    Rail roughness and rolling noise in tramways

    Companies which manage railway networks have to cope continually with the problem of operating safety and maintenance intervention issues related to rail surface irregularities. A lot of experience has been gained in recent years in railway applications but the case of tramways is quite different; in this field there are no specific criteria to define any intervention on rail surface restoration. This paper shows measurements carried out on some stretches of a tram network with the CAT equipment (Corrugation Analysis Trolley) for the principal purpose of detecting different states of degradation of the rails and identifying a level of deterioration to be associated with the need for maintenance through rail grinding. The measured roughness is used as an input parameter into prediction models for both rolling noise and ground vibration to show the potential effect that high levels of roughness can have in urban environment. Rolling noise predictions are also compared with noise measurements to illustrate the applicability of the modelling approach. Particular attention is given to the way the contact filter needs to be modelled in the specific case of trams that generally operate at low speed. Finally an empirical approach to assess vibration levels in buildings is presented

    Successful Transitions of Runaway/Homeless Youth from Shelter Care

    Previous research indicates that runaway and homeless youth often achieve positive outcomes after shelter stays however few studies have examined how these outcomes are achieved. This study employs qualitative methods to explicate this phenomenon. Twenty-five providers and 21 youth from four shelters participated in this study. Youth were recruited who had completed shelter care and returned home for minimally six months. Multiple raters identified themes and created a conceptual model. While in shelter, youths experienced structure and freedom, and the family experienced respite. Once youth became involved in treatment, the family re-connected and the youth returned home. After returning home, youth and family become involved in follow-up services. Results from our study provide insight into the process through which runaway/homeless youth return home after a shelter stay. Our findings emphasize the need for continued change by all members of the family system, highlighting the need for continued intervention to maintain positive changes

    Himmelfarb 911! Using LibGuides and Scenarios to Inform and Train Library Staff on Disaster Response

    Objectives Create and update disaster planning and response materials so that they are accessible at time of need and educate the library\u27s staff on the library\u27s safety and emergency response policies and procedures. Introduction This urban health sciences library has experienced a number of potentially serious disaster incidents in the local region in the last fifteen years including terrorist attack, earthquake, fire, tornado, and hurricane. The library has a standing Disaster Planning committee which has produced extensive policy and planning documents for disaster response including a Disaster Planning and Response Manual, a Continuity of Operations Plan, a Pocket Response Plan, and a staff telephone tree. The accessibility and usability of these documents along with staff training on emergency policies and procedures are a high priority. Methods During the last two fiscal years, the standing committee has engaged in initiatives to educate staff members and provide a stable, cloud based, centralized platform for resources. A Himmelfarb911 LibGuide was established in fall of 2013 and includes all of the library\u27s disaster documents, links to outside resources, and photo documentation of facilities, collections, and art work in the Library. Online availability of resources complements printed copies which are held at key locations both on- and off-site. In addition, the committee has engaged staff in disaster scenarios (Snowmaggeddon, severe thunderstorm, active shooter, etc.) at meetings to discuss appropriate responses and resources. The committee has also organized training with the Univerisity\u27s Office of Safety and Security to brief staff on evacuation and shelter in place procedures, and other important safety information. Results The disaster scenarios elicited enthusiastic response and discussion among library staff members and provided an opportunity for staff to ask questions and help refine library policies. Both initiatives help ensure that staff will be well prepared for whatever disasters or emergencies they might face on the job

    Cloning of the Complete Gene for Carcinoembryonic Antigen

    Carcinoembryonic antigen (CEA) is a widely used tumor marker, especially in the surveillance of colonic cancer patients. Although CEA is also present in some normal tissues, it is apparently expressed at higher levels in tumorous tissues than in corresponding normal tissues. As a first step toward analyzing the regulation of expression of CEA at the transcriptional level, we have isolated and characterized a cosmid clone (cosCEA1), which contains the entire coding region of the CEA gene. A close correlation exists between the exon and deduced immunoglobulin-like domain borders. We have determined a cluster of transcriptional starts for CEA and the closely related nonspecific cross-reacting antigen (NCA) gene and have sequenced their putative promoters. Regions of sequence homology are found as far as approximately 500 nucleotides upstream from the translational starts of these genes, but farther upstream they diverge completely. In both cases we were unable to find classic TATA or CAAT boxes at their expected positions. To characterize the CEA and NCA promoters, we carried out transient transfection assays with promoter-indicator gene constructs in the CEA-producing adenocarcinoma cell line SW403, as well as in nonproducing HeLa cells. A CEA gene promoter construct, containing approximately 400 nucleotides upstream from the translational start, showed nine times higher activity in the SW403 than in the HeLa cell line. This indicates that cis-acting sequences which convey cell type-specific expression of the CEA gene are contained within this region

