    Diversity of fungal endophytic community in Quercus suber L. under different climate scenarios

    Cork oak (Quercus suber L.) is an evergreen oak tree species, typically found throughout the Mediterranean Basin that presents a great ecological and economic importance in Portugal. The ecological importance is due to the role in the protection of the soil, biodiversity control, water quality and oxygen production. The cork production and processing confer cork oak a great economic relevance in Portugal. An increase of cork oak decline due to biotic and abiotic stresses has been reported, which could damage the ecosystem and lower cork production. Fungal species as D;plod;a corticola and Biscogniauxia mediterranea are cork oak pathogens that cause tree decline and affect cork productivity. The incidence of both diseases seems to increase when trees are under several environmental stresses, such as increased temperatures or drought. In this work, fungal endophytic species of cork oak were collected in different sites of Portugal (Bragança, Gerês, Alcobaça, Grândola e Gavião). The collection sites displayed differences in water availability and cork oak trees in different phytosanitary conditions. The community of fungal endophytes of young twigs and one/two-years old branches were studied by detecting fungal outgrowing from aseptic plant pieces and grouping them into morphotypes. A differentiation between live and death (xylem) branch tissue was considered by culturing the most superficial branch wood or the most inner wood. Collection sites with higher precipitation seem to display a more diverse community. Branches displayed a more diverse fungal community than young twigs, being the endophytic community, present in inner wood, more similar between sites, than young twigs or outer branch tissues. This study allowed the comparison of fungal endophytic communities living in cork oak trees with different water availability levels and disease symptoms.     Oscillating Economic Sphere: Sustainable Limits of the Biosphere in the Face of Climate Change

    This paper proposes a new geometric model to facilitate the understanding of the complex interaction between the economic system and the limits of the biosphere. The concept of the oscillating economic sphere is based on a central parameter of the economy, the Gross Domestic Product (GDP). In the model a hypothetical sphere was built, in which its radius (re) corresponds to the world GDP (economic sphere), and another hypothetical sphere with radius (rb) is also introduced to represent the size of the biosphere (sphere of life), on this geometric construction, these spheres are concentric (rb > re). Therefore, an expansion of the economic sphere will occupy the biosphere, since the fundamentals of economy predict unlimited growth of GDP. Thus we will have a limit to be reached because the biophysical forces oppose this expansion, creating harmful environmental effects such as climate change

    First association of scale insects (Hemiptera: Diaspididae) with \u3ci\u3eSalacia crassifolia\u3c/i\u3e (Mart. Ex Schult.) G. Don. (Celastraceae)

    Salacia crassifolia (Mart. ex Schult.) G.Don., (Celastraceae) is a native species of shrub/tree highly ap­preciated in Brazil for its fruits and medicinal properties. Scale insects have never been reported associated with S. crassifolia; nevertheless, this paper describes the occurrence of two diaspidids on S. crassifolia leaves in Brazil. Three mature trees were inspected in February and March 2018 and scale insect samples were collected and pre­served in 70% alcohol, then mounted and identified under an optical microscope. Two species of scale insects were found associated with this plant, Pseudoparlatoria argentata Hempel and Melanaspis aristotelesi Lepage and Gi­annotti, both from the family Diaspididae (Hemiptera). The three observed trees were infested by the diaspidids, with some leaves completely colonized by both species. This is the first reported occurrence of P. argentata and M. aristotelesi in plants of the Celastraceae family. It is also the first report of these insects in the Distrito Federal, Brazil, expanding the distribution and hosts in native plant species of the Cerrado biome

    Thermal paint production: the techno-economic evaluation of muscovite as insulating additive.

    Muscovite is known by its thermal and electrical insulating properties. Based on this, it was hypothesized that its addition on paints should increase the thermal resistance. The use of muscovite as mineral insulating is pointed out as advantageous due to its low cost compared to other materials used for this purpose, such as the ceramic microsphere. The use of a low cost material could open the access to the medium and low income families, implying two aspects: the life quality increase by thermal comfort and the increase of energy saving. Thus, this part of the population could open a new market to thermal paints. Aiming to contribute to this issue, this work evaluated the thermal insulation performance of commercial paints containing muscovite additions and determined the economic evaluation for its industrial production. The thermal paint was formulated by adding 10%, 20% and 40% of muscovite to the commercial paint. This was applied on steel reinforced mortar boards. Thermal insulation tests were carried out in bench scale using an adapted box. The economic evaluation of the industrial production of muscovite-based thermal paint was conducted, considering the Brazilian economic market in this activity. The results showed its ability as an insulating agent due to a reduction of 0.667 °C/mm board by the addition of 40% muscovite. The economic analysis also demonstrated the feasibility of the thermal paint industrial production. The payback is favorable to 5 years when compared to the Selic short-term lending rate, with 21.53% of internal rate return and a net present value of US$ 15,085.76

