    Does Nepal’s Climate Change Adaptation Policy and Practices Address Poor and Vulnerable Communities?

    Climate change (CC) impact, adaptation practices and policies are an emerging area of research and development in Nepal. Many households were deeply affected by, climatic variability and natural disasters coupled with a decade-long conflict, leaving them with few options to cope with CC in Nepal. The study was done through reviewed CC related policies and programs initiated at the national to local level.  Additionally, study was conducted to analyze different CC adaptation programs and practices in different eco-region from Himalaya to Inner Terai region of center and western Nepal through shared learning dialogue, field observation, key informant interview, regional and district level consultative workshop and focus group discussion. To address the issues of CC, different government and nongovernment institutions have initiated its adaptation programs in Nepal. NAPA and LAPA- Nepal highlighted the adaptation practices and adaptation framework from national to local level. These national programs initiatives have been endorsed by the GoN (Government of Nepal) through the promulgation of CC Policy Nepal 2011.  The policy has focused on interdisciplinary approach with 80% funds allocated at the local level particularly for poor and vulnerable groups. Some district level offices have initiated CC adaptation activities such as construction of water harvesting pond for irrigation, water source protection, agroforestry practices and training on CC adaptation and mitigation. Similarly, in some project supported districts, community level and VDC level five year local adaptation plans have been prepared. However, it is observed that there is a big gap in institutional arrangement, capacity to implement and mechanisms for adaptation programs at local level and poor communities. It was also observed that different GOs programs and NGOs were involved to improve the livelihood of the poor and vulnerable communities through pro-poor programs at local level which resulted improve CC adaptation capacity. It is suggested that CC policy in Nepal be formulated with holistic approach, institutionalized implementing mechanism and focus on livelihood improvement of the poor and vulnerable communities that have joint environmental, social or economic benefits.     Local people\u27s perception on Climate Change, its impact and adaptation practices in Himalaya to Terai regions of Nepal

    The study showed that average temperature has increased from 0.6 to 0.980 C over the last 30 years and precipitation is characterized by large inter-annual variability with substantial decrease in amount over the last five years. The annual temperature increasing trend followed 0.055\u3e0.0455\u3e0.035\u3e0.02 0C yr-1 from Middle Mountain, Siwlik, Himalayan and Teari regions respectively. More than 80 percent HHs responded that they have perceived increased temperature and expressed low amount snowfall in High mountain and rainfall in Mid mountain and Terai region over the last five years. Low amount of snow fall in the Himalayan region affected to the Nomad groups due low grass available to feed their livestock. Furthermore, local collector reported that there was lower availability of the medicinal plant such as Nirmasi and Jimbu due to low precipitation in the High mountain region. Similarly, more than 75 % respondent explored that monsoon has started one or two weeks later and increased the more number of drought days in the Mid-mountain and Terai. Study found that there was also decreased in water sources, ground water, and increased siltation and sedimentation in the downstream Terai region. Furthermore, change in flowering and fruiting time in some species, increased invade species like Agerative spp, Lantana camera in the farm land as well as forest land. Adaptation measures such as use of water source, community forest management, planting trees and grasses in the farm land, crop diversification were practiced by local people in their farm land as well as communal land. Natural resource degradation, poverty are already severe problems in this region, and there will be more severe problems in future if present scenario continues, particularly because small farmers do not have adequate resources to adopt to cope with CC impact. Study showed that Mid-mountain region is less vulnerable through climate change than other regions. Furthermore, no any policies and programs have been formulated for adaptation strateragy in this region. It is suggested that policy and program should formulate holistic approach and develop low cost technology for adaption to CC impact and improve livelihood of the local communities

    Spin-Polarized Transport Across an La0.7_{0.7}Sr0.3_{0.3}MnO3_{3}/YBa2_{2}Cu3_{3}O7_{7} Interface: Role of Andreev Bound States

