy and neonatal outcomes: A randomized controlled trial

    Background Maternal nutrition is a determinant of pregnancy outcomes. Few studies have evaluated the potential of online nutrition resources to modify behaviour. This randomized controlled trial aimed to determine whether access to a customized evidence-based nutrition website in pregnancy improved neonatal outcomes. Methods Women \u3c18 weeks gestation were recruited at their convenience. The control group received standard care. In addition to standard care, the intervention group received access to an evidence-based nutrition website, customized to the preferences of pregnant women. Results Of the 250 women, there were no differences in characteristics between the two groups. Of the women, 91.0% reported they make a conscious effort currently to eat a healthy diet. However, only 19.6% met dietary requirements for calcium, 13.2% for iron, 2.7% for folate and 2.3% for iodine. The most popular website section was pregnancy nutrition advice but engagement was not sustained. Access to the website was not associated with any improvement in clinical outcomes (P \u3e 0.05). Conclusions We found that provision of a customized website providing nutrition information, did not improve neonatal outcomes. Future studies should explore whether redesign with website interactivity or embedding information on popular digital platforms sustains women’s engagement and modifies dietary behaviour

    Black holes from high-energy beam--beam collisions

    Using a recent technique, proposed by Eardley and Giddings, we extend their results to the high-energy collision of two beams of massless particles, i.e. of two finite-front shock waves. Closed (marginally) trapped surfaces can be determined analytically in several cases even for collisions at non-vanishing impact parameter in D\ge 4 space-time dimensions. We are able to confirm and extend earlier conjectures by Yurtsever, and to deal with arbitrary axisymmetric profiles, including an amusing case of ``fractal'' beams.     Axion-photon Couplings in Invisible Axion Models

    We reexamine the axion-photon couplings in various invisible axion models motivated by the recent proposal of using optical interferometry at the ASST facility in the SSCL to search for axion. We illustrate that the assignment of U(1)PQU(1)_{PQ} charges for the fermion fields plays an important role in determining the couplings. Several simple non-minimal invisible axion models with suppressed and enhanced axion-photon couplings are constructed, respectively.     Renal pericytes: regulators of medullary blood flow

    Regulation of medullary blood flow (MBF) is essential in maintaining normal kidney function. Blood flow to the medulla is supplied by the descending vasa recta (DVR), which arise from the efferent arterioles of juxtamedullary glomeruli. DVR are composed of a continuous endothelium, intercalated with smooth muscle-like cells called pericytes. Pericytes have been shown to alter the diameter of isolated and in situ DVR in response to vasoactive stimuli that are transmitted via a network of autocrine and paracrine signalling pathways. Vasoactive stimuli can be released by neighbouring tubular epithelial, endothelial, red blood cells and neuronal cells in response to changes in NaCl transport and oxygen tension. The experimentally described sensitivity of pericytes to these stimuli strongly suggests their leading role in the phenomenon of MBF autoregulation. Because the debate on autoregulation of MBF fervently continues, we discuss the evidence favouring a physiological role for pericytes in the regulation of MBF and describe their potential role in tubulo-vascular cross-talk in this region of the kidney.     X-ray Spectroscopy and Variability of AGN Detected in the 2 Ms Chandra Deep Field-North Survey

    We investigate the nature of the faint X-ray source population through X-ray spectroscopy and variability analyses of 136 AGN detected in the 2 Ms Chandra Deep Field-North survey with > 200 background-subtracted 0.5-8.0 keV counts [F(0.5-8.0 keV)=(1.4-200)e-15 erg cm^{-2} s^{-1}]. Our preliminary spectral analyses yield median spectral parameters of Gamma=1.61 and intrinsic N_H=6.2e21 cm^{-2} (z=1 assumed when no redshift available) when the AGN spectra are fitted with a simple absorbed power-law model. However, considerable spectral complexity is apparent (e.g., reflection, partial covering) and must be taken into account to model the data accurately. Moreover, the choice of spectral model (i.e., free vs. fixed photon index) has a pronounced effect on the derived N_H distribution and, to a lesser extent, the X-ray luminosity distribution. Ten of the 136 AGN (~7%) show significant Fe Kalpha emission-line features with equivalent widths in the range 0.1-1.3 keV. Two of these emission-line AGN could potentially be Compton thick (i.e., Gamma < 1.0 and large Fe Kalpha equivalent width). Finally, we find that 81 (~60%) of the 136 AGN show signs of variability, and that this fraction increases significantly (~80-90%) when better photon statistics are available.Comment: Submitted to Advances in Space Research for New X-ray Results from Clusters of Galaxies and Black Holes (Oct 2002; Houston, TX), eds.     Astrophysical Axion Bounds

    Axion emission by hot and dense plasmas is a new energy-loss channel for stars. Observational consequences include a modification of the solar sound-speed profile, an increase of the solar neutrino flux, a reduction of the helium-burning lifetime of globular-cluster stars, accelerated white-dwarf cooling, and a reduction of the supernova SN 1987A neutrino burst duration.