    A yield comparison between small-plot and on-farm foliar fungicide trials in soybean and maize

    Agronomic research provides management recommendations based on small-plot trials (SPTs) and on-farm trials (OFTs) with very different characteristics. SPTs are traditionally conducted at agricultural experiment stations by research institutes or universities, while OFTs are conducted under commercial-scale conditions and managed by farmers using their own equipment. Several researchers claimed that discrepancies could occur between these two types of trials, which can make the extrapolation of results from SPTs to the farm level difficult. In our study, we conducted an extensive comparison of small-plot and on-farm trials to analyze the effect of foliar fungicide application on maize and soybean yields. We collected data on maize and soybean from five US states. Analysis of the soybean data showed similar mean yield responses and within-trial standard deviation to fungicide application between 479 OFTs and 83 SPTs. For maize, our comparison of 300 OFTs and 114 SPTs showed similar mean yield response in both. Nevertheless, the within-trial standard deviation was three times smaller in on-farm compared to small-plot trials. On the other hand, the between-trial standard deviation (measuring the variability of the effects of fungicide application across different environments) was almost twice as large in SPTs than in OFTs for both crops. Hence, the differences in the effects of fungicide on yield were similar whether they were estimated using OFTs or SPTs for both crops. This implies that OFTs can potentially detect significant yield differences with fewer replicates and thus reduce the cost of data generation. We argue that SPTs can be seen as a preliminary step before scaling up to OFTs to facilitate technology transfer and extrapolate the results in real farming conditions

    Field validation of a farmer supplied data approach to close soybean yield gaps in the US North Central region

    CONTEXT: Producer-reported data can be used to identify suites of management practices that lead to higher yield and profit. However, a rigorous validation of the approach in relation to its potential impact on farmer yield and profit is lacking. OBJECTIVE: This study aimed to validate a producer-data approach on its capability to guide on-farm evaluation of management practices with greatest potential for increasing producer yield and profit. We show proof of concept using soybean in the North Central US region as a case study. METHODS: We used a combination of regression tree analysis and a spatial framework to determine practices with highest influence on yield for specific climate domains across the region. These practices were used as a basis for designing an ‘improved’ management package for each domain. The impact associated with adoption of the ‘improved’ management package on producer yield, seed constituents, and profit was evaluated against a ‘reference’ treatment that follows farmer management via replicated on-farm trials across 100 sites over two crop seasons. RESULTS AND CONCLUSIONS: Average yield was 278 kg ha-1 higher in the improved versus reference management, equivalent to a closure of the current exploitable yield gap by 40%. In turn, adoption of the improved management led to an average increase of $76 ha-1 in net profit. Sensitivity analysis showed that adoption of the improved management packages should increase farmer profit across a wide range of grain price scenarios, with very small downside risk. Seed protein concentration was negatively associated with the positive yield advantage of the improved management, whereas seed oil concentration tended to increase. SIGNIFICANCE: Analysis of producer data can accelerate discovery, evaluation, and adoption of suites of management practices that consistently lead to higher farmer yield and profit, which, in turn, would help speed up current rates of yield gain

    Sequence and the Developmental and Tissue-Specific Regulation of the First Complete Vitellogenin Message From Ticks

    The first full-length mRNA for vitellogenin (Vg) from ticks was sequenced. This also represents the first complete sequence of Vg from the Chelicerata and of a heme binding Vg. The Vg cDNA from the American dog tick, Dermacentor variabilis was 5744nt in length (GenBank Accession number AY885250), which coded for a protein of 1843aa with a calculated molecular weight of 208kD. This protein had an 18 aa signal sequence, a single RXXR cleavage signal that would generate two subunits (49.5 and 157K in molecular weight) and lipoprotein N-terminal and carboxy von Willebrand factor type D domains. Tryptic digest MS analysis of vitellin protein confirmed the function of the cDNA as the tick yolk protein. Apparently, vitellin in D. variabilis is oligomeric (possibly dimeric) and is comprised of a mixture of the uncleaved monomer and subunits that were predicted from the single RXXR cleavage signal. The highly conserved GL/ICG motif close to the C-terminus in insect Vg genes was different in the tick Vg message, i.e., GLCS. This variant was also present in a partial sequence of Vg from Boophilus microplus. Phylogenic analysis showed that the full length Vg cDNA from D. variabilis and the partial cDNA from B. microplus were distinct from insects and Crustacea. The Vg message was not found in whole body RNA from unfed or fed males or in unfed and partially fed (virgin) females as determined by Northern blotting. The message was found in replete (mated) pre-ovipositional females, increased to higher levels in ovipositing females and was absent after egg laying was complete. The endocrine regulation of the Vg mRNA is discussed. The tissue sources of the Vg message are both the gut and fat body. Tryptic digest MS fingerprinting suggests that a second Vg mRNA might be present in the American dog tick, which needs further study

