    A Case Study of Excreta Disposal Following the 2006 Java Earthquake*

    Providing safe excreta disposal following disasters is important for disease prevention and the safety and dignity of the affected population. This is challenging because every emergency varies due to the nature of the disaster, local conditions and the characteristics of the affected population. This paper investigates the impact of the 2006 Java earthquake on excreta disposal needs and the response to those needs. Relevant documents were retrieved from the ReliefWeb database, complemented by a literature search. The case study highlights gaps in rapidly providing latrines on a large scale. Three months after the disaster, only 57% of the latrines targeted had been provided. One way to address this problem is to better understand the factors affecting excreta disposal needs and response, allowing appropriate solutions to be identified more effectively

    Light-color-induced Changes in Fatty Acid Biosynthesis in Chlorella SP. Strain Ks-ma2 in Early Stationary Growth Phase

    Optimization of light supply remains a critical issue in microalgae biotechnology. The impacts of light color on fatty acid production and biosynthesis in microalgae are poorly understood. The aim of this study was to determine the effect of light color on growth and fatty acid content in Chlorella strain KS-MA2. Cells were cultured on F/2 medium and incubated under blue, green, red or white light. The cells' growth, fatty acid composition and the expression levels of the ketoacyl synthase 1 (KAS-1), omega-6 desaturase (ω-6 FAD) and omega-3 desaturase (ω-3 FAD) genes were measured at the early stationary growth phase. Results of this study indicated that light color affected cell density and fatty acid profile produced by Chlorella sp. strain KS-MA2. Cells cultured under blue, red and white light had higher cell density than those cultured under green light. Palmitic acid (38.62 ± 3.29% of biomass dry weight) and linolenic acid (7.96 ± 0.88% of biomass dry weight) were highly accumulated under white light. Stearic acid was dominant under blue light (11.11 ± 0.14% of biomass dry weight), whereas oleic acid was dominant under red light (30.50 ± 0.14% of biomass dry weight). Linoleic acid was highly produced under green and blue light (28.63 ± 1.36% and 26.00 ± 0.81 % of biomass dry weight, respectively). KAS-1 and ω-6 FAD were highly expressed under blue light, whereas ω-3 FAD was highly expressed under green light. The production of particular fatty acids of interest from Chlorella could be achieved by shifting color of light used during the incubation of the cell cultures.     Learning task-optimal image registration with applications in localizing structure and function in the cerebral cortex

Registration is rarely the end-goal, but instead, the results of image registration are used in other tasks, such as voxel-based morphometry, functional group analysis, image segmentation and tracking. In this thesis, we argue that the quality of image registration should be evaluated in the context of the application. Consequently, we develop a framework for learning registration cost functions optimized for specific tasks. We demonstrate that by taking into account the application, we not only achieve better registration, but also potentially resolve certain ambiguities and ill-posed nature of image registration. We first develop a generative model for joint registration and segmentation of images. By jointly modeling registration and the application of image segmentation, we demonstrate improvements in parcellation of the cerebral cortex into different structural units. In this thesis, we work with spherical representations of the human cerebral cortex. Consequently, we develop a fast algorithm for registering spherical images. Application to the cortex shows that our algorithm achieves state-of-the-art accuracy, while being an order of magnitude faster than competing diffeomorphic, landmark-free algorithms. Finally, we consider the problem of automatically determining the "free" parameters of registration cost functions.(cont.) Registration is usually formulated as an optimization problem with multiple tunable parameters that are manually set. By introducing a second layer of optimization over and above the usual registration, this thesis provides the first effective approach to optimizing thousands of registration parameters to improve alignment of a new image as measured by an application-specific performance measure. Much previous work has been devoted to developing generic registration algorithms, which are then specialized to particular imaging modalities (e.g., MR), particular imaging targets (e.g., cardiac) and particular post- registration analyses (e.g., segmentation). Our framework provides a principled method for adapting generic algorithms to specific applications. For example, we estimate the optimal weights or cortical folding template of the generic weighted Sum of Squared Differences dissimilarity measure for localizing underlying cytoarchitecture and functional regions of the cerebral cortex.     Susceptibility to tuberculosis is associated with variants in the ASAP1 gene encoding a regulator of dendritic cell migration

