    The size and polydispersity of silica nanoparticles under simulated hot spring conditions

    The nucleation and growth of silica nanoparticles in supersaturated geothermal waters was simulated using a flow-through geothermal simulator system. The effect of silica concentration ([SiO2]), ionic strength (IS), temperature (T) and organic additives on the size and polydispersity of the forming silica nanoparticles was quantified. A decrease in temperature (58 to 33°C) and the addition of glucose restricted particle growth to sizes <20 nm, while varying [SiO2] or ISdid not affect the size (30-35 nm) and polydispersity (±9 nm) observed at 58°C. Conversely, the addition of xanthan gum induced the development of thin films that enhanced silica aggregation

    In order to determine the links between geochemical parameters controlling the formation of silica sinter in hot springs and their associated microbial diversity, a detailed characterisation of the waters and of in situ-grown silica sinters was combined with molecular phylogenetic analyses of the bacterial communities in Icelandic geothermal environments. At all but one site, the microorganisms clearly affected, and in part controlled, the formation of the macroscopic textures and structures of silica sinter edifices. In addition, the class and genera level phylogenetic diversity and distribution appeared to be closely linked to variations in temperature, salinity and pH regimes

    Controlled biomineralization of magnetite (Fe<sub>3</sub>O<sub>4</sub>) by <i>Magnetospirillum gryphiswaldense</i>

    Results from a study of the chemical composition and micro-structural characteristics of bacterial magnetosomes extracted from the magnetotactic bacterial strain Magnetospirillum gryphiswaldense are presented here. Using high-resolution transmission electron microscopy combined with selected-area electron diffraction and energy dispersive X-ray microanalysis, biogenic magnetite particles isolated from mature cultures were analysed for variations in crystallinity and particle size, as well as chain character and length. The analysed crystals showed a narrow size range (∼14-67 nm) with an average diameter of 46±6.8 nm, cuboctahedral morphologies and typical Gamma type crystal size distributions. The magnetite particles exhibited a high chemical purity (exclusively Fe3O4) and the majority fall within the single-magnetic-domain range

    As Geobase Information Systems increase in number, size and complexity, the format compatability of satellite remote sensing data becomes increasingly more important. Because of the vast and continually increasing quantity of data available from remote sensing systems the utility of these data is increasingly dependent on the degree to which their formats facilitate, or hinder, their incorporation into Geobase Information Systems. To merge satellite data into a geobase system requires that they both have a compatible geographic referencing system. Greater acceptance of satellite data by the user community will be facilitated if the data are in a form which most readily corresponds to existing geobase data structures.     Modeling user mobility via user psychological and geographical behaviors towards point of-interest recommendation

The pervasive employments of Location-based Social Network call for precise and personalized Point-of-Interest (POI) recommendation to predict which places the users prefer. Modeling user mobility, as an important component of understanding user preference, plays an essential role in POI recommendation. However, existing methods mainly model user mobility through analyzing the check-in data and formulating a distribution without considering why a user checks in at a specific place from psychological perspective. In this paper, we propose a POI recommendation algorithm modeling user mobility by considering check-in data and geographical information. Specifically, with check-in data, we propose a novel probabilistic latent factor model to formulate user psychological behavior from the perspective of utility theory, which could help reveal the inner information underlying the comparative choice behaviors of users. Geographical behavior of all the historical check-ins captured by a power law distribution is then combined with probabilistic latent factor model to form the POI recommendation algorithm. Extensive evaluation experiments conducted on two real-world datasets confirm the superiority of our approach over state-of-the-art methods

    Detection of changes in mitochondrial hydrogen sulfide (H2S) in vivo in the fish model Poecilia mexicana (Poeciliidae)