    Planning, implementation and scientific goals of the Studies of Emissions and Atmospheric Composition, Clouds and Climate Coupling by Regional Surveys (SEAC4RS) field mission

    The Studies of Emissions and Atmospheric Composition, Clouds and Climate Coupling by Regional Surveys (SEAC4RS) field mission based at Ellington Field, Texas, during August and September 2013 employed the most comprehensive airborne payload to date to investigate atmospheric composition over North America. The NASA ER-2, DC-8, and SPEC Inc. Learjet flew 57 science flights from the surface to 20 km. The ER-2 employed seven remote sensing instruments as a satellite surrogate and eight in situ instruments. The DC-8 employed 23 in situ and five remote sensing instruments for radiation, chemistry, and microphysics. The Learjet used 11 instruments to explore cloud microphysics. SEAC4RS launched numerous balloons, augmented AErosol RObotic NETwork, and collaborated with many existing ground measurement sites. Flights investigating convection included close coordination of all three aircraft. Coordinated DC-8 and ER-2 flights investigated the optical properties of aerosols, the influence of aerosols on clouds, and the performance of new instruments for satellite measurements of clouds and aerosols. ER-2 sorties sampled stratospheric injections of water vapor and other chemicals by local and distant convection. DC-8 flights studied seasonally evolving chemistry in the Southeastern U.S., atmospheric chemistry with lower emissions of NOx and SO2 than in previous decades, isoprene chemistry under high and low NOx conditions at different locations, organic aerosols, air pollution near Houston and in petroleum fields, smoke from wildfires in western forests and from agricultural fires in the Mississippi Valley, and the ways in which the chemistry in the boundary layer and the upper troposphere were influenced by vertical transport in convective clouds

    A Two-Phase Innate Host Response to Alphavirus Infection Identified by mRNP-Tagging In Vivo

    A concept fundamental to viral pathogenesis is that infection induces specific changes within the host cell, within specific tissues, or within the entire animal. These changes are reflected in a cascade of altered transcription patterns evident during infection. However, elucidation of this cascade in vivo has been limited by a general inability to distinguish changes occurring in the minority of infected cells from those in surrounding uninfected cells. To circumvent this inherent limitation of traditional gene expression profiling methods, an innovative mRNP-tagging technique was implemented to isolate host mRNA specifically from infected cells in vitro as well as in vivo following Venezuelan equine encephalitis virus (VEE) infection. This technique facilitated a direct characterization of the host defense response specifically within the first cells infected with VEE, while simultaneous total RNA analysis assessed the collective response of both the infected and uninfected cells. The result was a unique, multifaceted profile of the early response to VEE infection in primary dendritic cells, as well as in the draining lymph node, the initially targeted tissue in the mouse model. A dynamic environment of complex interactions was revealed, and suggested a two-step innate response in which activation of a subset of host genes in infected cells subsequently leads to activation of the surrounding uninfected cells. Our findings suggest that the application of viral mRNP-tagging systems, as introduced here, will facilitate a much more detailed understanding of the highly coordinated host response to infectious agents

    Greening of grey infrastructure should not be used as a Trojan horse to facilitate coastal development

    Climate change and coastal urbanization are driving the replacement of natural habitats with artificial structures and reclaimed land globally. These novel habitats are often poor surrogates for natural habitats. The application of integrated greening of grey infrastructure (IGGI) to artificial shorelines demonstrates how multifunctional structures can provide biodiversity benefits whilst simultaneously serving their primary engineering function. IGGI is being embraced globally, despite many knowledge gaps and limitations. It is a management tool to compensate anthropogenic impacts as part of the Mitigation Hierarchy. There is considerable scope for misuse and ‘greenwashing’ however, by making new developments appear more acceptable, thus facilitating the regulatory process. We encourage researchers to exercise caution when reporting on small-scale experimental trials. We advocate that greater attention is paid to when experiments ‘fail’ or yield unintended outcomes. We advise revisiting, repeating and expanding on experiments to test responses over broader spatio-temporal scales to improve the evidence base. Synthesis and applications. Where societal and economic demand makes development inevitable, particular attention should be paid to avoiding, minimizing and rehabilitating environmental impacts. Integrated greening of grey infrastructure (IGGI) should be implemented as partial compensation for environmental damage. Mutual benefits for both humans and nature can be achieved when IGGI is implemented retrospectively in previously developed or degraded environments. We caution, however, that any promise of net biodiversity gain from new developments should be scrutinized and any local ecological benefits set in the context of the wider environmental impacts. A ‘greened’ development will always impinge on natural systems, a reality that is much less recognized in the sea than on land.</p
