    Does Starting School Before Labor Day Affect High School Retention and Graduation: Evidence From Virginia\u27s Kings Dominion Law

    Several states have required K-12 public schools to start after Labor Day in an effort to aid the tourism and hospitality industry. However, little is known about how these policies impact educational outcomes. We examine the impact of Virginia\u27s post-Labor Day school start law on high school retention and graduation rates. We use a difference-in-differences model to exploit exogenous variation in school division start dates. Our results show small differences of up to three weeks have little effect on high school dropout and graduation rates. Our findings inform the debate on post-Labor Day school start laws and compulsory attendance age cutoff laws

    Modeling and Comparing Vertical Density Profiles

    The vertical density profile of pressed wood panels is influenced by the manufacturing process and is important to panel end-users. Modeling the vertical density profile and making statistical comparisons among profiles resulting from different manufacturing treatments are critical to understanding and improving panel properties. Nonparametric regression analysis was used to model the vertical density profile of aspen (Populus tremuloides) oriented strandboard panels. A methodology is presented to compare vertical density profile curves. Twenty-seven laboratory panels were manufactured at 608 kg/m3 incorporating three levels of furnish moisture content (4%, 8%, 12%) and three levels of press closure rate (20 s, 60 s, 100 s) in a replicated, experimental design.The nonparametric regression technique called cubic splines was used to fit the data, R2 ranged from 0.985 to 0.998. Detailed discussion is presented that describes the method and interpretation of the nonparametric regression analysis. Statistical comparison of vertical density profile curves among treatment levels revealed that the 4% furnish moisture content level was significantly different (P = 0.015) from the 8% and 12% levels; the 8% level was not significantly different (P > 0.99) from the 12% level. Vertical density profile curves for all press closure rate treatments were significantly different (P < 0.003)

    Absence of force suppression in rabbit bladder correlates with low expression of heat shock protein 20

    BACKGROUND: Nitroglycerin can induce relaxation of swine carotid artery without sustained reductions in [Ca(2+)](i )or myosin regulatory light chain (MRLC) phosphorylation. This has been termed force suppression and been found to correlate with ser(16)-phosphorylation of heat shock protein 20 (HSP20). We tested for the existence of this mechanism in a smooth muscle that is not responsive to nitric oxide. METHODS: Isometrically mounted mucosa free rabbit bladder strips were contracted with carbachol and relaxed with 8-Br-cGMP, forskolin, or isoprenaline. RESULTS: Contraction was associated with a highly cooperative relation between MRLC phosphorylation and force such that very small increases in MRLC phosphorylation induced large increases in force. Relaxation induced by 8-Br-cGMP, forskolin, or isoprenaline did not shift the MRLC phosphorylation-force relation from that observed with carbachol alone, i.e. there was no force suppression. HSP20 content was negligible (approximately two hundred-fold less than swine carotid). CONCLUSION: The lack of force suppression in the absence of HSP20 is consistent with the hypothesized role for HSP20 in the force suppression observed in tonic smooth muscles

    DeepAMR for predicting co-occurrent resistance of Mycobacterium tuberculosis.

    MOTIVATION: Resistance co-occurrence within first-line anti-tuberculosis (TB) drugs is a common phenomenon. Existing methods based on genetic data analysis of Mycobacterium tuberculosis (MTB) have been able to predict resistance of MTB to individual drugs, but have not considered the resistance co-occurrence and cannot capture latent structure of genomic data that corresponds to lineages. RESULTS: We used a large cohort of TB patients from 16 countries across six continents where whole-genome sequences for each isolate and associated phenotype to anti-TB drugs were obtained using drug susceptibility testing recommended by the World Health Organization. We then proposed an end-to-end multi-task model with deep denoising auto-encoder (DeepAMR) for multiple drug classification and developed DeepAMR_cluster, a clustering variant based on DeepAMR, for learning clusters in latent space of the data. The results showed that DeepAMR outperformed baseline model and four machine learning models with mean AUROC from 94.4% to 98.7% for predicting resistance to four first-line drugs [i.e. isoniazid (INH), ethambutol (EMB), rifampicin (RIF), pyrazinamide (PZA)], multi-drug resistant TB (MDR-TB) and pan-susceptible TB (PANS-TB: MTB that is susceptible to all four first-line anti-TB drugs). In the case of INH, EMB, PZA and MDR-TB, DeepAMR achieved its best mean sensitivity of 94.3%, 91.5%, 87.3% and 96.3%, respectively. While in the case of RIF and PANS-TB, it generated 94.2% and 92.2% sensitivity, which were lower than baseline model by 0.7% and 1.9%, respectively. t-SNE visualization shows that DeepAMR_cluster captures lineage-related clusters in the latent space.     On the dependent recognition of some long zinc finger proteins

