    Secondary metabolite variability in lichen genus Usnea in India: A potential source for bioprospection

    The species of lichen genus Usnea are well known for their worldwide distribution and medicinal properties due to presence of usnic acid. The genus is represented by more than 300 species in the world out of which 57 species are reported from India. In India, the species of Usnea grows profusely in higher altitudes of Western Ghats and Himalayan regions. 20 different secondary metabolites are known from Indian Usnea species which have potential biological and physiological significance. Salazinic acid, stictic acid, norstictic acid and barbatic acid are the most dominant secondary metabolites along with usnic acid. Apart from the natural thalli the cultivated mycobiont of Usnea species also produced many of these compounds in culture conditions. The secondary compounds present in Usnea species are significant source of natural products for commercial uses

    Ca(2+) contributes to a myriad of important cellular processes in all organisms, including the apicomplexans, Plasmodium and Toxoplasma. Due to its varied and essential roles, free Ca(2+) is tightly regulated by complex mechanisms. These mechanisms are therefore of interest as putative drug targets. One pathway in Ca(2+) homeostatic control in apicomplexans uses a Ca(2+)/H(+) exchanger (a member of the cation exchanger family, CAX). The P. falciparum CAX (PfCAX) has recently been characterised in asexual blood stage parasites. To determine the physiological importance of apicomplexan CAXs, tagging and knock-out strategies were undertaken in the genetically tractable T. gondii and P. berghei parasites. In addition, a yeast heterologous expression system was used to study the function of apicomplexan CAXs. Tagging of T. gondii and P. berghei CAXs (TgCAX and PbCAX) under control of their endogenous promoters could not demonstrate measureable expression of either CAX in tachyzoites and asexual blood stages, respectively. These results were consistent with the ability of parasites to tolerate knock-outs of the genes for TgCAX and PbCAX at these developmental stages. In contrast, PbCAX expression was detectable during sexual stages of development in female gametocytes/gametes, zygotes and ookinetes, where it was dispersed in membranous networks within the cytosol (with minimal mitochondrial localisation). Furthermore, genetically disrupted parasites failed to develop further from "round" form zygotes, suggesting that PbCAX is essential for ookinete development and differentiation. This impeded phenotype could be rescued by removal of extracellular Ca(2+). Therefore, PbCAX provides a mechanism for free living parasites to multiply within the ionic microenvironment of the mosquito midgut. Ca(2+) homeostasis mediated by PbCAX is critical and suggests plasmodial CAXs may be targeted in approaches designed to block parasite transmission

    Editing talking-head video to change the speech content or to remove filler words is challenging. We propose a novel method to edit talking-head video based on its transcript to produce a realistic output video in which the dialogue of the speaker has been modified, while maintaining a seamless audio-visual flow (i.e. no jump cuts). Our method automatically annotates an input talking-head video with phonemes, visemes, 3D face pose and geometry, reflectance, expression and scene illumination per frame. To edit a video, the user has to only edit the transcript, and an optimization strategy then chooses segments of the input corpus as base material. The annotated parameters corresponding to the selected segments are seamlessly stitched together and used to produce an intermediate video representation in which the lower half of the face is rendered with a parametric face model. Finally, a recurrent video generation network transforms this representation to a photorealistic video that matches the edited transcript. We demonstrate a large variety of edits, such as the addition, removal, and alteration of words, as well as convincing language translation and full sentence synthesis

    Work in Progress -- Does a Technology-Rich Transportation Engineering Experiences Increase Interest in Civil Engineering

    This paper examines whether a technology-rich transportation engineering experience generates interest among college freshman students towards transportation related degrees and careers, specifically, whether it increases interest in civil engineering. Louisiana Tech’s first-year engineering experience provides relevant, project-focused education for over 500 students each year and an opportunity to introduce transportation-related projects. The six semester-hour, three-course sequence, called “Living with the Lab,” boosts experiential learning through student ownership of inexpensive laboratory equipment. In one section of the Living with the Lab course, a transportation engineer visited the class and the students were led through a technology-rich transportation engineering activity. They used an ultrasonic distance sensor to build a device to measure traffic speed. At the end of the term, students were asked to rate their interest level in 22 fields of study (one of which is civil engineering) from “not at all interested” (1) to “extremely interested” (7). Asking for interest level in each field provides finer resolution than only looking at which major a student selects. Interest levels in civil engineering of the section that did the transportation engineering activity were slightly higher than those of the other sections, but the difference was not statistically significant

    Evaluation of the Immunoprotective Potential of Recombinant Paraflagellar Rod Proteins of Trypanosoma evansi in Mice

    Trypanosomosis, caused by Trypanosoma evansi, is an economically significant disease of livestock. Systematic antigenic variation by the parasite has undermined prospects for the development of a protective vaccine that targets the immunodominant surface antigens, encouraging exploration of alternatives. The paraflagellar rod (PFR), constituent proteins of the flagellum, are prominent non-variable vaccine candidates for T. evansi owing to their strategic location. Two major PFR constituent proteins, PFR1 (1770bp) and PFR2 (1800bp), were expressed using Escherichia coli. Swiss albino mice were immunized with the purified recombinant TePFR1 (89KDa) and TePFR2 (88KDa) proteins, as well as with the mix of the combined proteins at equimolar concentrations, and subsequently challenged with virulent T. evansi. The PFR-specific humoral response was assessed by ELISA. Cytometric bead-based assay was used to measure the cytokine response and flow cytometry for quantification of the cytokines. The recombinant TePFR proteins induced specific humoral responses in mice, including IgG1 followed by IgG2a and IgG2b. A balanced cytokine response induced by rTePFR 1 and 2 protein vaccination associated with extended survival and improved control of parasitemia following lethal challenge. The observation confirms the immunoprophylactic potential of the covert antigens of T. evansi

    Integrated nutrient management in calendula (Calendula officinalis L.) grown in partially reclaimed sodic soil condition

    To study the effect of nutrient management on growth and flower yield of Calendula, field experiments were carried out during 2009 and 2010. Results indicated that significantly higher plant height (40.6 cm), number of leaves (142) at 90 days, total number of flowers plant-1 (126), total fresh weight of flowers (87.5 q ha-1) and dry weight of flowers (25.0 q ha-1) were observed by the application of 10 t farmyard manure (FYM) + ½ NPK + spraying of micronutrients followed by sole application of recommended dose of inorganic fertilizer (i.e. NPK@ 80:30:30 kg ha-1) which was at par with application of half dose of recommended dose of inorganic fertilizer supplemented with half dose of organic fertilizer. Application of 20 t FYM ha-1 improved the soil physicochemical parameters i.e. pH, EC, organic carbon and available NPK in comparison to control. &nbsp

Medicinal plants survive very well in the current scheme of crop diversification in various types of agro-climatic conditions of India. Agronomists are posed with challenge of scientifically fitting most suitable medicinal plants in different ago-climatic regions. Among the losses caused by different pests in the agriculture, the weeds account for about 45% and it may be more or less equal in the case of medicinal plants. Integrated weed management increases the factor productivity, income of the farmer, quality of produce and is eco-friendly in nature. By taking examples of two important medicinal plants viz. Satawar (Asparagus racemosus Willd.) and Kalmegh (Andrographis paniculata Nees.), the present review discusses the different methods of weed management and how they may be integrated to develop new paradigm as an integrated weed management

    An integrable model for SU(ν\nu) electrons with inverse-square interaction is studied for the system with confining harmonic potential. We develop a new description of the spectrum based on the {\it renormalized harmonic-oscillators} which incorporate interaction effects via the repulsion of energy levels.
