    Calculation of multiple-trait sire reliability for traits included in a dairy cattle fertility index

    The advent of genetic evaluations for fertility traits in the UK offers valuable information to farmers that can be used to control fertility problems and safeguard against involuntary culling. In addition to estimated genetic merit, proof reliabilities are required to make correct use of this genetic information. Exact reliabilities, based on the inverse of the coefficient matrix, cannot be estimated for large data sets because of computational restrictions. A method to calculate approximate reliabilities was implemented based on a six-trait sire model. Traits considered were interval between first and second calving, interval between first calving and first service, non-return rate 56 days post first service, number of inseminations per conception, daily milk yield at test nearest day 110 and body condition score. Sire reliabilities were calculated in four steps. Firstly, the number of effective daughters was calculated for each bull, separately for each trait, based on total number of daughters and daughter distribution across herd-year-seasons. Secondly, multiple-trait reliabilities were calculated, based on bull daughter contribution, applying selection index theory on independent daughter groups. Thirdly, (great-) grand-daughter contribution was added to the reliability of each bull, using daughter-based reliability of sons and maternal grandsons. An adjustment was made to account for the probability of bull and son or grandson having daughters in the same herd-year-season. Without the adjustment, reliabilities were inflated by proportionately 0·15 to 0·25. Finally, parent (sire and maternal grandsire) contribution was added to the reliability of each bull. The procedure was first tested on a data subset of 28 061 cow records from 285 bulls. Approximate reliabilities were compared with exact estimates based on the inverse of the coefficient matrix. Mean absolute differences ranged from 0·014 to 0·020 for the six traits and correlation between exact and approximate estimates neared unity. In a full-scale application, sire reliability for the fertility traits increased by proportionately 0·47 to 0·79 over single-trait estimates and the number of bulls with a reliability of 0·60 or more increased by 42 to 115%

    Estimation of prediction error variances via Monte Carlo sampling methods using different formulations of the prediction error variance

    Calculation of the exact prediction error variance covariance matrix is often computationally too demanding, which limits its application in REML algorithms, the calculation of accuracies of estimated breeding values and the control of variance of response to selection. Alternatively Monte Carlo sampling can be used to calculate approximations of the prediction error variance, which converge to the true values if enough samples are used. However, in practical situations the number of samples, which are computationally feasible, is limited. The objective of this study was to compare the convergence rate of different formulations of the prediction error variance calculated using Monte Carlo sampling. Four of these formulations were published, four were corresponding alternative versions, and two were derived as part of this study. The different formulations had different convergence rates and these were shown to depend on the number of samples and on the level of prediction error variance. Four formulations were competitive and these made use of information on either the variance of the estimated breeding value and on the variance of the true breeding value minus the estimated breeding value or on the covariance between the true and estimated breeding values

    Chandra Monitoring of the Candidate Anomalous X-ray Pulsar AX J1845.0-0258

    The population of clearly identified anomalous X-ray pulsars has recently grown to seven, however, one candidate anomalous X-ray pulsar (AXP) still eludes re-confirmation. Here, we present a set of seven Chandra ACIS-S observations of the transient pulsar AX J1845.0-0258, obtained during 2003. Our observations reveal a faint X-ray point source within the ASCA error circle of AX J1845.0-0258's discovery, which we designate CXOU J184454.6-025653 and tentatively identify as the quiescent AXP. Its spectrum is well described by an absorbed single-component blackbody (kT~2.0 keV) or power law (Gamma~1.0) that is steady in flux on timescales of at least months, but fainter than AX J1845.0-0258 was during its 1993 period of X-ray enhancement by at least a factor of 13. Compared to the outburst spectrum of AX J1845.0-0258, CXOU J184454.6-025653 is considerably harder: if truly the counterpart, then its spectral behaviour is contrary to that seen in the established transient AXP XTE J1810-197, which softened from kT~0.67 keV to ~0.18 keV in quiescence. This unexpected result prompts us to examine the possibility that we have observed an unrelated source, and we discuss the implications for AXPs, and magnetars in general.Comment: 4 pages, 3 figures.     Compassion-focused self-help for psychological distress associated with skin conditions: a randomized feasibility trial

