    Mental Strategies of Elite High Altitude Climbers: Overcoming Adversity on Mount Everest

    The purpose of this study was to explore the mental strategies used by elite Mount Everest climbers to overcome obstacles while ascending and descending the mountain. Individual interviews were carried out with 10 climbers who have successfully reached the summit of Mount Everest. Common strategies of success were identified, as well as factors that created difficulty on the mountain. The themes of focus, mental toughness, short-term goals, drawing on past experience, connecting to one\u27s body, feeling support from other climbers, and belief in personal capacities were seen to have importance in overcoming adversity on the mountain. These results support mental training as an effective tool for overcoming obstacles in the high stress environment of high altitude climbing

    The Residual Income Approach to Housing Affordability: The Theory and the Practice

    The residual income approach to housing affordability is one that looks at what different household types can afford to spend on housing after taking into account the other necessary expenditures of living. It is an alternative to benchmark measures of affordability as used in social housing rent setting in Australia (the 25% rule) or assessing the overall affordability in the wider housing market (the 30/40 rule) as commonly used by a range of housing affordability researchers in Australia. This Positioning Paper does two things. Part A provides an overview, using existing literature, of the various semantic, substantive and definitional issues around the notion of affordability, leading to an argument in support of the soundness of the residual income approach. This overview is set in the historical contexts of discussions about affordability measures in the US, UK and Australia. Part B is methodological; it shows for various household types and income ranges, both for home purchase and rental, how the residual income method can be operationalised and its potential policy applications. This is still work in progress and there may be minor refinements to the method by the time of the production of the Final Report. However, we are confident that the method is sufficiently robust at this point to indicate its substantial potential as a problem identification and policy tool

    The house sparrow (Passer domesticus) is an important model species in ecology and evolution. However, until recently, genomic resources for molecular ecological projects have been lacking in this species. Here, we present transcriptome sequencing data (RNA-Seq) from three different house sparrow tissues (spleen, blood and bursa). These tissues were specifically chosen to obtain a diverse representation of expressed genes and to maximize the yield of immune-related gene functions. After de novo assembly, 15250 contigs were identified, representing sequence data from a total of 8756 known avian genes (as inferred from the closely related zebra finch). The transcriptome assembly contain sequence data from nine manually annotated MHC genes, including an almost complete MHC class I coding sequence. There were 407, 303 and 68 genes overexpressed in spleen, blood and bursa, respectively. Gene ontology terms related to ribosomal function were associated with overexpression in spleen and oxygen transport functions with overexpression in blood. In addition to the transcript sequences, we provide 327 gene-linked microsatellites (SSRs) with sufficient flanking sequences for primer design, and 3177 single-nucleotide polymorphisms (SNPs) within genes, that can be used in follow-up molecular ecology studies of this ecological well-studied species

    Size and Distribution of Resting Eggs in a Natural Population of the Rotifer Brachionus plicatilis

    Resting eggs of the rotifer Brachionus plicatilis were collected from the sediments of a brackish-water pond near Tampa, Florida. These core samples were examined for resting egg density and size. Resting egg density decreased exponentially from the surface down to 7 cm in the sediments at all stations, with depth in the sediments accounting for 42.3% of the total variance. The maximum resting egg density recorded was 194 RE/cm3 with no significant differences in density among stations. Resting eggs were found to be significantly larger in the top 2 cm of sediment than resting eggs from deeper in the sediments. These data are discussed with regard to other work on resting egg densities in natural populations. Hypotheses are offered to explain the observed decrease in resting egg size with depth in sediments

    Enhanced cross-species utility of conserved microsatellite markers in shorebirds

    Background: Microsatellite markers are popular genetic markers frequently used in forensic biology. Despite their popularity, the characterisation of polymorphic microsatellite loci and development of suitable markers takes considerable effort. Newly-available genomic databases make it feasible to identify conserved genetic markers. We examined the utility and characteristics of conserved microsatellite markers in Charadriiformes (plovers, sandpipers, gulls and auks). This order harbours many species with diverse breeding systems, life histories and extraordinary migration biology whose genetics warrant investigation. However, research has been largely restrained by the limited availability of genetic markers. To examine the utility of conserved microsatellite loci as genetic markers we collated a database of Charadriiformes microsatellites, searched for homologues in the chicken genome and tested conserved markers for amplification and polymorphism in a range of charadriiform species. Results: Sixty-eight (42%) of 161 charadriiform microsatellite loci were assigned to a single location in the chicken genome based on their E-value. Fifty-five primers designed from conserved microsatellite loci with an E-value of E-10 or lower amplified across a wider range of charadriiform species than a control group of primers from ten anonymous microsatellite loci. Twenty-three of 24 examined conserved markers were polymorphic, each in on average 3 of 12 species tested. Conclusion: Genomic sequence databases are useful tools to identify conserved genetic markers including those located in non-coding regions. By maximising primer sequence similarity between source species and database species, markers can be further improved and provide additional markers to study the molecular ecology of populations of non-model organisms

    Comparison between Normalised and Unnormalised 454-Sequencing Libraries for Small-Scale RNA-Seq Studies

    Next-generation sequencing of transcriptomes (RNA-Seq) is being used increasingly in studies of nonmodel organisms. Here, we evaluate the effectiveness of normalising cDNA libraries prior to sequencing in a small-scale study of the zebra finch. We find that assemblies produced from normalised libraries had a larger number of contigs but used fewer reads compared to unnormalised libraries. Considerably more genes were also detected using the contigs produced from normalised cDNA, and microsatellite discovery was up to 73% more efficient in these. There was a positive correlation between the detected expression level of genes in normalised and unnormalised cDNA, and there was no difference in the number of genes identified as being differentially expressed between blood and spleen for the normalised and unnormalised libraries. We conclude that normalised cDNA libraries are preferable for many applications of RNA-Seq and that these can also be used in quantitative gene expression studies