    Human genetic factors predispose to tuberculosis (TB). We studied 7.6 million genetic variants in 5,530 people with pulmonary TB and in 5,607 healthy controls. In the combined analysis of these subjects and the follow-up cohort (15,087 TB patients and controls altogether), we found an association between TB and variants located in introns of the ASAP1 gene on chromosome 8q24 (P = 2.6 × 10−11 for rs4733781; P = 1.0 × 10−10 for rs10956514). Dendritic cells (DCs) showed high ASAP1 expression that was reduced after Mycobacterium tuberculosis infection, and rs10956514 was associated with the level of reduction of ASAP1 expression. The ASAP1 protein is involved in actin and membrane remodeling and has been associated with podosomes. The ASAP1-depleted DCs showed impaired matrix degradation and migration. Therefore, genetically determined excessive reduction of ASAP1 expression in M. tuberculosis–infected DCs may lead to their impaired migration, suggesting a potential mechanism of predisposition to TB

    Emergency toilets for the people affected by the Mount Sinabung eruptions

    Mount Sinabung erupted in September 2013, causing the mass evacuation of more than 30,000 people. Evacuees were placed in public buildings such as churches, mosques, schools and universities, as well as tent camps. Pour-flush toilets were provided by the government. However, the level of provision was generally inadequate. The continuity of water was a major concern, with the people relying on water trucks for the supply of water. Toilets were not properly maintained and rubbish was found around toilets, which could clog drainage channels. Insufficient cleaning tools were provided. Coverage far exceeded generally accepted targets of twenty people per toilet. To address these issues, water supply should be closely monitored, additional toilets installed, adequate cleaning tools should be provided and people should be better organised to maintain toilets

In the United Kingdom, over 1 million women suffer from CPP, which has been highlighted as a key area of unmet need. Current medical treatments for CPP are often associated with unacceptable side effects. A specific style of acupuncture, the meridian balance method electro-acupuncture (BMEA) and traditional Chinese medicine health consultation (TCM HC (BMEA + TCM HC = BMEA treatment)), may be effective for CPP in women. Aim: Three focus group discussions and semi-structured telephone interviews were embedded in a randomised controlled feasibility trial to gain in-depth description of the perceived benefits of the participants’ respective interventions. Methods: Women with CPP were randomised into the BMEA treatment, TCM HC or National Health Service standard care (NHS SC). Focus group discussions were recorded, transcribed and analysed thematically. Semi-structured telephone interviews were conducted post focus group discussions. Findings: A total of 30 women were randomised into BMEA treatment, TCM HC or NHS SC. A total of 11 participants attended the three focus group discussions. Thematic analysis of focus group discussions showed: a perceived pain reduction, enhanced sleep, energy level and sense of well-being in the BMEA treatment and TCM HC groups; a dislike for the adverse effects of medications, frustration at the lack of effective treatment, heavy reliance on medications and services that are helpful, in the NHS SC group. Semi-structured telephone interviews showed that the methodology was acceptable to the participants. Conclusion: The embedded focus group discussions captured the rich and complex narratives of the participants and provided insights into the perceived benefits of the BMEA treatment, TCM HC and NHS SC interventions.The research was partly supported by the Morag Robinson Legacy, the Alexander Dykes Fund and Barbour Watson Trust.13pubpub

    Evidence for metallic 1T phase, 3d1 electronic configuration and charge density wave order in molecular-beam epitaxy grown monolayer VTe2

    We present a combined experimental and theoretical study of monolayer VTe2 grown on highly oriented pyrolytic graphite by molecular-beam epitaxy. Using various in-situ microscopic and spectroscopic techniques, including scanning tunneling microscopy/spectroscopy, synchrotron X-ray and angle-resolved photoemission, and X-ray absorption, together with theoretical analysis by density functional theory calculations, we demonstrate direct evidence of the metallic 1T phase and 3d1 electronic configuration in monolayer VTe2 that also features a (4 x 4) charge density wave order at low temperatures. In contrast to previous theoretical predictions, our element-specific characterization by X-ray magnetic circular dichroism rules out a ferromagnetic order intrinsic to the monolayer. Our findings provide essential knowledge necessary for understanding this interesting yet less explored metallic monolayer in the emerging family of van der Waals magnets.Comment: 21 pages, 5 figure

