Hieroglyph: Hierarchical Glia Graph Skeletonization and Matching
Automatic 3D reconstruction of glia morphology is a powerful tool necessary
for investigating the role of microglia in neurological disorders in the
central nervous system. Current glia skeleton reconstruction techniques fail to
capture an accurate tracing of the processes over time, useful for the study of
the microglia motility and morphology in the brain during healthy and diseased
states. We propose Hieroglyph, a fully automatic temporal 3D skeleton
reconstruction algorithm for glia imaged via 3D multiphoton microscopy.
Hieroglyph yielded a 21% performance increase compared to state of the art
automatic skeleton reconstruction methods and outperforms the state of the art
in different measures of consistency on datasets of 3D images of microglia. The
results from this method provide a 3D graph and digital reconstruction of glia
useful for a myriad of morphological analyses that could impact studies in
brain immunology and disease.Comment: submitted to IEEE International Conference on Image Processing, 201

Osteoprotegerin reduces osteoclast resorption activity without affecting osteogenesis on nanoparticulate mineralized collagen scaffolds.
The instructive capabilities of extracellular matrix-inspired materials for osteoprogenitor differentiation have sparked interest in understanding modulation of other cell types within the bone regenerative microenvironment. We previously demonstrated that nanoparticulate mineralized collagen glycosaminoglycan (MC-GAG) scaffolds efficiently induced osteoprogenitor differentiation and bone healing. In this work, we combined adenovirus-mediated delivery of osteoprotegerin (AdOPG), an endogenous anti-osteoclastogenic decoy receptor, in primary human mesenchymal stem cells (hMSCs) with MC-GAG to understand the role of osteoclast inactivation in augmentation of bone regeneration. Simultaneous differentiation of osteoprogenitors on MC-GAG and osteoclast progenitors resulted in bidirectional positive regulation. AdOPG expression did not affect osteogenic differentiation alone. In the presence of both cell types, AdOPG-transduced hMSCs on MC-GAG diminished osteoclast-mediated resorption in direct contact; however, osteoclast-mediated augmentation of osteogenic differentiation was unaffected. Thus, the combination of OPG with MC-GAG may represent a method for uncoupling osteogenic and osteoclastogenic differentiation to augment bone regeneration
Finding the Needles in the Haystacks: High-Fidelity Models of the Modern and Archean Solar System for Simulating Exoplanet Observations
We present two state-of-the-art models of the solar system, one corresponding
to the present day and one to the Archean Eon 3.5 billion years ago. Each model
contains spatial and spectral information for the star, the planets, and the
interplanetary dust, extending to 50 AU from the sun and covering the
wavelength range 0.3 to 2.5 micron. In addition, we created a spectral image
cube representative of the astronomical backgrounds that will be seen behind
deep observations of extrasolar planetary systems, including galaxies and Milky
Way stars. These models are intended as inputs to high-fidelity simulations of
direct observations of exoplanetary systems using telescopes equipped with
high-contrast capability. They will help improve the realism of observation and
instrument parameters that are required inputs to statistical observatory yield
calculations, as well as guide development of post-processing algorithms for
telescopes capable of directly imaging Earth-like planets.Comment: Accepted for publication in PAS
Cold-gas outflows in typical low-redshift galaxies are driven by star formation, not AGN
This article has been accepted for publication in Monthly Notices of the Royal Astronomical Society. © 2015 The Authors. Published by Oxford University Press on behalf of the Royal Astronomical Society. Available online at: https://doi.org/10.1093/mnrasl/slv165Energetic feedback from active galactic nuclei (AGN) is an important ingredient for regulating the star-formation history of galaxies in models of galaxy formation, which makes it important to study how AGN feedback actually occurs in practice. In order to catch AGNs in the act of quenching star formation we have used the interstellar NaD absorption lines to look for cold-gas outflows in a sample of 456 nearby galaxies for which we could unambigously ascertain the presence of radio AGN activity, thanks to radio imaging at milli-arcsecond scales. While compact radio emission indicating a radio AGN was found in 103 galaxies (23% of the sample), and 23 objects (5%) exhibited NaD absorption-line kinematics suggestive of cold-gas outflows, not one object showed evidence of a radio AGN and of a cold-gas outflow simultaneously. Radio AGN activity was found predominantly in early-type galaxies, while cold-gas outflows were mainly seen in spiral galaxies with central star-formation or composite star-formation/AGN activity. Optical AGNs also do not seem capable of driving galactic winds in our sample. Our work adds to a picture of the low-redshift Universe where cold-gas outflows in massive galaxies are generally driven by star formation and where radio-AGN activity occurs most often in systems in which the gas reservoir has already been significantly depleted.Peer reviewedFinal Published versio
Starting on the Right Foot: Carryover Effects of Larval Hydroperiod and Terrain Moisture on Post-metamorphic Frog Movement Behavior
Changing patterns of precipitation and drought will dramatically influence the distribution and persistence of lentic habitats. Pond-breeding amphibians can often respond to changes in habitat by plastically shifting behavioral and developmental trait response. However, fitness tradeoffs inherent in life history strategies can carry over to impact development, behavior, and fitness in later life stages. In this experiment, we investigated carryover effects of hydroperiod permanence on the movement behavior of newly-metamorphosed juvenile Northern Red-legged Frogs (Rana aurora). Frogs were raised through metamorphosis in mesocosms under either permanent or ephemeral hydroperiod conditions. After metamorphosis, individuals were removed from the mesocosms, measured, uniquely tagged with elastomer, and moved to holding terrariums. Movement behavior was quantified under two terrain conditions: a physiologically-taxing, dry runway treatment, or a control, moist runway treatment. Individuals were given 30 min to move down the 1 × 20m enclosed structure before distance was measured. We applied a hurdle model to examine two distinct components of movement behavior: (1) the probability of moving away from the start location, and (2) movement distance. We found that hydroperiod condition had an indirect carryover effect on movement via the relationship between individual size and the propensity to move. Individuals from ephemeral mesocosm conditions metamorphosed at a smaller size but showed increased growth rates as compared to individuals from permanent hydroperiod conditions. Individual snout-vent length and runway condition (moist or dry) were significant predictors of both aspects of movement behavior. Larger individuals were more likely to move down the runway and able to move a farther distance than smaller individuals. In addition to the influence of size, dry runway conditions reduced the probability of individuals moving from the start location, but increased the distance traveled relative to the moist runway. The demonstrated cumulative impact of stressors suggests the importance of addressing direct, indirect, and carryover effects of stressors throughout ontogeny
Sustainable Development Goal 3
Sustainable Development Goals (SDGs) aim to develop a better and sustainable future for the world and the goals are part of an action plan to address poverty, hunger, health, gender equity and various pressing world issues. One of these goals looks at health and wellness. Ageing populations have become a crucial issue worldwide and this short monograph explores ageing and how the consequences of an ageing population may affect our health care system through the case study on Hong Kong's population. The book looks at several critical health issues related to ageing. The elderly, particularly those with low socioeconomic status, rely more on the acute-centric care rather than primary care. The book suggests that secondary care service may only be effective to limited extent as a healthcare measure and an optimum health care system should be one that focuses on primary care. The authors put forth a compelling argument for disease prevention and screening schemes and explain how they are more cost-effective and beneficial to the society and the system. This thoughtful book will provide beneficial insights into the relationship of ageing and sustainable development goals in the context of health and wellness for policy makers and healthcare professionals
