    Antibacterial property of neem nanoemulsion against Vibrio anguillarium infection in Asian sea bass (Lates calcarifer)

    1222-1226Fish vibriosis is among the most common diseases that is caused by a bacteria belonging to the genus Vibrio (Vibrio anguillarium). It causes considerable economic loss in the commercial cultivation of Asian sea bass (Lates calcarifer). The resistance of microorganisms to antibiotics has resulted in a growing need for developing a new antibacterial therapy that is effective in aquaculture. The aim of this study is to develop neem nanoemulsion with antibacterial activity against V. anguillarium to identify a possible alternative to the commonly used antibiotics in aquaculture. Neem nanoemulsion was prepared and the effectiveness was studied both in vitro and in vivo (agar well diffusion assay and artificial infection). Injection and immersion challenge of neem nanoemulsion formulated the fish less susceptible to V. anguillarium infection. The results confirmed the potential use of neem nanoemulsion as a source of antibacterial compounds or as a health-promoting medicine for fish culture

The nucleus is visible in the zygotes on the 7th day after the introduction of spores into fresh wort. A nuclear membrane delimits the nucleus from the cytoplasm and encloses formed structures. The structure of the nucleus is identical in a zygote and its bud. 3. Iodine-formaldehyde-acetic gives a life-like preservation of cell structures. After removal of the cytoplasmic basophilia by acid hydrolysis, staining with hæmatoxylin gives pictures of nuclear details comparable to that of the living nucleus.     Serum adiponectin is positively associated with lung function in young adults, independent of obesity: The CARDIA study

    <p>Abstract</p> <p>Rationale</p> <p>Adipose tissue produces adiponectin, an anti-inflammatory protein. Adiponectin deficiency in mice is associated with abnormal post-natal alveolar development.</p> <p>Objective</p> <p>We hypothesized that lower serum adiponectin concentrations are associated with lower lung function in humans, independent of obesity. We explored mediation of this association by insulin resistance and systemic inflammation.</p> <p>Methods and Measurements</p> <p>Spirometry testing was conducted at years 10 and 20 follow-up evaluation visits in 2,056 eligible young adult participants in the Coronary Artery Risk Development in Young Adults (CARDIA) study. Body mass index, serum adiponectin, serum C-reactive protein (a marker of systemic inflammation), and insulin resistance were assessed at year 15.</p> <p>Main Results</p> <p>After controlling for body mass index, years 10 and 20 forced vital capacity (FVC) were 81 ml and 82 ml lower respectively (p = 0.004 and 0.01 respectively) in the lowest <it>vs</it>. highest adiponectin quartiles. Similarly, years 10 and 20 forced expiratory volume in one second (FEV<sub>1</sub>) were 50 ml and 38 ml lower (p = 0.01 and 0.09, respectively) in the lowest <it>vs</it>. highest adiponectin quartiles. These associations were no longer significant after adjustment for insulin resistance and C-reactive protein. Serum adiponectin was not associated with FEV<sub>1</sub>/FVC or peak FEV<sub>1</sub>.</p> <p>Conclusions</p> <p>Independent of obesity, lower serum adiponectin concentrations are associated with lower lung function. The attenuation of this association after adjustment for insulin resistance and systemic inflammation suggests that these covariates are on a causal pathway linking adiponectin and lung function.</p

    Measuring phased-array antenna beampatterns with high dynamic range for the Murchison Widefield Array using 137MHz ORBCOMM satellites

    Detection of the fluctuations in a 21 cm line emission from neutral hydrogen during the Epoch of Reionization in thousand hour integrations poses stringent requirements on calibration and image quality, both of which necessitate accurate primary beam models. The Murchison Widefield Array (MWA) uses phased-array antenna elements which maximize collecting area at the cost of complexity. To quantify their performance, we have developed a novel beam measurement system using the 137 MHz ORBCOMM satellite constellation and a reference dipole antenna. Using power ratio measurements, we measure the in situ beampattern of the MWA antenna tile relative to that of the reference antenna, canceling the variation of satellite flux or polarization with time. We employ angular averaging to mitigate multipath effects (ground scattering) and assess environmental systematics with a null experiment in which the MWA tile is replaced with a second-reference dipole. We achieve beam measurements over 30 dB dynamic range in beam sensitivity over a large field of view (65% of the visible sky), far wider and deeper than drift scans through astronomical sources allow. We verify an analytic model of the MWA tile at this frequency within a few percent statistical scatter within the full width at half maximum. Toward the edges of the main lobe and in the sidelobes, we measure tens of percent systematic deviations. We compare these errors with those expected from known beamforming errors

