    Humor creation and appreciation as an indicator of intercultural communication effectiveness : toward a theoretical model

    A review of the theoretical and empirical literature from the field of humor and from that of intercultural communication showed that no investigations dealt with the possibility that a sense of humor might serve as an indicator of intercultural communication effectiveness. This study was conducted in order to assess that possibility. This paper has set out to answer the following questions: What cognitive processes are common to a sense of humor and to intercultural communication effectiveness? What does one\u27s ability to create and appreciate humor say about that individual\u27s potential for intercultural communication effectiveness? Through the review of the literature, specific connections were suggested as theoretical connecting points or parallels. The processes suggested as common to humor and intercultural communication included the following: the ability to note difference; the ability to note and appreciate incongruity; the ability to process information both analytically and synthetically; the ability to shift frame of reference; the ability to perceive, communicate and maintain multiple perspectives; a tolerance for ambiguity; the possession of an internal locus of control; and the ability to act and react appropriately to others and to context. In an attempt to develop a theoretical link between these processes considered crucial to a sense of humor and to intercultural communication effectiveness, cognitive complexity theory was examined. Through this examination it was found that the cognitively complex individual possessed qualities and abilities consistent with those which had been found to be typical of people with a sense of humor and with people considered to be effective intercultural communicators. This suggested that cognitive complexity could serve as the theoretical tool by which a sense of humor may be seen as an indicator of intercultural communication effectiveness

    Two attentive strategies reducing subjective distortions in serial duration perception

    Humans tend to perceptually distort (dilate/shrink) the duration of brief stimuli presented in a sequence when discriminating the duration of a second stimulus (Comparison) from the duration of a first stimulus (Standard). This type of distortion, termed “Time order error” (TOE), is an important window into the determinants of subjective perception. We hypothesized that stimulus durations would be optimally processed, suppressing subjective distortions in serial perception, if the events to be compared fell within the boundaries of rhythmic attentive sampling (4–8 Hz, theta band). We used a two-interval forced choice (2IFC) experimental design, and in three separate experiments tested different Standard durations: 120-ms, corresponding to an 8.33 Hz rhythmic attentive window; 160 ms, corresponding to a 6.25 Hz window; and 200 ms, for a 5 Hz window. We found that TOE, as measured by the Constant Error metric, is sizeable for a 120-ms Standard, is reduced for a 160-ms Standard, and statistically disappears for 200-ms Standard events, confirming our hypothesis. For 120- and 160-ms Standard events, to reduce TOEs it was necessary to increase the interval between the Standard and the Comparison event from sub-second (400, 800 ms) to supra-second (1600, 2000 ms) lags, suggesting that the orienting of attention in time waiting for the Comparison event to onset may work as a back-up strategy to optimize its encoding. Our results highlight the flexible use of two different attentive strategies to optimize subjective time perception

    Neural entrainment via perceptual inferences

    Entrainment depends on sequential neural phase reset by regular stimulus onset, a temporal parameter. Entraining to sequences of identical stimuli also entails stimulus feature predictability, but this component is not readily separable from temporal regularity. To test if spectral regularities concur with temporal regularities in determining the strength of auditory entrainment, we devised sound sequences that varied in conditional perceptual inferences based on deviant sound repetition probability: strong inference (100% repetition probability: If a deviant appears, then it will repeat), weak inference (75% repetition probability) and no inference (50%: A deviant may or may not repeat with equal probability). We recorded EEG data from 15 young human participants pre-attentively listening to the experimental sound sequences delivered either isochronously or anisochronously (±20% jitter), at both delta (1.67 Hz) and theta (6.67 Hz) stimulation rates. Strong perceptual inferences significantly enhanced entrainment at either stimulation rate and determined positive correlations between precision in phase distribution at the onset of deviant trials and entrained power. We conclude that both spectral predictability and temporal regularity govern entrainment via neural phase control