    Transport across an La0.7_{0.7}Sr0.3MnO_{0.3}MnO_{3}/YBa2Cu_{2}Cu_{3}OO_{7}(LSMO/YBCO),interfaceisstudiedasafunctionoftemperatureandsurfacemorphology.Forcomparison,controlmeasurementsareperformedinnon−magneticheterostructuresofLaNiO(LSMO/YBCO), interface is studied as a function of temperature and surface morphology. For comparison, control measurements are performed in non-magnetic heterostructures of LaNiO_{3}$/YBCO and Ag/YBCO. In all cases, YBCO is used as bottom layer to eliminate the channel resistance and to minimize thermal effects. The observed differential conductance re ects the role of Andreev bound states in a-b planes, and brings out for the first time the suppression of such states by the spin-polarized transport across the interface. The theoretical analysis of the measured data reveals decay of the spin polarization near the LSMO surface with temperature, consistent with the reported photoemission data.Comment: 5 pages LaTeX, 3 eps figures included, accepted by Physical Review

    Evaluation of an Electricity-free, Culture-based Approach for Detecting Typhoidal Salmonella Bacteremia during Enteric Fever in a High Burden, Resource-limited Setting

    Background: In many rural areas at risk for enteric fever, there are few data on Salmonella enterica serotypes Typhi (S. Typhi) and Paratyphi (S. Paratyphi) incidence, due to limited laboratory capacity for microbiologic culture. Here, we describe an approach that permits recovery of the causative agents of enteric fever in such settings. This approach involves the use of an electricity-free incubator based upon use of phase-change materials. We compared this against conventional blood culture for detection of typhoidal Salmonella. Methodology/Principal Findings: Three hundred and four patients with undifferentiated fever attending the outpatient and emergency departments of a public hospital in the Kathmandu Valley of Nepal were recruited. Conventional blood culture was compared against an electricity-free culture approach. Blood from 66 (21.7%) patients tested positive for a Gram-negative bacterium by at least one of the two methods. Sixty-five (21.4%) patients tested blood culture positive for S. Typhi (30; 9.9%) or S. Paratyphi A (35; 11.5%). From the 65 individuals with culture-confirmed enteric fever, 55 (84.6%) were identified by the conventional blood culture and 60 (92.3%) were identified by the experimental method. Median time-to-positivity was 2 days for both procedures. The experimental approach was falsely positive due to probable skin contaminants in 2 of 239 individuals (0.8%). The percentages of positive and negative agreement for diagnosis of enteric fever were 90.9% (95% CI: 80.0%–97.0%) and 96.0% (92.7%–98.1%), respectively. After initial incubation, Salmonella isolates could be readily recovered from blood culture bottles maintained at room temperature for six months. Conclusions/Significance: A simple culture approach based upon a phase-change incubator can be used to isolate agents of enteric fever. This approach could be used as a surveillance tool to assess incidence and drug resistance of the etiologic agents of enteric fever in settings without reliable local access to electricity or local diagnostic microbiology laboratories.Boston Children's Hospital (Frederick H.     Breeding tomato (Solanum lycopersicum L.) for resistance to biotic and abiotic stresses