    Systemic Inflammation in Young Adults Is Associated with Abnormal Lung Function in Middle Age

    BACKGROUND:Systemic inflammation is associated with reduced lung function in both healthy individuals and those with chronic obstructive pulmonary disease (COPD). Whether systemic inflammation in healthy young adults is associated with future impairment in lung health is uncertain. METHODOLOGY/PRINCIPAL FINDINGS:We evaluated the association between plasma fibrinogen and C-reactive protein (CRP) in young adults and lung function in the Coronary Artery Risk Development in Young Adults cohort study. Higher year 7 fibrinogen was associated with greater loss of forced vital capacity (FVC) between years 5 and 20 (439 mL in quartile 4 vs. 398 mL in quartile 1, P<0.001) and forced expiratory volume in 1 second (FEV(1)) (487 mL in quartile 4 vs. 446 mL in quartile 1, P<0.001) independent of cigarette smoking, body habitus, baseline lung function and demographic factors. Higher year 7 CRP was also associated with both greater loss of FVC (455 mL in quartile 4 vs. 390 mL in quartile 1, P<0.001) and FEV(1) (491 mL in quartile 4 vs. 442 mL in quartile 1, P = 0.001). Higher year 7 fibrinogen and CRP were associated with abnormal FVC at year 20 (odds ratio (OR) per standard deviation 1.51 (95% confidence interval (CI): 1.30-1.75) for fibrinogen and 1.35 (95% CI: 1.14-1.59) for CRP). Higher year 5 fibrinogen was additionally associated with abnormal FEV(1). A positive interaction was observed between pack-years cigarette smoking and year 7 CRP for the COPD endpoint, and among participants with greater than 10 pack-years of cigarette exposure, year 7 CRP was associated with greater odds of COPD at year 20 (OR per standard deviation 1.53 (95% CI: 1.08-2.16). CONCLUSION/SIGNIFICANCE:Systemic inflammation in young adults is associated with abnormal lung function in middle age. In particular, elevated CRP may identify vulnerability to COPD among individuals who smoke. TRIAL REGISTRATION:ClinicalTrials.gov NCT00005130

    Beam-forming errors in Murchison Widefield array phased array antennas and their effects on epoch of reionization science

    Accurate antenna beam models are critical for radio observations aiming to isolate the redshifted 21 cm spectral line emission from the Dark Ages and the Epoch of Reionization (EOR) and unlock the scientific potential of 21 cm cosmology. Past work has focused on characterizing mean antenna beam models using either satellite signals or astronomical sources as calibrators, but antenna-to-antenna variation due to imperfect instrumentation has remained unexplored. We characterize this variation for the Murchison Widefield Array (MWA) through laboratory measurements and simulations, finding typical deviations of the order of ±10%-20% near the edges of the main lobe and in the sidelobes. We consider the ramifications of these results for image- and power spectrum-based science. In particular, we simulate visibilities measured by a 100 m baseline and find that using an otherwise perfect foreground model, unmodeled beam-forming errors severely limit foreground subtraction accuracy within the region of Fourier space contaminated by foreground emission (the "wedge"). This region likely contains much of the cosmological signal, and accessing it will require measurement of per-antenna beam patterns. However, unmodeled beam-forming errors do not contaminate the Fourier space region expected to be free of foreground contamination (the "EOR window"), showing that foreground avoidance remains a viable strategy

    Sleep disordered breathing and fibroblast growth factor 23 in the Hispanic Community Health Study/Study of Latinos

    Preclinical data suggest that hypoxia stimulates fibroblast growth factor 23 (FGF23) transcription and cleavage in osteocytes, resulting in elevated circulating c-terminal (cFGF23) levels but normal intact FGF23 (iFGF23) levels. We conducted a case-control study within the Hispanic Community Health Study/Study of Latinos to investigate whether sleep disordered breathing, as a model of hypoxemia, is independently associated with elevated cFGF23 levels in the general population and with elevated cFGF23 and iFGF23 levels in patients with chronic kidney disease (CKD), in whom FGF23 cleavage may be impaired. Cases (n = 602) had severe sleep disordered breathing defined as an apnea/hypopnea index (AHI) of ≥30. Controls without severe sleep disordered breathing (n = 602) were matched for sex and CKD stage. The median AHI in the cases was 45.8 (IQR 35.5–62.5) compared to 2.6 (IQR 0.6–8.2) in the controls. Cases had higher cFGF23 levels than controls (66.2 RU/mL, IQR 52.8–98.4 vs. 61.2 RU/mL, IQR 49.5–80.1, p value <0.001). There were no differences in iFGF23 levels between cases and controls. In adjusted linear regression and multinomial regression analyses, body mass index attenuated the relationship between severe sleep disordered breathing and cFGF23 levels. No significant relationships were seen in analyses of severe sleep disordered breathing and iFGF23 levels or in analyses of iFGF23 and cFGF23 stratified by CKD status. Additional studies using other models of intermittent and chronic hypoxia are needed to confirm whether hypoxia stimulates FGF23 transcription in humans and to determine the impact on iFGF23 levels in CKD

