    MBMS—IP Multicast/Broadcast in 3G Networks

    In this article, the Multimedia Broadcast and Multicast Service (MBMS) as standardized in 3GPP is presented. With MBMS, multicast and broadcast capabilities are introduced into cellular networks. After an introduction into MBMS technology, MBMS radio bearer realizations are presented. Different MBMS bearer services like broadcast mode, enhanced broadcast mode and multicast mode are discussed. Streaming and download services over MBMS are presented and supported media codecs are listed. Service layer components as defined in Open Mobile Alliance (OMA) are introduced. For a Mobile TV use case capacity improvements achieved by MBMS are shown. Finally, evolution of MBMS as part of 3GPP standardization is presented

    Persistent parental RNAi in the Beetle Tribolium castaneum involves maternal transmission of long double‐stranded RNA

    Parental RNA interference (pRNAi) is a powerful and widely used method for gene‐specific knockdown. Yet in insects its efficacy varies between species, and how the systemic response is transmitted from mother to offspring remains elusive. Using the beetle Tribolium castaneum, an RT‐qPCR strategy to distinguish the presence of double‐stranded RNA (dsRNA) from endogenous mRNA is reported. It is found that injected dsRNA is directly transmitted into the egg and persists throughout embryogenesis. Despite this depletion of dsRNA from the mother, it is shown that strong pRNAi can persist for months before waning at strain‐specific rates. In seeking the receptor proteins for cellular uptake of long dsRNA into the egg, a phylogenomics profiling approach of candidate proteins is also presented. A visualization strategy based on taxonomically hierarchical assessment of orthology clustering data to rapidly assess gene age and copy number changes, refined by sequence‐based evidence, is demonstrated. Repeated losses of SID‐1‐like channel proteins in the arthropods, including wholesale loss in the Heteroptera (true bugs), which are nonetheless highly sensitive to pRNAi, are thereby documented. Overall, practical considerations for insect pRNAi against a backdrop of outstanding questions on the molecular mechanism of dsRNA transmission for long‐term, systemic knockdown are elucidated

    Measuring telomere length and telomere dynamics in evolutionary biology and ecology

    Telomeres play a fundamental role in the protection of chromosomal DNA and in the regulation of cellular senescence. Recent work in human epidemiology and evolutionary ecology suggests adult telomere length (TL) may reflect past physiological stress and predict subsequent morbidity and mortality, independent of chronological age. Several different methods have been developed to measure TL, each offering its own technical challenges. The aim of this review is to provide an overview of the advantages and drawbacks of each method for researchers, with a particular focus on issues that are likely to face ecologists and evolutionary biologists collecting samples in the field or in organisms that may never have been studied in this context before. We discuss the key issues to consider and wherever possible try to provide current consensus view regarding best practice with regard to sample collection and storage, DNA extraction and storage, and the five main methods currently available to measure TL. Decisions regarding which tissues to sample, how to store them, how to extract DNA, and which TL measurement method to use cannot be prescribed, and are dependent on the biological question addressed and the constraints imposed by the study system. What is essential for future studies of telomere dynamics in evolution and ecology is that researchers publish full details of their methods and the quality control thresholds they employ

    Technische Optimierungsmöglichkeiten von Holzerntemaschinen zur Reduktion von SpannungseintrÀgen und ihre Auswirkungen auf die Bodenstruktur von Waldböden

    Anhand eines Vortrages wurden Möglichkeiten zur Reduktion von SpannungseintrĂ€gen unter Holzerntemaschinen prĂ€sentiert. Die Auswirkungen auf die Bodenstruktur als Funktion der Auflast der Holzerntemaschinen wurden ausfĂŒhrlich untersucht und Möglichkeiten zur Vermeidung dieser erforscht. Die Untersuchungen fanden auf befahrungsempfindlichen Lössböden an mehreren Standorten in Niedersachsen statt. An jedem Standort wurde eine unbelastete Variante mit verschiedenen Belastungsvarianten verglichen. Dazu wurden verschiedene moderne Holzerntemaschinen (Rad- und Kettenmaschinen) bzw. Maschinenmodifikationen getestet. Sowohl die Messungen der SpannungseintrĂ€ge, mit dem Stress State Transducer System [SST], als auch die bodenphysikalischen Untersuchungen (z.B. Vorbelastung, gesĂ€ttigte WasserleitfĂ€higkeit, LuftkapazitĂ€t, -leitfĂ€higkeit) erfolgten in 20?cm, 40?cm und 60 cm Bodentiefe. Die Ergebnisse zeigen, dass moderne Forstmaschinen ohne weitere Modifikationen immense SchĂ€den an der Bodenstruktur hervorrufen, obwohl sie sich bereits durch bodenschonende Bereifung (geringer Reifeninnendruck, grĂ¶ĂŸere Reifen, grĂ¶ĂŸere KontaktflĂ€che) und Erhöhung der Achsanzahl auszeichnen. Diese SchĂ€den wurden unter den Punkten „Bodenökologie“ sowie „technische Befahrbarkeit“ detaillierter betrachtet und bewertet. Mit Hilfe von Kettenlaufwerken oder BogiebĂ€ndern können die SchĂ€den an der Bodenökologie nur unwesentlich vermindert werden, allerdings beeinflussen diese Modifikationen die Auswirkungen auf die technische Befahrbarkeit einer potenziellen RĂŒckegasse deutlich positiv und fĂŒhren somit indirekt zur Sicherung der Bodenfunktionen im Wald