    Sp110 transcription is induced and required by Anaplasma phagocytophilum for infection of human promyelocytic cells

    <p>Abstract</p> <p>Background</p> <p>The tick-borne intracellular pathogen, <it>Anaplasma phagocytophilum </it>(Rickettsiales: Anaplasmataceae) causes human granulocytic anaplasmosis after infection of polymorphonuclear leucocytes. The human Sp110 gene is a member of the nuclear body (NB) components that functions as a nuclear hormone receptor transcriptional coactivator and plays an important role in immunoprotective mechanisms against pathogens in humans. In this research, we hypothesized that Sp110 may be involved in the infection of human promyelocytic HL-60 cells with <it>A. phagocytophilum</it>.</p> <p>Methods</p> <p>The human Sp110 and <it>A. phagocytophilum msp4 </it>mRNA levels were evaluated by real-time RT-PCR in infected human HL-60 cells sampled at 0, 12, 24, 48, 72 and 96 hours post-infection. The effect of Sp110 expression on <it>A. phagocytophilum </it>infection was determined by RNA interference (RNAi). The expression of Sp110 was silenced in HL-60 cells by RNAi using pre-designed siRNAs using the Nucleofector 96-well shuttle system (Amaxa Biosystems, Gaithersburg, MD, USA). The <it>A. phagocytophilum </it>infection levels were evaluated in HL-60 cells after RNAi by real-time PCR of <it>msp4 </it>and normalizing against human <it>Alu </it>sequences.</p> <p>Results</p> <p>While Sp110 mRNA levels increased concurrently with <it>A. phagocytophilum </it>infections in HL-60 cells, the silencing of Sp110 expression by RNA interference resulted in decreased infection levels.</p> <p>Conclusion</p> <p>These results demonstrated that Sp110 expression is required for <it>A. phagocytophilum </it>infection and multiplication in HL-60 cells, and suggest a previously undescribed mechanism by which <it>A. phagocytophilum </it>modulates Sp110 mRNA levels to facilitate establishment of infection of human HL-60 cells.</p

    Anthropogenic edge effects in habitat selection by sun bears in a protected area

    Wildlife populations in southeast Asia are increasingly experiencing a broad array of anthropogenic threats, and mammalian carnivores are particularly vulnerable. Populations of the Malayan sun bear Helarctos malayanus are estimated to have declined by 30% over the last 30 years from forest conversion to industrial plantations and mortality associated with human–bear conflicts and illegal wildlife trade. However, the effects of industrial plantations on habitat selection and activity patterns of mammals that live at the protected area-plantation interface, including sun bears, are not well known. We investigated habitat selection and activity patterns of sun bears in Tabin Wildlife Reserve in Sabah, Malaysia. We deployed 83 remote camera sites to record sun bear detections during two sampling periods (2012–2013 and 2017). We used generalized linear models to examine relationships between sun bear presence and site covariates representing physical, environmental and anthropogenic elements of the landscape. Relative probability of sun bear presence was positively associated with distance to roads and elevation. Because most roads were on the reserve boundary and often associated with oil palm plantations, proximity to roads likely served as a surrogate measure of human accessibility and activity in peripheral areas of the reserve. Supporting that interpretation, sun bears close to the reserve boundary were primarily active at night, whereas daytime activity was more common for bears in the interior. Our findings indicate that sun bears alter behaviour and habitat selection likely in response to anthropogenic activities at the edges of Tabin Wildlife Reserve (112 200 ha). Because the ratio of edge to interior increases steeply with declining habitat area, smaller protected areas bordered by plantations are predicted to have greater impacts on sun bear behaviour and, potentially, population persistence. Effective conservation actions may benefit from management to improve the security of edge habitats for sun bears and other vulnerable species
