    Restoration of spermatogenesis after transplantation of c-Kit positive testicular cells in the fowl

    Transplantation of male germ line cells into sterilized recipients has been used in mammals for conventional breeding as well as for transgenesis. We have previously adapted this approach for the domestic chicken and we present now an improvement of the germ cell transplantation technique by using an enriched subpopulation of c-Kit-positive spermatogonia as donor cells. Dispersed c-Kit positive testicular cells from 16 to 17 week-old pubertal donors were transplanted by injection directly into the testes of recipient males sterilized by repeated gamma irradiation. We describe the repopulation of the recipient’s testes with c-Kit positive donor testicular cells, which resulted in the production of functional heterologous spermatozoa. Using manual semen collection, the first sperm production in the recipient males was observed about nine weeks after the transplantation. The full reproduction cycle was accomplished by artificial insemination of hens and hatching of chickens

    Evaluation of the slag regime and desulphurization of steel with synthetic slag containing Cr2O3

    The paper focuses on an evaluation of the slag regime and steel desulphurisation in a ladle with the help of synthetic slags based on Al2O3 containing various mass amounts of Cr2O3 varying from 0.3 % to 3.0 %. The aim of the plant experiments was to assess the achieved results when using two types of synthetic slag focusing on the course of desulphurisation, an analysis of the achieved chemical composition of the slag and an evaluation of the influence of oxide Cr2O3 on the increase in the chromium amounts in steel during secondary metallurgy. During the evaluation of the slag regime in the ladle, samples of steel from various technological nodes for a determination of the desulphurisation degree and the achieved chromium amounts in steel were taken. Samples of slags for the evaluation of the achieved chemical composition and amounts of Cr2O3 in the slag were also taken. The results mentioned in this paper represent the basic information about the possibilities of applying synthetic slags with increased amounts of Cr2O3 within secondary metallurgy.Web of Science4896597

    Biomechanical assessment of cannulated nails for the treatment of proximal femur fractures

    This article focuses on a type of surgical implant used in orthopaedics and traumatology-cannulated femoral nails. Femoral nails are used in medical treatment for purposes of osteosynthesis, i.e., when treating various types of complicated fractures, in this case fractures of the femur. The article investigates cases in which a nail has been implanted in the proximal part of the femur for a short time (with the fracture still not healed), compared with cases in which the bone has already healed. According to AO classification, examined fractures are described as AO 31B3 AO 32A3. The main focus is on strength-deformation analysis using the finite element method (FEM), which makes it possible to determine the behaviour of the femur-implant system. FEM analysis was used to compare 1.4441 steel nails made by two manufacturers, Medin (Czech Republic) and Tantum (Germany). Boundary conditions including external loading, prescribed supports and elastic foundation are defined. There were solved FEM analyses for five cases of healed femur and five cases of broken femur both including implants with prescribed collo-diaphyseal angles. The results of the analysis were used to assess stress-deformation states from the perspective of appropriateness for clinical treatment, biomechanical reliability and safety. All examined femoral nails are compared, safe and suitable for patient treatment.Web of Science1215art. no. 747

    Mechanics of screw joints solved as beams placed in a tangential elastic foundation

    This article deals with a new original analytical solution of deformation, force and stress states in wood screw joints up to the limit values of pulling out/breaking the screw. The screws are under tension. The wood-to-screw interaction is effectively simplified by introducing several physical model variants using a tangential elastic non-linear foundation. The experimental verification of the proposed models using pull-out tests (i.e., pulling out screws from dry spruce wood in laboratory conditions) confirms the correctness of the proposed models of the elastic linear/non-linear foundation. The validity of the model is also analytically and experimentally verified in the biomechanical model of pulling out screws from the femur of a bovine/human cadaver, which confirms and expands the validity of newly designed screw joint models outside the timber structure area.Web of Science1112art. no. 561

    Cílem diplomové práce bylo navrhnout metodiku poskytování outsourcingu v oblasti tiskových a reprografických služeb z pohledu společnosti, která outsourcing poskytuje. . Navržená metodika reflektuje proces outsourcingu formou projektu. Práce se skládá ze čtyř částí. První část se zabývá obecným vysvětlením pojmu outsourcing, jeho přínosy, výhodami a nevýhodami. Druhá část se věnuje zhodnocení současného stavu outsourcingu tiskových služeb ve firmě Konica Minolta a navržení zlepšení, kterých je možno dosáhnout. Ve třetí části je vypracován návrh metodiky poskytování outsourcingu v Konica Minolta. V poslední, čtvrté části, je pro přiblížení situace na trhu a pro možnost srovnání uvedena nabídka služeb největších konkurentů společnosti

    Transplantation of male germ cells into sterilized recipients has been widely used in mammals for conventional breeding and transgenesis purposes. This study presents a workable approach for germ cell transplantation between male chickens. Testicular cells from adult and prepubertal donors were dispersed and transplanted by injection directly into the testes of recipient males sterilized by repeated gamma irradiation. We describe the repopulation of the recipient seminiferous epithelium up to the production of heterologous sperm in about 50% of transplanted males. In comparison to males transplanted with testicular cell preparations from adult donors, in which the first ejaculates with sperm were recovered about 5 wk after transfer, a substantial interval (about 10 wk) was necessary to obtain ejaculates after the transfer of testicular cells from prepubertal donors. However, in both cases, recipient males produced ejaculates capable of fertilizing ova and producing progeny expressing donor genes