    The murchison widefield array 21 cm power spectrum analysis methodology

    We present the 21 cm power spectrum analysis approach of the Murchison Widefield Array Epoch of Reionization project. In this paper, we compare the outputs of multiple pipelines for the purpose of validating statistical limits cosmological hydrogen at redshifts between 6 and 12. Multiple independent data calibration and reduction pipelines are used to make power spectrum limits on a fiducial night of data. Comparing the outputs of imaging and power spectrum stages highlights differences in calibration, foreground subtraction, and power spectrum calculation. The power spectra found using these different methods span a space defined by the various tradeoffs between speed, accuracy, and systematic control. Lessons learned from comparing the pipelines range from the algorithmic to the prosaically mundane; all demonstrate the many pitfalls of neglecting reproducibility. We briefly discuss the way these different methods attempt to handle the question of evaluating a significant detection in the presence of foregrounds

    KNOWN PULSARS IDENTIFIED in the GMRT 150 MHz ALL-SKY SURVEY

    We have used the 150 MHz radio continuum survey (TGSS ADR) from the Giant Metrewave Radio Telescope (GMRT) to search for phase-averaged emission toward all well-localized radio pulsars north of -53° decl. We detect emission toward 200 pulsars with high confidence (=5s) and another 88 pulsars at fainter levels. We show that most of our identifications are likely from pulsars, except for a small number where the measured flux density is confused by an associated supernova or pulsar-wind nebula, or a globular cluster. We investigate the radio properties of the 150 MHz sample and find an unusually high number of gamma-ray binary millisecond pulsars with very steep spectral indices. We also note a discrepancy in the measured flux densities between GMRT and LOFAR pulsar samples, suggesting that the flux density scale for the LOFAR pulsar sample may be in error by approximately a factor of two. We carry out a separate search of 30 well-localized gamma-ray, radio-quiet pulsars in an effort to detect a widening of the radio beam into the line of sight at lower frequencies. No steep-spectrum emission was detected either toward individual pulsars or in a weighted stack of all 30 images

    ReishiMax, mushroom based dietary supplement, inhibits adipocyte differentiation, stimulates glucose uptake and activates AMPK

    <p>Abstract</p> <p>Background</p> <p>Obesity is a health hazard which is closely associated with various complications including insulin resistance, hypertension, dyslipidemia, atherosclerosis, type 2 diabetes and cancer. In spite of numerous preclinical and clinical interventions, the prevalence of obesity and its related disorders are on the rise demanding an urgent need for exploring novel therapeutic agents that can regulate adipogenesis. In the present study, we evaluated whether a dietary supplement ReishiMax (RM), containing triterpenes and polysaccharides extracted from medicinal mushroom <it>Ganoderma lucidum</it>, affects adipocyte differentiation and glucose uptake in 3T3-L1 cells.</p> <p>Methods</p> <p>3T3-L1 pre-adipocytes were differentiated into adipocytes and treated with RM (0-300 μg/ml). Adipocyte differentiation/lipid uptake was evaluated by oil red O staining and triglyceride and glycerol concentrations were determined. Gene expression was evaluated by semi-quantitative RT-PCR and Western blot analysis. Glucose uptake was determined with [<sup>3</sup>H]-glucose.</p> <p>Results</p> <p>RM inhibited adipocyte differentiation through the suppresion of expression of adipogenic transcription factors peroxisome proliferator-activated receptor-γ (PPAR-γ), sterol regulatory element binding element protein-1c (SREBP-1c) and CCAAT/enhancer binding protein-α (C/EBP-α). RM also suppressed expression of enzymes and proteins responsible for lipid synthesis, transport and storage: fatty acid synthase (FAS), acyl-CoA synthetase-1 (ACS1), fatty acid binding protein-4 (FABP4), fatty acid transport protein-1 (FATP1) and perilipin. RM induced AMP-activated protein kinase (AMPK) and increased glucose uptake by adipocytes.</p> <p>Conclusion</p> <p>Our study suggests that RM can control adipocyte differentiation and glucose uptake. The health benefits of ReishiMax warrant further clinical studies.</p