    Objective: This study tested the feasibility of a self-help intervention based on Compassion-Focused Theory (CFT), and estimated treatment effects in a population of adults with skin conditions and associated psychological distress. Design: A randomized-controlled design was used, with 176 participants being allocated to either CFT-based self-help or a waitlist control group, who received usual medical care. The two-week intervention was provided by email. Main Outcome Measures: Treatment adherence and attrition rates were calculated, and effectiveness was estimated using measures of perceived stress, anxiety, depression, dermatology-specific quality of life and self-compassion. Results: Eighty-seven participants completed the post-intervention questionnaires (51%), and practiced on a median of 9/14 days. Study completers demonstrated significant, moderate improvements on measures of stress, anxiety, depression, self-compassion and dermatology-specific quality of life, relative to controls. In intention-to-treat (ITT) analyses, these findings remained significant, however effect sizes reduced from moderate to small. Conclusions: The findings indicate that CFT self-help shows promise in the treatment of psychological distress associated with skin conditions, however further testing of the intervention is not feasible without significant methodological changes, including the method of treatment delivery. Future studies should also include a follow-up period, as the duration of treatment effects could not be shown

    Coulomb drag as a signature of the paired quantum Hall state

    Motivated by the recent Coulomb drag experiment of M. P. Lilly et. al, we study the Coulomb drag in a two-layer system with Landau level filling factor ν=1/2\nu=1/2. We find that the drag conductivity in the incompressible paired quantum Hall state at zero temperature can be finite. The drag conductivity is also greatly enhanced above TcT_c, at which the transition between the weakly coupled compressible liquids and the paired quantum Hall liquid takes place.     Formation of giant iron oxide-copper-gold deposits by superimposed, episodic hydrothermal pulses

    Iron oxide-copper-gold deposits are a globally important source of copper, gold and critical commodities. However, they possess a range of characteristics related to a variety of tectono-magmatic settings that make development of a general genetic model challenging. Here we investigate micro-textural and compositional variations in actinolite, to constrain the thermal evolution of the Candelaria iron oxide-copper-gold deposit in Chile. We identify at least two mineralization stages comprising an early 675–800 °C iron oxide-apatite type mineralization overprinted by a later copper-rich fluid at around 550–700 °C. We propose that these distinct stages were caused by episodic pulses of injection of magmatic-hydrothermal fluids from crystallizing magmas at depth. We suggest that the mineralisation stages we identify were the result of temperature gradients attributable to changes in the magmatic source, rather than variations in formation depth, and that actinolite chemistry can be used as a proxy for formation temperature in iron oxide-copper-gold systems

    Triple oral beta-lactam containing therapy for Buruli ulcer treatment shortening

    The potential use of clinically approved beta-lactams for Buruli ulcer (BU) treatment was investigated with representative classes analyzed in vitro for activity against Mycobacterium ulcerans. Beta-lactams tested were effective alone and displayed a strong synergistic profile in combination with antibiotics currently used to treat BU, i.e. rifampicin and clarithromycin; this activity was further potentiated in the presence of the beta-lactamase inhibitor clavulanate. In addition, quadruple combinations of rifampicin, clarithromycin, clavulanate and beta-lactams resulted in multiplicative reductions in their minimal inhibitory concentration (MIC) values. The MIC of amoxicillin against a panel of clinical isolates decreased more than 200-fold within this quadruple combination. Amoxicillin/clavulanate formulations are readily available with clinical pedigree, low toxicity, and orally and pediatric available; thus, supporting its potential inclusion as a new anti-BU drug in current combination therapies

    Dynamics near the Surface Reconstruction of W(100)

    Using Brownian molecular dynamics simulation, we study the surface dynamics near the reconstruction transition of W(100) via a model Hamiltonian. Results for the softening and broadening of the surface phonon spectrum near the transition are compared with previous calculations and with He atom scattering data. From the critical behavior of the central peak in the dynamical structure factor, we also estimate the exponent of the power law anomaly for adatom diffusion near the transition temperature.Comment: 8 pages, 8 figures, to appear in Phys.     Adsorption-desorption kinetics in nanoscopically confined oligomer films under shear

    The method of molecular dynamics computer simulations is employed to study oligomer melts confined in ultra-thin films and subjected to shear. The focus is on the self-diffusion of oligomers near attractive surfaces and on their desorption, together with the effects of increasing energy of adsorption and shear. It is found that the mobility of the oligomers near an attractive surface is strongly decreased. Moreover, although shearing the system forces the chains to stretch parallel to the surfaces and thus increase the energy of adsorption per chain, flow also promotes desorption. The study of chain desorption kinetics reveals the molecular processes responsible for the enhancement of desorption under shear. They involve sequences of conformations starting with a desorbed tail and proceeding in a very fast, correlated, segment-by-segment manner to the desorption of the oligomers from the surfaces.
