    Quantum Enhancement of the Zero-Area Sagnac Interferometer Topology for Gravitational Wave Detection

    Only a few years ago, it was realized that the zero-area Sagnac interferometer topology is able to perform quantum nondemolition measurements of position changes of a mechanical oscillator. Here, we experimentally show that such an interferometer can also be efficiently enhanced by squeezed light. We achieved a nonclassical sensitivity improvement of up to 8.2 dB, limited by optical loss inside our interferometer. Measurements performed directly on our squeezed-light laser output revealed squeezing of 12.7 dB. We show that the sensitivity of a squeezed-light enhanced Sagnac interferometer can surpass the standard quantum limit for a broad spectrum of signal frequencies without the need for filter cavities as required for Michelson interferometers. The Sagnac topology is therefore a powerful option for future gravitational-wave detectors, such as the Einstein Telescope, whose design is currently being studied.Comment: 4 pages, 4 figure

    Glutathione reductase-catalyzed cascade of redox reactions to bioactivate potent antimalarial 1,4-naphthoquinones--a new strategy to combat malarial parasites.

    Our work on targeting redox equilibria of malarial parasites propagating in red blood cells has led to the selection of six 1,4-naphthoquinones, which are active at nanomolar concentrations against the human pathogen Plasmodium falciparum in culture and against Plasmodium berghei in infected mice. With respect to safety, the compounds do not trigger hemolysis or other signs of toxicity in mice. Concerning the antimalarial mode of action, we propose that the lead benzyl naphthoquinones are initially oxidized at the benzylic chain to benzoyl naphthoquinones in a heme-catalyzed reaction within the digestive acidic vesicles of the parasite. The major putative benzoyl metabolites were then found to function as redox cyclers: (i) in their oxidized form, the benzoyl metabolites are reduced by NADPH in glutathione reductase-catalyzed reactions within the cytosols of infected red blood cells; (ii) in their reduced forms, these benzoyl metabolites can convert methemoglobin, the major nutrient of the parasite, to indigestible hemoglobin. Studies on a fluorinated suicide-substrate indicate as well that the glutathione reductase-catalyzed bioactivation of naphthoquinones is essential for the observed antimalarial activity. In conclusion, the antimalarial naphthoquinones are suggested to perturb the major redox equilibria of the targeted infected red blood cells, which might be removed by macrophages. This results in development arrest and death of the malaria parasite at the trophozoite stage

    Laparoscopic mesh-augmented hiatoplasty without fundoplication as a method to treat large hiatal hernias

    PURPOSE: Laparoscopic hiatal hernia repair with additional fundoplication is a commonly recommended standard surgical treatment for symptomatic large hiatal hernias with paraesophageal involvement (PEH). However, due to the risk of persistent side effects, this method remains controversial. Laparoscopic mesh-augmented hiatoplasty without fundoplication (LMAH), which combines hiatal repair and mesh reinforcement, might therefore be an alternative. METHODS: In this retrospective study of 55 (25 male, 30 female) consecutive PEH patients, the perioperative course and symptomatic outcomes were analyzed after a mean follow-up of 72 months. RESULTS: The mean DeMeester symptom score decreased from 5.1 to 1.8 (P < 0.001) and the gas bloating value decreased from 1.2 to 0.5 (P = 0.001). The dysphagia value was 0.7 before surgery and 0.6 (P = 0.379) after surgery. The majority of the patients were able to belch and vomit (96 and 92 %, respectively). Acid-suppressive therapy on a regular basis was discontinued in 68 % of patients. In 4 % of patients, reoperation was necessary due to recurrent or persistent reflux. A mesh-related stenosis that required endoscopic dilatation occurred in 2 % of patients. CONCLUSIONS: LMAH is feasible, safe and provides an anti-reflux effect, even without fundoplication. As operation-related side effects seem to be rare, LMAH is a potential treatment option for large hiatal hernias with paraesophageal involvement