    The human genome contains about 800 C2H2 zinc finger proteins (ZFPs), and most of them are composed of long arrays of zinc fingers. Standard ZFP recognition model asserts longer finger arrays should recognize longer DNA-binding sites. However, recent experimental efforts to identify in vivo ZFP binding sites contradict this assumption, with many exhibiting short motifs. Here we use ZFY, CTCF, ZIM3, and ZNF343 as examples to address three closely related questions: What are the reasons that impede current motif discovery methods? What are the functions of those seemingly unused fingers and how can we improve the motif discovery algorithms based on long ZFPs\u27 biophysical properties? Using ZFY, we employed a variety of methods and find evidence for \u27dependent recognition\u27 where downstream fingers can recognize some previously undiscovered motifs only in the presence of an intact core site. For CTCF, high-throughput measurements revealed its upstream specificity profile depends on the strength of its core. Moreover, the binding strength of the upstream site modulates CTCF\u27s sensitivity to different epigenetic modifications within the core, providing new insight into how the previously identified intellectual disability-causing and cancer-related mutant R567W disrupts upstream recognition and deregulates the epigenetic control by CTCF. Our results establish that, because of irregular motif structures, variable spacing and dependent recognition between sub-motifs, the specificities of long ZFPs are significantly underestimated, so we developed an algorithm, ModeMap, to infer the motifs and recognition models of ZIM3 and ZNF343, which facilitates high-confidence identification of specific binding sites, including repeats-derived elements. With revised concept, technique, and algorithm, we can discover the overlooked specificities and functions of those \u27extra\u27 fingers, and therefore decipher their broader roles in human biology and diseases

    Theory and application of draw control for elastic webs with nipped pull rollers

    Draw control is defined as changing or maintaining web tension by controlling surface speeds of consecutive driven rollers. Even though draw control is commonly used in web handling applications, there remains some misunderstanding of the relationship between machine draw and the resulting web elongation and tension. Fundamentals of draw control and a new analytical solution will be presented.Draw control is commonly used downstream of a constant tension zone. The strain at any point in the process is dependent on the upstream tension and the added web strain from the machine draw. Recent experimental studies of draw control have revealed a previously unknown influence of nip roller load, similar to pack rollers at winding. Models for nipped pull roller slip conditions, and nip induced draw and tension will be presented

    Understanding Implementation Strategies to Support Classroom-Based Physical activity approaches in Elementary Schools: a Qualitative Study

    BACKGROUND: Classroom-based physical activity approaches can improve children\u27s physical activity levels during school. However, the implementation of these approaches remains a challenge. The purpose of this study was to examine implementation strategies to support the delivery of classroom-based physical activity approaches from the perspectives of elementary school staff. METHODS: We conducted individual interviews with elementary school staff from a mid-sized school district in Texas. Interviews lasted approximately 60 min and were audio recorded and transcribed for analyses. We used directed content analysis and an iterative categorization approach to identify emerging themes related to implementation strategies. RESULTS: We interviewed 15 participants (4 classroom teachers, 4 physical education teachers, 3 assistant principals, and 4 principals) about implementation strategies supporting classroom-based physical activity approaches. Four prominent themes related to implementation strategies emerged: 1) the role of program champions, 2) the use and function of staff training, 3) the importance of strategic planning, and 4) the use of positive reinforcements to support implementation. CONCLUSIONS: Results highlight the need for multiple implementation strategies to support the delivery of classroom-based physical activity approaches. Results also highlight potential mechanisms through which the implementation strategies operate. This information is valuable to future planning efforts for classroom-based physical activity approaches

    A Cure for HIV Infection: "Not in My Lifetime" or "Just Around the Corner"?

    With the advent and stunning success of combination antiretroviral therapy (ART) to prolong and improve quality of life for persons with HIV infection, HIV research has been afforded the opportunity to pivot towards studies aimed at finding "a cure." The mere idea that cure of HIV might be possible has energized researchers and the community towards achieving this goal. Funding agencies, both governmental and private, have targeted HIV cure as a high priority; many in the field have responded to these initiatives and the cure research agenda is robust. In this "salon" two editors of Pathogens and Immunity, Michael Lederman and Daniel Douek ask whether curing HIV is a realistic, scalable objective. We start with an overview perspective and have asked a number of prominent HIV researchers to add to the discussion