    A perceptual glitch in serial perception generates temporal distortions

    Precisely estimating event timing is essential for survival, yet temporal distortions are ubiquitous in our daily sensory experience. Here, we tested whether the relative position, duration, and distance in time of two sequentially-organized events—standard S, with constant duration, and comparison C, with duration varying trial-by-trial—are causal factors in generating temporal distortions. We found that temporal distortions emerge when the first event is shorter than the second event. Importantly, a significant interaction suggests that a longer inter-stimulus interval (ISI) helps to counteract such serial distortion effect only when the constant S is in the first position, but not if the unpredictable C is in the first position. These results imply the existence of a perceptual bias in perceiving ordered event durations, mechanistically contributing to distortion in time perception. We simulated our behavioral results with a Bayesian model and replicated the finding that participants disproportionately expand first-position dynamic (unpredictable) short events. Our results clarify the mechanisms generating time distortions by identifying a hitherto unknown duration-dependent encoding inefficiency in human serial temporal perception, something akin to a strong prior that can be overridden for highly predictable sensory events but unfolds for unpredictable ones

    Indications for transcatheter valve implantation have been extended to treat native valve aortic regurgitation in case of disproportionate risk for open surgery. Transcatheter aortic valves are also an attractive alternative in patients who refuse blood transfusions. We report the successful off-label implantation of a self-expandable transcatheter valve in a Jehovah's Witness with prior replacement of the ascending aorta for Type A dissection, residual severe aortic regurgitation and refractory heart failure

    Isolation and pathogenicity of Xylella fastidiosa associated to the olive quick decline syndrome in southern Italy

    In autumn 2013, the presence of Xylella fastidiosa, a xylem-limited Gram-negative bacterium, was detected in olive stands of an area of the Ionian coast of the Salento peninsula (Apulia, southern Italy), that were severely affected by a disease denoted olive quick decline syndrome (OQDS). Studies were carried out for determining the involvement of this bacterium in the genesis of OQDS and of the leaf scorching shown by a number of naturally infected plants other than olive. Isolation in axenic culture was attempted and assays were carried out for determining its pathogenicity to olive, oleander and myrtle-leaf milkwort. The bacterium was readily detected by quantitative polymerase chain reaction (qPCR) in all diseased olive trees sampled in different and geographically separated infection foci, and culturing of 51 isolates, each from a distinct OQDS focus, was accomplished. Needle-inoculation experiments under different environmental conditions proved that the Salentinian isolate De Donno belonging to the subspecies pauca is able to multiply and systemically invade artificially inoculated hosts, reproducing symptoms observed in the field. Bacterial colonization occurred in prick-inoculated olives of all tested cultivars. However, the severity of and timing of symptoms appearance differed with the cultivar, confirming their differential reaction

    Prosody abilities in a large sample of affective and non-affective first episode psychosis patients

    Objective: Prosody comprehension deficits have been reported in major psychoses. It is still not clear whether these deficits occur at early psychosis stages. The aims of our study were to investigate a) linguistic and emotional prosody comprehension abilities in First Episode Psychosis (FEP) patients compared to healthy controls (HC); b) performance differences between non-affective (FEP-NA) and affective (FEP-A) patients, and c) association between symptoms severity and prosodic features. Methods: A total of 208 FEP (156 FEP-NA and 52 FEP-A) patients and 77 HC were enrolled and assessed with the Italian version of the “Protocole Montréal d'Evaluation de la Communication” to evaluate linguistic and emotional prosody comprehension. Clinical variables were assessed with a comprehensive set of standardized measures. Results: FEP patients displayed significant linguistic and emotional prosody deficits compared to HC, with FEP-NA showing greater impairment than FEP-A. Also, significant correlations between symptom severity and prosodic features in FEP patients were found. Conclusions: Our results suggest that prosodic impairments occur at the onset of psychosis being more prominent in FEP-NA and in those with severe psychopathology. These findings further support the hypothesis that aprosodia is a core feature of psychosis

    Screening of olive germplasm for resistance to Xylella fastidiosa ST53: the state of the art

