    Automatic Chemical Processing Systems for Extraterrestrial Biochemical Investigations

    Implementing the concept of planetary exploration is a formidable task. Laboratory procedures and techniques that are performed routinely by scientists and technicians must be performed remotely, automatically, and reliably for long periods of time without direct human intervention. The subtle and sophisticated reasoning, planning, and execution that lies behind well-conducted research in terrestrial laboratories must be incorporated into the careful original design and mechanization of extraterrestrial experiments. Mechanisms capable of automation for use in batch chemical processing of surface samples for scientific payloads have been devised and tested. Elemental processing operations common to a wide variety of possible life detection and biochemical characterization experiments were identified. It was determined that one device could perform a multiplicity of analyses if it could perform a limited number of elemental processing operations. Mechanisms were then designed for performing such elemental processing operations, and a breadboard processing device was fabricated incorporating the various processing operations, some completely automated (such as reagent metering) and others manually operated. The tremendous flexibility of the device to perform a wide variety of chemical procedures is evident. Tests of the breadboard in performing aqueous and organic extractions of soil samples indicate that completely automated, flight worthy chemical processing systems possessing great experimental flexibility are feasible and reliable

    International audienceMarket-gardening cropping systems in protected cultivation are very sensitive to soil-borne pests and diseases. Their productivity used to rely on pesticides, but alternative systems have now to be found for environmental, societal and health reasons. Many cultural techniques are known to provide some control of soilborne diseases, but are only partially efficient. The aim of the project is to design alternative systems with professionals, by improving the efficiency of the present techniques and/or imagining more innovative systems. The research project takes place in two steps. The first one consists in building a tool to assess the resistance or resilience of a given cropping system to soil-borne pests; the second one consists in using the tool with professionals in order to build alternative cropping and farming systems in cooperation. The model built for evaluation is a qualitative multicriteria tool. As scientific knowledge is not available for each technique or combination of techniques, empirical knowledge collected from growers and technical advisers is used to fill the gaps. The model is already built for root-knot nematodes and under construction for the other fungi. The second step will consist in using the tool with technical advisers and growers to redesign cropping systems and select the promising ones that should be put into trial in R&D stations. Co-building farming systems with stakeholders appears as an absolute necessity, to imagine solutions both efficient and acceptable for growers. The presentation will enable to discuss how combining expert and scientific knowledge may not only fill the knowledge gap, but also enable to build innovative solutions thanks to the diversity of experts' standpoints

    Young adults with a history of out-of-home care report poorer mental health and life satisfaction compared to non-care-experienced peers. Social support is a known protective factor for mental health. There is limited evidence, however, on the relationship between sources (e.g., family members) and types (e.g., information) of social support and mental health symptoms and life satisfaction in this population. Reporting cross-sectional survey data from 215 young adults aged 18–22 years with a history of out-of-home care, the current study conducted descriptive, bivariate, and linear regression analysis to examine the different sources and types of support young adults receive and their relation to mental health symptoms and life satisfaction. Participants had high levels of support from family members, friends, and other adults. Most participants had informational support, but less than half had consistent material support. Regression analyses demonstrated that having enough informational and material support were associated with fewer mental health symptoms. Having family support and material support were associated with greater life satisfaction. Further longitudinal research is needed to understand the trajectory between social supports and mental health functioning and life satisfactio

    Public policies clearly aim at an ecologisation of agriculture. However, few concrete goals or incentives have been established to mobilize the experimental research. Applied to 13-year results of four experimental stations of the South-East of France, the Direct-Indirect-System multi-level analysis grid showed a diversification of their experimental strategies. These three levels of analysis, combined with the identification of different types of agroecosystems, appears as promising for instigating assessment and determining agroecological innovation priorities for an experimental station, a territory or a food chain

    Use of a cAMP BRET Sensor to Characterize a Novel Regulation of cAMP by the Sphingosine 1-Phosphate/G13 Pathway

    Regulation of intracellular cyclic adenosine 3',5'-monophosphate (cAMP) is integral in mediating cell growth, cell differentiation, and immune responses in hematopoietic cells. To facilitate studies of cAMP regulation we developed a BRET (bioluminescence resonance energy transfer) sensor for cAMP, CAMYEL (cAMP sensor using YFP-Epac-RLuc), which can quantitatively and rapidly monitor intracellular concentrations of cAMP in vivo. This sensor was used to characterize three distinct pathways for modulation of cAMP synthesis stimulated by presumed Gs-dependent receptors for isoproterenol and prostaglandin E2. Whereas two ligands, uridine 5'-diphosphate and complement C5a, appear to use known mechanisms for augmentation of cAMP via Gq/calcium and Gi, the action of sphingosine 1-phosphate (S1P) is novel. In these cells, S1P, a biologically active lysophospholipid, greatly enhances increases in intracellular cAMP triggered by the ligands for Gs-coupled receptors while having only a minimal effect by itself. The enhancement of cAMP by S1P is resistant to pertussis toxin and independent of intracellular calcium. Studies with RNAi and chemical perturbations demonstrate that the effect of S1P is mediated by the S1P2 receptor and the heterotrimeric G13 protein. Thus in these macrophage cells, all four major classes of G proteins can regulate intracellular cAMP

    Review Essay: Biopolitics 2.0 – Reclaiming the Power of Life in the Anthropocene

    Biopolitics was once the preserve of critical Foucauldian-influenced theorists. This is no longer the case. The rise of new materialisms, networked and object-oriented ontologies and demands for the ‘renaturalization of politics’ (Grosz, 2011; Sharp, 2011) have turned the political into the biopolitical in much of radical contemporary political theory. Here, the biopolitical is often in tension with the Foucauldian discourses of the last few decades, affirming alternative possibilities rather than merely critiquing regimes of power (Malabou, 2008; Kirby, 2011; Taussig, 2018). One factor in this shift has been the impact of anthropogenic climate change and global warming on the contemporary political imagination, increasingly refracted through conceptualizations of the Anthropocene (Tsing, 2015; Cohen et al, 2016; Chandler, 2018). A second factor has been that the life sciences no longer appear to support understandings of deterministic differences but rather to cast evolution in sympoietic and entangled ways, making biology appear as no longer essentialising but as increasingly importable into politics, in ways which disrupt modernist or liberal conceptions of the culture/nature divide (Haraway, 2016). Two important but very different engagements, which reflect these affirmative readings of biopolitics, are reviewed here, from the fields of postcolonial literature and gender and race in international politics