    Fourier Analysis of Gapped Time Series: Improved Estimates of Solar and Stellar Oscillation Parameters

    Quantitative helio- and asteroseismology require very precise measurements of the frequencies, amplitudes, and lifetimes of the global modes of stellar oscillation. It is common knowledge that the precision of these measurements depends on the total length (T), quality, and completeness of the observations. Except in a few simple cases, the effect of gaps in the data on measurement precision is poorly understood, in particular in Fourier space where the convolution of the observable with the observation window introduces correlations between different frequencies. Here we describe and implement a rather general method to retrieve maximum likelihood estimates of the oscillation parameters, taking into account the proper statistics of the observations. Our fitting method applies in complex Fourier space and exploits the phase information. We consider both solar-like stochastic oscillations and long-lived harmonic oscillations, plus random noise. Using numerical simulations, we demonstrate the existence of cases for which our improved fitting method is less biased and has a greater precision than when the frequency correlations are ignored. This is especially true of low signal-to-noise solar-like oscillations. For example, we discuss a case where the precision on the mode frequency estimate is increased by a factor of five, for a duty cycle of 15%. In the case of long-lived sinusoidal oscillations, a proper treatment of the frequency correlations does not provide any significant improvement; nevertheless we confirm that the mode frequency can be measured from gapped data at a much better precision than the 1/T Rayleigh resolution.Comment: Accepted for publication in Solar Physics Topical Issue "Helioseismology, Asteroseismology, and MHD Connections

    Inheritance of Telomere Length in a Bird

    Telomere dynamics are intensively studied in human ageing research and epidemiology, with many correlations reported between telomere length and age-related diseases, cancer and death. While telomere length is influenced by environmental factors there is also good evidence for a strong heritable component. In human, the mode of telomere length inheritance appears to be paternal and telomere length differs between sexes, with females having longer telomeres than males. Genetic factors, e.g. sex chromosomal inactivation, and non-genetic factors, e.g. antioxidant properties of oestrogen, have been suggested as possible explanations for these sex-specific telomere inheritance and telomere length differences. To test the influence of sex chromosomes on telomere length, we investigated inheritance and sex-specificity of telomere length in a bird species, the kakapo (Strigops habroptilus), in which females are the heterogametic sex (ZW) and males are the homogametic (ZZ) sex. We found that, contrary to findings in humans, telomere length was maternally inherited and also longer in males. These results argue against an effect of sex hormones on telomere length and suggest that factors associated with heterogamy may play a role in telomere inheritance and sex-specific differences in telomere length

    Precipitation with polyethylene glycol followed by washing and pelleting by ultracentrifugation enriches extracellular vesicles from tissue culture supernatants in small and large scales

    Extracellular vesicles (EVs) provide a complex means of intercellular signalling between cells at local and distant sites, both within and between different organs. According to their cell-type specific signatures, EVs can function as a novel class of biomarkers for a variety of diseases, and can be used as drug-delivery vehicles. Furthermore, EVs from certain cell types exert beneficial effects in regenerative medicine and for immune modulation. Several techniques are available to harvest EVs from various body fluids or cell culture supernatants. Classically, differential centrifugation, density gradient centrifugation, size-exclusion chromatography and immunocapturing-based methods are used to harvest EVs from EV-containing liquids. Owing to limitations in the scalability of any of these methods, we designed and optimised a polyethylene glycol (PEG)based precipitation method to enrich EVs from cell culture supernatants. We demonstrate the reproducibility and scalability of this method and compared its efficacy with more classical EV-harvesting methods. We show that washing of the PEG pellet and the re-precipitation by ultracentrifugation remove a huge proportion of PEG co-precipitated molecules such as bovine serum albumine (BSA). However, supported by the results of the size exclusion chromatography, which revealed a higher purity in terms of particles per milligram protein of the obtained EV samples, PEG-prepared EV samples most likely still contain a certain percentage of other non-EV associated molecules. Since PEG-enriched EVs revealed the same therapeutic activity in an ischemic stroke model than corresponding cells, it is unlikely that such co-purified molecules negatively affect the functional properties of obtained EV samples. In summary, maybe not being the purification method of choice if molecular profiling of pure EV samples is intended, the optimised PEG protocol is a scalable and reproducible method, which can easily be adopted by laboratories equipped with an ultracentrifuge to enrich for functional active EVs
