    Estimation of the Postmortem Duration of Mouse Tissue by Electron Spin Resonance Spectroscopy

    Electron spin resonance (ESR) method is a simple method for detecting various free radicals simultaneously and directly. However, ESR spin trap method is unsuited to analyze weak ESR signals in organs because of water-induced dielectric loss (WIDL). To minimize WIDL occurring in biotissues and to improve detection sensitivity to free radicals in tissues, ESR cuvette was modified and used with 5,5-dimethtyl-1-pyrroline N-oxide (DMPO). The tissue samples were mouse brain, hart, lung, liver, kidney, pancreas, muscle, skin, and whole blood, where various ESR spin adduct signals including DMPO-ascorbyl radical (AsA∗), DMPO-superoxide anion radical (OOH), and DMPO-hydrogen radical (H) signal were detected. Postmortem changes in DMPO-AsA∗ and DMPO-OOH were observed in various tissues of mouse. The signal peak of spin adduct was monitored until the 205th day postmortem. DMPO-AsA∗ in liver (y = 113.8–40.7 log (day), R1 = −0.779, R2 = 0.6, P < .001) was found to linearly decrease with the logarithm of postmortem duration days. Therefore, DMPO-AsA∗ signal may be suitable for detecting an oxidation stress tracer from tissue in comparison with other spin adduct signal on ESR spin trap method

    We investigated with 17 cases of deaf children in born, wearing Cochlear Implants and Having habilitation for 7 years 8 months on the average by the clinical-audiologists using auditoryoral method, for the Speech-language and cognitive-neuropsychological ability of 6 years. It became clear that the achievement level of them were various and had individual differences, so one could catch up normal children in age 3 years, the other had severe retardation in age 9 years. (1) We researched on the factors effected the individual differences, especially concerned about the developmental retardation by using the checklists of Mori. And we got the following results ; the late age of wearing Cochlear Implants, the shortage of habilitation period before the entrance of primary school, the problems of parents (e.g., the difficulty of home training), and in relation for these, the poverty of systematic, continuous and gradual Speech-language habilitation, and also the lack of attention, the retardation of Performance Intelligence Quotient. (2) It was suggested that we should educate Japanese Speech-Therapists intensively who can be the specialists in the field of habilitation for the hearing-impaired children

    Development of diffuse large B-cell lymphoma in a patient with Waldenström's macroglobulinemia/lymphoplasmacytic lymphoma: clonal identity between two B-cell neoplasms

    Waldenström's macroglobulinemia (WM)/ lymphoplasmacytic lymphoma (LPL) is an indolent mature B-cell neoplasm. In rare cases of WM/LPL, diffuse large B-cell lymphoma (DLBCL) develops as a result of histologic transformation. In this report, we present a case of DLBCL developing in a patient with WM/LPL. Combination chemotherapy for DLBCL was effective and complete remission was eventually achieved. We attempted to determine the clonal relatedness between WM/LPL and DLBCL in the patient by analyzing complementarity-determining region 3 (CDR3) in the immunoglobulin heavy chain gene. A common CDR3 sequence was found in tumor cells of DLBCL and those of WM/LPL, indicating that tumor cells of DLBCL are clonally identical to those of WM/LPL. Therefore, in the present case, DLBCL is developed from WM/LPL cells by clonal evolution

    Evidence of mature adipocyte proliferation regulated by proliferin

    Despite much research, whether mature adipocytes proliferate remains controversial. Here, we examined 5-bromo-2&#x2032;-deoxyuridine (BrdU)-labelling of mature adipocytes. Although BrdU incorporation into subcutaneous adipocytes was less than that in visceral adipocytes, pioglitazone (Pio) treatment increased BrdU incorporation in subcutaneous, but not visceral, adipocytes in rats. Fully differentiated 3T3-L1 adipocytes exhibited an increase in cell number and BrdU incorporation with time, with this increase enhanced by Pio treatment. We therefore screened for genes that encode growth factors regulated by Pio, and selected proliferin (PLF). Both gene silencing of PLF by small interfering RNA and treatment with anti-PLF antibody suppressed proliferation in 3T3-L1 adipocytes. In adipocytes isolated from Pio-treated rats, the tissue-specific pattern of PLF expression was similar to that of BrdU incorporation. Administration of an anti-PLF antibody to mice reduced BrdU incorporation into adipocytes. Mature adipocytes thus have the ability to replicate, and this proliferation is positively regulated by PLF

    本研究の目的は,看護職者のキャリアディベロップメント支援のための継続教育プログラムの構築の基礎資料を得るために,A県下の看護職者のがん看護に関する学習状況やキャリア志向の実態を明らかにすることである.A県下病床数100床以上の外・内科を併設する病院17施設の看護職者1017名中686名(回収率67.5%)を分析対象とした.結果,がん看護に関する学習項目全てにおいて学習ニードが高かった.また,がん看護の学習機会を持つ者や学習の阻害要因はないと回答した者が多かった.職業的満足感では“看護の仕事にやりがいを感じている”“仕事上での目標がある”と回答した者が多く,“今の看護の仕事に満足している”とした者は約半数であった.がんに関する資格取得希望者は少なかった.がん看護に関する学習ニードと対象者の背景および学習機会,阻害要因,キャリア志向との関係については,学習機会があると回答した方が学習ニードの合計得点の平均値が高かった(P<0.05).また,阻害要因があると回答した方が学習ニードの合計得点の平均値が高かった(P<0.05).職業的満足感では“看護の仕事にやりがいを感じている”で〈そう思う群〉の方が学習ニードの合計得点の平均値が高かった(P<0.05).“仕事上での目標がある”では〈そう思う群〉の方が学習ニードの合計得点の平均値が高かった(P<0.05).“今の看護の仕事に満足している”では〈思わない群〉の方が学習ニードの合計得点の平均値が高かった(P<0.05).資格取得希望者の方が学習ニードの合計得点の平均値が高かった(P<0.05).以上より,がん看護に関する学習ニードの高さが示された.がん看護に関するキャリアディベロップにおいて,施設側と教育側の連携と学習ニードを充実させていくことが鍵となることが示唆された.The objective of this study was to elucidate the actual conditions of learning needs for cancer nursing and career-intended mind among nursing staff in Prefecture A in order to obtain basic information for establishing a continuing educational programs as support for developing a career of nursing staff. A survey was conducted on 1017 nurses working at one of 17 hospitals with ≥100 beds in Prefecture A that have both a department of surgery and a department of internal medicine. Answers from 686 nurses (response rate, 67.5%)were analyzed. The results indicated that there was a high level of learning needs in all learning items regarding cancer nursing. In addition, many nurses answered that they had opportunities to learn cancer nursing or that they had no obstacles to learn. Regarding job satisfaction, many nurses answered that they “feel worthwhile in nursing work” and “have objectives in their jobs” and approximately half of them answered that they “are satisfied with current nursing work”. Few nurses had a desire to acquire qualifications for being engaged in a high level of cancer care. In comparison of average scores of learning needs, nurses who answered there was a learning opportunities have higher scores than total average scores(P<0.05). In addition, learning need was high the one where had a hindrances to learning(P<0.05). As for the learning needs regarding cancer nursing and the relations of the career intention, learning need was higher Agree group in “find nursing work rewarding” (P<0.05) and “have career objectives” (P<0.05). Furthermore, learning need was higher Disagree group in “are satisfied with current nursing work” (P< 0.05). Learning need was higher a qualification applicant (P<0.05). These findings indicate a high level of learning needs regarding cancer nursing. It is necessary for “career development regarding cancer nursing” to be filled up in cooperation and learning need