    Tomato (Solanum lycopersicum L.) is an important vegetable crop cultivated in the tropical and sub-tropical regions of the world. Low productivity in India is due to occurrence of both biotic and abiotic stresses. Among the biotic stresses, tomato leaf curl disease, bacterial wilt, early blight and Groundnut Bud Necrosis Virus disease have become serious production constraints causing considerable yield loss in the major tomato growing areas of the country. Adoption of multiple disease resistant varieties or F1 hybrids would be the most appropriate way to address these diseases. At ICAR-IIHR, Bengaluru systematic breeding strategies were employed to pyramid genes for resistance to early blight, bacterial wilt and tomato leaf curl diseases and to develop advanced breeding lines& F1 hybrids with triple disease resistance. Stable source of resistance to early blight and bi-partite begomo-virus (Tomato Leaf Curl New Delhi Virus) has been identified in Solanum habrochaites LA-1777. Validation with molecular markers linked to tomato leaf curl virus resistance revealed that LA-1777 carryTy2 and other putative resistant genes. Several high yielding dual purpose hybrids were also developed for fresh market and processing with high level of resistance to multiple diseases. Cherry tomato lines have also been bred for high TSS, total carotenoids, total phenols, flavonoids, vitamin C, acidity and lycopene content. IIHR-249-1, IIHR-2101 (Solanum habrochaites LA-1777), IIHR- 2866 and IIHR-2864 recorded high values for quality parameters like total carotenoids, lycopene, vitamin C, total phenols, flavonoids and TSS. Drought tolerant root stock has been developed by an interspecific cross between S. habrochaites LA-1777 and S. lycopersicum (15 SB SB). Resistant sources have also been identified against Tuta absoluta, a serious insect pest reported from major tomato growing areas in the country in recent time.     The Phosphodiesterase-5 Inhibitor Vardenafil Is a Potent Inhibitor of ABCB1/P-Glycoprotein Transporter

    One of the major causes of chemotherapy failure in cancer treatment is multidrug resistance (MDR) which is mediated by the ABCB1/P-glycoprotein. Previously, through the use of an extensive screening process, we found that vardenafil, a phosphodiesterase 5 (PDE-5) inhibitor significantly reverses MDR in ABCB1 overexpressing cancer cells, and its efficacy was greater than that of tadalafil, another PDE-5 inhibitor. The present study was designed to determine the reversal mechanisms of vardenafil and tadalafil on ABC transporters-mediated MDR. Vardenafil or tadalafil alone, at concentrations up to 20 µM, had no significant toxic effects on any of the cell lines used in this study, regardless of their membrane transporter status. However, vardenafil when used in combination with anticancer substrates of ABCB1, significantly potentiated their cytotoxicity in ABCB1 overexpressing cells in a concentration-dependent manner, and this effect was greater than that of tadalafil. The sensitivity of the parenteral cell lines to cytotoxic anticancer drugs was not significantly altered by vardenafil. The differential effects of vardenafil and tadalafil appear to be specific for the ABCB1 transporter as both vardenafil and tadalafil had no significant effect on the reversal of drug resistance conferred by ABCC1 (MRP1) and ABCG2 (BCRP) transporters. Vardenafil significantly increased the intracellular accumulation of [3H]-paclitaxel in the ABCB1 overexpressing KB-C2 cells. In addition, vardenafil significantly stimulated the ATPase activity of ABCB1 and inhibited the photolabeling of ABCB1 with [125I]-IAAP. Furthermore, Western blot analysis indicated the incubation of cells with either vardenafil or tadalafil for 72 h did not alter ABCB1 protein expression. Overall, our results suggest that vardenafil reverses ABCB1-mediated MDR by directly blocking the drug efflux function of ABCB1

    Current and Future Drug Targets in Weight Management

    Obesity will continue to be one of the leading causes of chronic disease unless the ongoing rise in the prevalence of this condition is reversed. Accumulating morbidity figures and a shortage of effective drugs have generated substantial research activity with several molecular targets being investigated. However, pharmacological modulation of body weight is extremely complex, since it is essentially a battle against one of the strongest human instincts and highly efficient mechanisms of energy uptake and storage. This review provides an overview of the different molecular strategies intended to lower body weight or adipose tissue mass. Weight-loss drugs in development include molecules intended to reduce the absorption of lipids from the GI tract, various ways to limit food intake, and compounds that increase energy expenditure or reduce adipose tissue size. A number of new preparations, including combinations of the existing drugs topiramate plus phentermine, bupropion plus naltrexone, and the selective 5-HT2C agonist lorcaserin have recently been filed for approval. Behind these leading candidates are several other potentially promising compounds and combinations currently undergoing phase II and III testing. Some interesting targets further on the horizon are also discussed