    In this paper, we outline the use of a mitochondria-targeted ratiometric mass spectrometry probe, MitoA, to detect in vivo changes in mitochondrial hydrogen sulfide (H2S) in Poecilia mexicana (family Poeciliidae). MitoA is introduced via intraperitoneal injection into the animal and is taken up by mitochondria, where it reacts with H2S to form the product MitoN. The MitoN/MitoA ratio can be used to assess relative changes in the amounts of mitochondrial H2S produced over time. We describe the use of MitoA in the fish species P. mexicana to illustrate the steps for adopting the use of MitoA in a new organism, including extraction and purification of MitoA and MitoN from tissues followed by tandem mass spectrometry. In this proof-of-concept study we exposed H2S tolerant P. mexicana to 59 µM free H2S for 5 h, which resulted in increased MitoN/MitoA in brain and gills, but not in liver or muscle, demonstrating increased mitochondrial H2S levels in select tissues following whole-animal H2S exposure. This is the first time that accumulation of H2S has been observed in vivo during whole-animal exposure to free H2S using MitoA

    Mitogenomes reveal two major influxes of Papuan ancestry across Wallacea following the last glacial maximum and Austronesian contact

    The tropical archipelago of Wallacea contains thousands of individual islands interspersed between mainland Asia and Near Oceania, and marks the location of a series of ancient oceanic voyages leading to the peopling of Sahul—i.e., the former continent that joined Australia and New Guinea at a time of lowered sea level—by 50,000 years ago. Despite the apparent deep antiquity of human presence in Wallacea, prior population history research in this region has been hampered by patchy archaeological and genetic records and is largely concentrated upon more recent history that follows the arrival of Austronesian seafarers ~3000–4000 years ago (3–4 ka). To shed light on the deeper history of Wallacea and its connections with New Guinea and Australia, we performed phylogeographic analyses on 656 whole mitogenomes from these three regions, including 186 new samples from eight Wallacean islands and three West Papuan populations. Our results point to a surprisingly dynamic population history in Wallacea, marked by two periods of extensive demographic change concentrated around the Last Glacial Maximum ~15 ka and post-Austronesian contact ~3 ka. These changes appear to have greatly diminished genetic signals informative about the original peopling of Sahul, and have important implications for our current understanding of the population history of the region.1.     Reduced dose of subcutaneous cladribine induces identical response rates but decreased toxicity in pretreated chronic lymphocytic leukaemia

    Purpose To study the efficacy and the safety of cladribine (2-chlorodeoxyadenosine, 2-CDA) administered as 24-hour infusions or as subcutaneous bolus injections at two different doses to patients with relapsing or refractory chronic lymphocytic leukaemia (CLL). Patients and methods In this non randomised 2-cohort study, 20 patients with pretreated CLL received cladribine at a dose of 0.7 mg/kg/cycle as continuous i.v. infusions over seven days (group 1) and 35 patients were treated at a reduced dose of 0.5 mg/kg/cycle given as s.c. bolus injections for five days (group 2). After two cycles of four week duration, response was assessed. In the case of progressive disease, therapy was discontinued, otherwise a maximum of four additional cycles were administered until best response. Results A total of 130 cycles were administered (group 1: 41, group 2: 89). Patient characteristics in both groups were comparable. The median dose intensities were 0.172 mg/kg per week and 0.123 mg/kg per week for groups 1 and 2, respectively (P ≤ 0.0001). The overall response rate for all 55 patients was 38% (95% confidence interval (95% CI): 25%-52%), with 5% CR and 33% PR. Response was similar in both patient groups (35% in group 1, 40% in group 2). No association between cladribine dose intensity and response rate was found, and there was no difference between patients relapsing after or refractory to previous therapies (11 of 24 vs. 10 of 31). Median remission duration was six months in both groups. Toxicity, in particular infections (all WHO grades, 34% in group 1 versus 7% in group 2) and myelosuppression (grade 1-4 neutropenia, 72% versus 41% of cladribine cycles) were statistically significantly more frequent in group 1. Conclusion Cladribine is active in heavily pretreated patients with chronic lymphocytic leukaemias. Dose reduction by 29% led to similar response and remission duration, but to a significant decrease of myelotoxicity and risk of infection. Cladribine administered as s.c. bolus injections at 0.5 mg/kg per cycle is safe and this dose level should not be exceeded in this patient populatio