    GPlates – Building a Virtual Earth Through Deep Time

    GPlates is an open‐source, cross‐platform plate tectonic geographic information system, enabling the interactive manipulation of plate‐tectonic reconstructions and the visualization of geodata through geological time. GPlates allows the building of topological plate models representing the mosaic of evolving plate boundary networks through time, useful for computing plate velocity fields as surface boundary conditions for mantle convection models and for investigating physical and chemical exchanges of material between the surface and the deep Earth along tectonic plate boundaries. The ability of GPlates to visualize subsurface 3‐D scalar fields together with traditional geological surface data enables researchers to analyze their relationships through geological time in a common plate tectonic reference frame. To achieve this, a hierarchical cube map framework is used for rendering reconstructed surface raster data to support the rendering of subsurface 3‐D scalar fields using graphics‐hardware‐accelerated ray‐tracing techniques. GPlates enables the construction of plate deformation zones—regions combining extension, compression, and shearing that accommodate the relative motion between rigid blocks. Users can explore how strain rates, stretching/shortening factors, and crustal thickness evolve through space and time and interactively update the kinematics associated with deformation. Where data sets described by geometries (points, lines, or polygons) fall within deformation regions, the deformation can be applied to these geometries. Together, these tools allow users to build virtual Earth models that quantitatively describe continental assembly, fragmentation and dispersal and are interoperable with many other mapping and modeling tools, enabling applications in tectonics, geodynamics, basin evolution, orogenesis, deep Earth resource exploration, paleobiology, paleoceanography, and paleoclimate

    One or two trainees per workplace in a structured multimodality training curriculum for laparoscopic surgery? Study protocol for a randomized controlled trial – DRKS00004675

    BACKGROUND: Laparoscopy training courses have been established in many centers worldwide to ensure adequate skill learning before performing operations on patients. Different training modalities and their combinations have been compared regarding training effects. Multimodality training combines different approaches for optimal training outcome. However, no standards currently exist for the number of trainees assigned per workplace. METHODS: This is a monocentric, open, three-arm randomized controlled trial. The participants are laparoscopically-naive medical students from Heidelberg University. After a standardized introduction to laparoscopic cholecystectomy (LC) with online learning modules, the participants perform a baseline test for basic skills and LC performance on a virtual reality (VR) trainer. A total of 100 students will be randomized into three study arms, in a 2:2:1 ratio. The intervention groups participate individually (Group 1) or in pairs (Group 2) in a standardized and structured multimodality training curriculum. Basic skills are trained on the box and VR trainers. Procedural skills and LC modules are trained on the VR trainer. The control group (Group C) does not receive training between tests. A post-test is performed to reassess basic skills and LC performance on the VR trainer. The performance of a cadaveric porcine LC is then measured as the primary outcome using standardized and validated ratings by blinded experts with the Objective Structured Assessment of Technical Skills. The Global Operative Assessment of Laparoscopic Surgical skills score and the time taken for completion are used as secondary outcome measures as well as the improvement of skills and VR LC performance between baseline and post-test. Cognitive tests and questionnaires are used to identify individual factors that might exert influence on training outcome. DISCUSSION: This study aims to assess whether workplaces in laparoscopy training courses for beginners should be used by one trainee or two trainees simultaneously, by measuring the impact on operative performance and learning curves. Possible factors of influence, such as the role of observing the training partner, exchange of thoughts, active reflection, model learning, motivation, pauses, and sympathy will be explored in the data analysis.     Hotspots of land use change in Europe

Yet, changes in the spatial patterns of land management intensity, and thus how they might relate to changes in the extent of land uses, remains unclear for many world regions.Wecompiled and analyzed high-resolution, spatiallyexplicit land-use change indicators capturing changes in both the extent and management intensity of cropland, grazing land, forests, and urban areas for all of Europe for the period 1990–2006. Based on these indicators, we identified hotspots of change and explored the spatial concordance of area versus intensity changes.Wefound a clear East–West divide with regard to agriculture, with stronger cropland declines and lower management intensity in the East compared to the West. Yet, these patterns were not uniform and diverging patterns of intensification in areas highly suitable for farming, and disintensification and cropland contraction in more marginal areas emerged. Despite the moderate overall rates of change, many regions in Europe fell into at least one land-use change hotspot during 1990–2006, often related to a spatial reorganization of land use (i.e., co-occurring area decline and intensification or co-occurring area increase and disintensification). Our analyses highlighted the diverse spatial patterns and heterogeneity of land-use changes in Europe, and the importance of jointly considering changes in the extent and management intensity of land use, as well as feedbacks among land-use sectors. Given this spatial differentiation of land-use change, and thus its environmental impacts, spatially-explicit assessments of land-use dynamics are important for context-specific, regionalized land-use policy making.Peer Reviewe