The deposited article version contains attached the supplementary materials within the pdf.LysR-type transcriptional regulators (LTTR) are the most commonly found regulators in Burkholderia cepacia complex, comprising opportunistic pathogens causing chronic respiratory infections in cystic fibrosis (CF) patients. Despite LTTRs being global regulators of pathogenicity in several bacteria, few have been characterized in Burkholderia Here, we showed that gene ldhR of B. multivorans encoding a LTTR is co-transcribed with ldhA encoding a d-lactate dehydrogenase, and evaluate their implication in virulence traits like exopolysaccharide (EPS) synthesis and biofilm formation. Comparison of wild-type (WT) and its isogenic ΔldhR mutant grown in medium with 2% d-glucose revealed a negative impact on EPS biosynthesis and on cells' viability in the presence of LdhR. Loss of viability in WT cells was caused by intracellular acidification as consequence of cumulative organic acids secretion including d-lactate, this last one absent from the ΔldhR mutant supernatant. Furthermore, LdhR is implicated in the formation of planktonic cellular aggregates. WT cell aggregates reached 1000 μm after 24 hours in liquid cultures; in contrast to ΔldhR mutant aggregates that never grew more than 60 μm. Overexpression of d-lactate dehydrogenase LdhA in the ΔldhR mutant partially restored formed aggregates size, suggesting a role for fermentation inside aggregates. Similar results were obtained for surface-attached biofilms, with WT cells producing more biofilm. A systematic evaluation of planktonic aggregates in Burkholderia CF clinical isolates showed aggregates in 40 out of 74. As CF patients' lung environment is microaerophilic and bacteria are found as free aggregates/biofilms, LdhR and LdhA might have central roles in adaptation to this environment.IMPORTANCE Cystic fibrosis patients often suffer from chronic respiratory infections caused by several microorganisms. Among them are the Burkholderia cepacia complex bacteria which cause progressive deterioration of lung function and, in some patients, might develop into fatal necrotizing pneumoniae with bacteremia, known as "cepacia syndrome". Burkholderia pathogenesis is multifactorial since they express several virulence factors, form biofilms, and are highly resistant to antimicrobial compounds, making their eradication from the CF patients' airways very difficult. As Burkholderia is commonly found in the CF lungs in the form of cell aggregates and biofilms, the need to investigate the mechanisms of cellular aggregation is obvious. In this study we demonstrate the importance of a d-lactate dehydrogenase and a regulator, in regulating carbon overflow, cellular aggregates and surface-attached biofilm formation.     Hypercalcitoninemia is not Pathognomonic of Medullary Thyroid Carcinoma

    Hypercalcitoninemia has frequently been reported as a marker for medullary thyroid carcinoma. Currently, calcitonin measurements are mostly useful in the evaluation of tumor size and progression, and as an index of biochemical improvement of medullary thyroid carcinomas. Although measurement of calcitonin is a highly sensitive method for the detection of medullary thyroid carcinoma, it presents a low specificity for this tumor. Several physiologic and pathologic conditions other than medullary thyroid carcinoma have been associated with increased levels of calcitonin. Several cases of thyroid nodules associated with increased values of calcitonin are not medullary thyroid carcinomas, but rather are related to other conditions, such as hypercalcemias, hypergastrinemias, neuroendocrine tumors, renal insufficiency, papillary and follicular thyroid carcinomas, and goiter. Furthermore, prolonged treatment with omeprazole (> 2–4 months), beta-blockers, glucocorticoids and potential secretagogues, have been associated with hypercalcitoninemia. An association between calcitonin levels and chronic auto-immune thyroiditis remains controversial. Patients with calcitonin levels >100 pg/mL have a high risk for medullary thyroid carcinoma (~90%–100%), whereas patients with values from 10 to 100 pg/mL (normal values: <8.5 pg/mL for men, < 5.0 pg/mL for women; immunochemiluminometric assay) have a <25% risk for medullary thyroid carcinoma

    Características clínicas e epidemiológicas do câncer da boca em hospital-escola de 1994 a 2002: aumento da incidência em mulheres, predomínio de doença local avançada e baixa incidência de metástases regionais

