    Les jeux vidéo dans l'accompagnement éducatif: quelle utilisation par les éducateurs sociaux ?

    Ce travail de recherche vise à éclairer l’utilisation faite des jeux vidéo par les professionnels de l’éducation sociale. Il est composé de deux axes principaux, à savoir le développement de concepts théoriques et la recherche empirique. La première thématique développée dans le cadre conceptuel se rapporte aux jeux vidéo. Leurs caractéristiques et composantes sont précisées, leurs effets – tant positifs que négatifs – sont exposés et les joueurs adolescents suisses sont identifiés. La seconde thématique approfondie traite de l’adolescence, en termes de développement cognitif et d’accès au jugement moral, de développement identitaire et de socialisation. La troisième thématique évoquée concerne l’intervention socio- éducative, principalement en milieu institutionnel. Elle aborde les notions de placement institutionnel, d’accompagnement, de relation éducative ainsi que de projet éducatif. De ces approfondissements, la question de recherche a été ciblée. Celle-ci a été formulée de la manière suivante : « Quelle utilisation des jeux vidéo est faite par les éducateurs sociaux dans leur accompagnement éducatif ?

    Toll-like receptors in domestic animals

    Toll-like receptors are pattern recognition receptors with which hosts recognize pathogen-associated molecular patterns (PAMP). This recognition process is translated rapidly into a meaningful defense reaction. This form of innate host defense is preserved in the animal kingdom: invertebrates heavily depend on it; higher vertebrates also have an adaptive immune system. Both adaptive and innate immune systems are intertwined in that the former also depends on an intact innate recognition and response system. Members of the TLR system cover recognition of parasitic, bacterial or viral germs. Due to the constraints imposed by the necessity to recognize PAMP and to interact with downstream signaling molecules, the TLR system is relatively conserved in evolution. Nevertheless, subtle species differences have been reported for several mammalian TLR members. Examples of this will be given. In all mammalian species investigated, part of the coding sequence is available for the most important TLR members, thus allowing study of expression of these TLR members in various tissues by reverse-transcription polymerase chain reaction in its classical (RT-PCR) and quantitative real time RT-PCR (qRT-PCR) form. In some species, the whole coding sequences of the most important or even all TLR members are known. This allows construction of cDNA and transfection of common host cells, thus permitting functional studies. Extensive investigations were devoted to the study of non-synonymous single nucleotide polymorphisms.     Particle and particle pair dispersion in turbulence modeled with spatially and temporally correlated stochastic processes

    In this paper we present a new model for modeling the diffusion and relative dispersion of particles in homogeneous isotropic turbulence. We use an Heisenberg-like Hamiltonian to incorporate spatial correlations between fluid particles, which are modeled by stochastic processes correlated in time. We are able to reproduce the ballistic regime in the mean squared displacement of single particles and the transition to a normal diffusion regime for long times. For the dispersion of particle pairs we find a t2t^{2}-dependence of the mean squared separation at short times and a tt-dependence for long ones. For intermediate times indications for a Richardson t3t^{3} law are observed in certain situations.     Molecular Basis for Converting (2S)-Methylsuccinyl-CoA Dehydrogenase into an Oxidase

    Although flavoenzymes have been studied in detail, the molecular basis of their dioxygen reactivity is only partially understood. The members of the flavin adenosine dinucleotide (FAD)-dependent acyl-CoA dehydrogenase and acyl-CoA oxidase families catalyze similar reactions and share common structural features. However, both enzyme families feature opposing reaction specificities in respect to dioxygen. Dehydrogenases react with electron transfer flavoproteins as terminal electron acceptors and do not show a considerable reactivity with dioxygen, whereas dioxygen serves as a bona fide substrate for oxidases. We recently engineered (2S)-methylsuccinyl-CoA dehydrogenase towards oxidase activity by rational mutagenesis. Here we characterized the (2S)-methylsuccinyl-CoA dehydrogenase wild-type, as well as the engineered (2S)-methylsuccinyl-CoA oxidase, in detail. Using stopped-flow UV-spectroscopy and liquid chromatography-mass spectrometry (LC-MS) based assays, we explain the molecular base for dioxygen reactivity in the engineered oxidase and show that the increased oxidase function of the engineered enzyme comes at a decreased dehydrogenase activity. Our findings add to the common notion that an increased activity for a specific substrate is achieved at the expense of reaction promiscuity and provide guidelines for rational engineering efforts of acyl-CoA dehydrogenases and oxidases

    Paramyxoviruses constitute a large family of enveloped RNA viruses including important pathogens in veterinary and human medicine. Recently, feline paramyxoviruses, genus morbillivirus, were detected in cats from Hong Kong and Japan. Here we describe the discovery of several new feline paramyxoviruses. Infections with these diverse viruses were detected in urine samples from cats suffering from chronic kidney disease (CKD). No viral RNA was found in cats without clinical signs of uropathy highlighting an association between feline paramyxovirus (FPaV) infections and CKD. Phylogenetic analyses of the detected viruses showed that they represent at least two different species, one of them representing the feline morbilliviruses detected previously in Hong Kong and Japan. In addition, a new FPaV was detected sharing only 73 % homology on the nucleotide level of the viral L-gene to currently known paramyxoviral species

    A prospective, randomized, blinded, placebo-controlled pilot study on the effect of Enterococcus faecium on clinical activity and intestinal gene expression in canine food-responsive chronic enteropathy

    BACKGROUND: Canine chronic enteropathies (CE) are believed to be caused by an aberrant immune response towards the intestinal microbiome. Administration of probiotics can alleviate colitis in people. In vitro effects of the probiotic Enterococcus faecium NCIMB 10415 E1707 (EF) previously have been evaluated using canine cells (e.g., whole blood, intestinal biopsies), but data on in vivo efficacy are lacking. HYPOTHESIS/OBJECTIVES: Administration of EF to dogs with food‐responsive CE will improve clinical outcome and decrease the intestinal inflammatory profile. ANIMALS: Dogs diagnosed with CE were prospectively recruited to receive a hydrolyzed elimination diet plus either a synbiotic product containing EF or placebo for 6 weeks. Both veterinary staff and owners were blinded to the treatment. METHODS: Clinical severity index (CCECAI), clinicopathological data and gene expression using intestinal biopsies (TLR2/4/5/9, IL‐17A, IL‐22, IL‐23p19, RORC, IL‐2, IL‐12p35, TNFα, IL‐4, IFNy, IL‐10, TGFβ, IL‐1β, IL‐18, NLRP3, casp‐1, TFF1, TFF3 and PPARy) before and after 6 weeks of treatment were analyzed using linear mixed modeling. RESULTS: Of the 45 cases recruited, 12 finished the clinical trial. Seven received the synbiotic and 5 the placebo product. There was no difference between groups or treatments regarding clinical efficacy, histology scores or expression of any of the investigated genes. CONCLUSIONS AND CLINICAL IMPORTANCE: Standard dietary treatment induced rapid clinical response in all cases. Because the study was underpowered, it was not possible to determine whether or not EF had an additional effect within the time period of 6 weeks