    While different sources of natural resistance to X. fastidiosa have been described in grapevines and citrus, lack of solid information exists on possible sources of resistance/tolerance in the cultivars that characterize the wide olive germplasm. Preliminary field observations and laboratory analyses of a few cultivars, have shown that differential responses to X. fastidiosa infections exist. To confirm these preliminary findings, a large panel of olive cultivars is being specifically investigated. Currently, the screening procedure relies on field observations looking for symptomless subjects (trees of known cultivars/volunteer seedlings), mechanical inoculations, qualitative and quantitative diagnostic assays (ELISA & qPCR) and, in selected cases, comparative transcriptomic profiling. Field experiments include the planting of the target cultivars/selections in an infected area under high inoculum pressure. All the plots are located in the Apulia Region (Italy) in the demarcated infected area, surrounded by X. fastidiosa heavily affected olive groves. A first experimental plot was established in April 2015 with 10 different cultivars, which was extended in 2016 to 49 cultivars, and will be further enlarged in 2017 with the addition of 40 new accessions. Other plots, comprising newly planted or grafted cultivars (over 260 cvs) have been also established, bringing to over 300 the total number of accessions under evaluation. Cvs Leccino and FS-17®, both expressing interesting traits of resistance, have already been identified

    The Soluble Recombinant Neisseria meningitidis Adhesin NadAΔ351–405 Stimulates Human Monocytes by Binding to Extracellular Hsp90

    The adhesin NadA favors cell adhesion/invasion by hypervirulent Neisseria meningitidis B (MenB). Its recombinant form NadAΔ351–405, devoid of the outer membrane domain, is an immunogenic candidate for an anti-MenB vaccine able to stimulate monocytes, macrophages and dendritic cells. In this study we investigated the molecular mechanism of NadAΔ351–405 cellular effects in monocytes. We show that NadAΔ351–405 (against which we obtained polyclonal antibodies in rabbits), binds to hsp90, but not to other extracellular homologous heat shock proteins grp94 and hsp70, in vitro and on the surface of monocytes, in a temperature dependent way. Pre-incubation of monocytes with the MenB soluble adhesin interfered with the binding of anti-hsp90 and anti-hsp70 antibodies to hsp90 and hsp70 at 37°C, a condition in which specific cell-binding occurs, but not at 0°C, a condition in which specific cell-binding is very diminished. Conversely, pre-incubation of monocytes with anti-hsp90 and anti-hsp70 antibodies did not affected NadAΔ351–405 cell binding in any temperature condition, indicating that it associates to another receptor on their plasma membrane and then laterally diffuses to encounter hsp90. Consistently, polymixin B interfered with NadAΔ351–405 /hsp90 association, abrogated the decrease of anti-hsp90 antibodies binding to the cell surface due to NadAΔ351–405 and inhibited adhesin-induced cytokine/chemokine secretion without affecting monocyte-adhesin binding. Co-stimulation of monocytes with anti-hsp90 antibodies and NadAΔ351–405 determined a stronger but polymixin B insensitive cell activation. This indicated that the formation of a recombinant NadA/hsp90/hsp70 complex, although essential for full monocyte stimulation, can be replaced by anti-hsp90 antibody/hsp90 binding. Finally, the activation of monocytes by NadAΔ351–405 alone or in the presence of anti-hsp90 antibodies were both inhibited by neutralizing anti-TLR4 antibodies, but not by anti-TLR2 antibodies. We propose that hsp90-dependent recruitment into an hsp90/hsp70/TLR4 transducing signal complex is necessary for the immune-stimulating activity of NadAΔ351–405 anti-MenB vaccine candidate

    Targeted delivery of photosensitizers: efficacy and selectivity issues revealed by multifunctional ORMOSIL nanovectors in cellular systems

    PEGylated and non-PEGylated ORMOSIL nanoparticles prepared by microemulsion condensation of vinyltriethoxy-silane (VTES) were investigated in detail for their micro-structure and ability to deliver photoactive agents. With respect to pure silica nanoparticles, organic modification substantially changes the microstructure and the surface properties. This in turn leads to a modulation of both the photophysical properties of embedded photosensitizers and the interaction of the nanoparticles with biological entities such as serum proteins. The flexibility of the synthetic procedure allows the rapid preparation and screening of multifunctional nanosystems for photodynamic therapy (PDT). Selective targeting of model cancer cells was tested by using folate, an integrin specific RGD peptide and anti-EGFR antibodies. Data suggest the interference of the stealth-conferring layer (PEG) with small targeting agents, but not with bulky antibodies. Moreover, we showed that selective photokilling of tumour cells may be limited even in the case of efficient targeting because of intrinsic transport limitations of active cellular uptake mechanisms or suboptimum localization