    PURPOSE: Description of clinical and epidemiological characteristics of patients who underwent surgery for oral cancer in a Medical School Teaching Hospital, and determination of differences with respect to other institutions and/or periods of time. METHOD: The charts of patients undergoing surgery for oral cancer from 1994 to 2002 were reviewed. Data were collected in a spreadsheet in order to analyze clinical and epidemiological features.. RESULTS: A total of 374 patients having undergone 406 operations was identified. Their ages varied from 14 to 94 years (mean = 57.4 years), with 255 men (68.2%), and 295 out 366 Caucasian (80.6%). A majority had tumors of the tongue and/or floor of mouth (55.6%), while 20.3% had lip cancer. Squamous cell carcinoma was found in 90.3%, and glandular carcinoma in 4%. T4 tumors in 39.6%, Tis or T1 lesions in 15.2% of all patients. Nearly 62% had no regional metastases, and the relative incidence in young patients (40 years or younger) reached 8.6%. CONCLUSION: In spite of the predominance of locally advanced tumors, a majority of patients had no neck metastases. The 31.8% incidence in females indicates an increasing incidence of oral cavity cancer among women when compared to previous periods at the same institution.OBJETIVO: Caracterizar epidemiológica e clinicamente a população atendida por câncer de boca em hospital-escola de atenção terciária e quaternária e verificar variações em relação a outras casuísticas e períodos. MÉTODO: Procedeu-se à revisão dos prontuários dos operados por câncer oral de 1994 a 2002. As características clínicas e epidemiológicas foram colocadas em planilhas e analisadas. RESULTADOS: Foram identificados 374 portadores de câncer da boca que foram submetidos a 406 operações. A idade dos doentes variou de 14 a 94 anos (média = 57,4 anos). Duzentos e cinqüenta e cinco eram do gênero masculino (68,2%). Duzentos e noventa e cinco entre 366 doentes eram caucasianos (80,6%). A maioria era portadora de tumores da língua e/ou do soalho da boca (194 entre 349 doentes; 55,6%), e 71 (20,3%) tinham tumores dos lábios. O carcinoma epidermóide foi encontrado em 90,3% e carcinomas glandulares em 4%. Os tumores T4 estavam presentes em 39,6% dos doentes; 15,2% tinham lesões Tis ou T1. Quase 62% dos pacientes não tinham metástases cervicais. A incidência relativa em jovens (com até 40 anos de idade) atingiu 8,6%. CONCLUSÃO: Apesar dos achados freqüentemente referidos na literatura terem se confirmado (como o predomínio de lesões avançadas localmente), a maioria dos doentes não apresentava metástases cervicais. Acima do observado em outras casuísticas, os 31,8% de mulheres na presente casuística indica a preocupante e crescente incidência no gênero feminino

    Non-pharmacological treatments in the stimulation of osteogenesis

    Mechanical loads cause bone deformation leading to bone resorption and an increase in local bone formation. However, the stimulus for bone formation depends on the amount and frequency of bone deformation. High calcium intake is required to increase bone formation. There are also non-pharmacological treatments, such as electrical stimulation, low-intensity ultrasound, and laser, which revealed to promote osteogenesis. The article intends to evaluate non-pharmacological, alternative methods which assist in the increase of bone formation or in the osteogenic stimulus. A literature review was carried out for the period between 1982 to 2001.A carga mecânica provoca microdeformações ósseas, que estimulam células osteoblásticas e, conseqüentemente, promovem adaptações nos ossos, muitas vezes relacionadas com uma menor reabsorção óssea e um aumento na formação óssea local. No entanto, o estímulo para formação óssea depende do número e freqüência das deformações aplicadas no osso. Concomitantemente aos estímulos ósseos providos pelas cargas mecânicas, é importante uma dieta rica em cálcio, a qual também beneficia o aumento da massa óssea. Entretanto, existem também outros métodos não farmacológicos, como o ultra-som pulsado de baixa intensidade, a estimulação elétrica e o laser, que já mostraram efeitos positivos na promoção da osteogênese. Assim, realizou-se um levantamento na literatura, no período de 1982 a 2001, sobre esses métodos não farmacológicos com o objetivo de avaliar tais métodos alternativos, que ajudam no aumento da massa óssea ou no estímulo à osteogênese.64765

    HPLC-DAD analysis, antinociceptive and anti-inflammatory properties of the ethanolic extract of Hyptis umbrosa in mice

    Hyptis umbrosa (syn. Mesosphaerum sidifolium) (Lamiaceae Family) has been used to treat several conditions such as gastrointestinal disorders, skin infections, nasal congestion, fever and cramps. The objective of this study was to evaluate the chemical composition, analgesic and anti-inflammatory profiles of ethanol extract from leaves of Hyptis umbrosa (EEB). HPLC-DAD was used to determine the fingerprint chromatogram of the extract. Male Swiss mice were orally pretreated with EEB (100, 200 or 400 mg/kg; 60 min before initiating algesic stimulation) and antinociceptive activity was assessed using the acetic acid-induced writhing model, formalin test and hyperalgesia induced by glutamate or capsaicin. Also, peritonitis was induced by the intrathoracic injection of carrageenan to quantify the total number of leukocytes. The presence of phenolic compounds in the extract was confirmed using HPLC-DAD. The treatment with EEB, at all doses, produced a significant analgesic effect against acetic acid-induced antinociceptive activity. In the formalin test, only the 400-mg/kg-dose of EEB had a significant effect in the first phase. However, all doses tested were able to reverse nociception in the second phase. The effect of all doses of EEB also showed a significant antinociceptive effect in the glutamate and capsaicin tests and inhibited the carrageenan-induced leukocyte migration to the peritoneal cavity. The present study suggests that the EEB possesses peripheral analgesic action and showed potential in reducing the spreading of the inflammatory processes.
