    Análise de variáveis não diretamente observáveis: influência na tomada de decisão durante o processo de investigação

    El tamaño de la muestra, el tipo de variables, su medida y la construcción de instrumentos para la recogida de datos válidos y fiables son aspectos a considerar en el proceso de investigación. En el ámbito de las ciencias sociales, de la salud y particularmente en el área de enfermería, los instrumentos para la recogida de datos son muchas veces compuestos por variables componentes o indicadores que originan variables latentes o no observables directamente, lo que muestra la importancia de decidir cuidadosamente cómo se miden (escala ordinal, Likert o de tipo Likert). Las escalas psicométricas son ejemplos de instrumentos, por el tipo de variables que lo componen, que pueden traer problemas de medición y de análisis estadístico (test paramétricos versus no paramétricos). Así, el investigador cuando usa estas variables, debe respetar algunos supuestos basados en estudios de simulación o recomendaciones basadas en la evidencia científica, lo que permite una mejor toma de decisiones.The sample dimension, types of variables, format used for measurement, and construction of instruments to collect valid and reliable data must be considered during the research process. In the social and health sciences, and more specifically in nursing, data-collection instruments are usually composed of latent variables or variables that cannot be directly observed. Such facts emphasize the importance of deciding how to measure study variables (using an ordinal scale or a Likert or Likert-type scale). Psychometric scales are examples of instruments that are affected by the type of variables that comprise them, which could cause problems with measurement and statistical analysis (parametric tests versus non-parametric tests). Hence, investigators using these variables must rely on suppositions based on simulation studies or recommendations based on scientific evidence in order to make the best decisions.A dimensão da amostra, o tipo de variáveis, o seu formato de medida, a construção dos instrumentos de recolha de dados válidos e fiáveis, são aspectos a ter em consideração no processo de investigação. No âmbito das ciências sociais, da saúde e especificamente na área de enfermagem, os instrumentos de recolha de dados são muitas vezes compostos por variáveis componentes ou indicadores que dão origem a variáveis latentes ou não observáveis diretamente, daí a importância da decisão relativa à forma como são medidas (escala ordinal, Likert ou de tipo Likert). As escalas psicométricas são exemplos de instrumentos, pelo tipo de variáveis que as integram, que podem trazer problemas de medida e de análise estatística (testes paramétricos versus não paramétricos). Assim o investigador quando usa estas variáveis deve respeitar alguns pressupostos baseados em estudos de simulação ou em recomendações fundamentadas na evidência científica, de forma a tomar a melhor decisão

    GROUND REACTION FORCE AND PLANTAR PRESSURE OF TWO JUMPING HEAD-OUT AQUATIC EXERCISES

    The aim of this study was to compare the biomechanical load, measured by the vertical ground reaction force (GRF), plantar pressure, and contact area of two jumping head-out aquatic exercises (cross country ski and jumping jacks) performed at 1.30 m water depth. Ten healthy male volunteers, with mean (SD) age 26.5 (4.2) yr, height 175.8 (2.6) cm, weight 73.7 (8.5) kg, body mass index 22.3 (2.5) kg.m-2 participated in this study. Data from both GRF and plantar pressures were obtained by using an insole system. Pairedsamples t-tests were conducted to compare each variable between conditions. Significant differences between exercises were found in all variables (peak force, t(9)=2.52, p = 0.033; peak pressure, t(9)=2.41, p = 0.040 and contact area, t(9)=-2.60, p = 0.029). Jumping exercises can be a suitable option when prescribing water-based exercises

    In situ formation of gold nanoparticles in polymer inclusion membrane: Application as platform in a label-free potentiometric immunosensor for Salmonella typhimurium detection

    Polymeric ion selective electrodes are highly sensitive to changes in zero current ion flow and this offers a route to signal amplification in label-free potentiometric immunosensors. In this work, a label-free potentiometric immunosensor toward Salmonella typhimurium (ST) assembled in a home-made pipette-tip electrode is described. The signal-output amplification was implemented on a gold nanoparticle polymer inclusion membrane (AuNPs-PIM) which was used as sensing platform and for antibody immobilization. Additionally, a marker ion was used to detect the antibody-antigen binding event at the electrode surface. The immunosensor construction was performed in several steps: i) gold salt ions extraction in PVC membrane; ii) AuNPs formation using Na2EDTA as reduction agent; iii) antibody anti-Salmonella conjugation on AuNPs-PIM in pipette-tip electrodes. The potential shift observed in potentiometric measurements was derived simply from the blocking effect in the ionic flux caused by antigen-antibody conjugation, without no extra steps, mimetizing the ion-channel sensors. A detection limit of 6 cells mL-1 was attained. As proof-of-concept, recovery studies were performed in spiked commercial apple juice samples with success. Due to the simplicity of use, the appealing cost of equipment and sensor production and being able to provide a quick analytical response (less than 1 h for a complete assay, including sample preparation for analysis), this scheme represents a good prototype device for the detection of foodborne pathogens like ST or other immune-responsive bacteria.This work received financial support from the European Union (FEDER funds through COMPETE) and National Funds from Portugal; FCT-Fundação para a Ciência e a Tecnologia through projects UID/QUI/50006/2013 and Norte-01-0145-FEDER-000011-RL1-QUALIFOOD.     We found that production parameters of a European Portuguese corpus are influenced by body composition: more prominently in the frequency parameters (f0, DP f0 and formants), perturbation parameters and noise parameters, for vowel segments [a] and [i]. We concluded that extralinguistic patterns of voice quality were significantly correlated with body composition and somatotype.
