    Assessing a hierarchical sustainable solid waste management structure with qualitative information:policy and regulations drive social impacts and stakeholder participation

    This study contributes to building a valid hierarchical sustainable solid waste management (SSWM) attribute set with qualitative information, which is a complicated and ambiguous problem with uncertainties. Few studies have provided qualitative information and have addressed the interrelationships and interdependencies among the hierarchical SSWM attributes. SSWM has recently become a difficult problem to solve due to urbanization, inequality, and economic growth. This study aims to propose an SSWM attribute set and identify a causal model through linguistic preferences by using a fuzzy decision-making trial and evaluation laboratory approach to simultaneously handle the uncertainty and the interrelationships. The analytic network process is used to develop the hierarchical structure to weigh the aspects and criteria. Qualitative information is transformed into crisp and comparable values to examine the causal relationships between attributes and confirm the consistency between the theoretical structure and industry phenomena. The results indicate that policy and regulations, stakeholder participation, and social impacts play essential roles in these causal interrelationships. Political leadership in SSWM is required to drive stakeholder participation and achieve social impacts. Population growth and migration, institutional settings, waste recycling and energy recovery, households, and private contractors are the main aspects involved in improving SSWM in Vietnam

    A proposed conceptual model of determinants of supply chain performance in the Malaysian electronic manufacturing services (EMS) industry

    Electronic manufacturing services (EMS) provide electronics manufacturing services for other companies. This industry is one of the industries that require supply chain management (SCM) to optimize the operations. EMS focuses on printed circuit board fabrication, electronic design, assembly and testing. In this article, the authors propose a conceptual model by linking the relationships with customer and supplier, information and communication technology, material flows management, corporate culture, performance measurement; and SCM performance in a single study. The findings of this article would provide important implications for the management in the manufacturing companies to understand determinants that contribute to the SCM success. More importantly, based of the obtained results, these companies can enhance the SCM performance by improving the current practices/strategies through focusing on the determinants that significantly influence SCM performance

    Critical success factors of supply chain management: An empirical study on a manufacturing company in Malaysia

    The aim of this study is to shed the light on the critical success factors that lead to high supply chain performance in a manufacturing company. The critical success factors consist of relationships with customer and supplier, information communication and technology (ICT), material flows management, corporate culture and performance measurement. Questionnaire was the main instrument for the study and it was distributed to 84 staffs from departments of purchasing, planning,logistics and operation. Hypotheses were testing by using Statistical Package for Social Science (SPSS) of Windows software. Data analysis was conducted by employing descriptive analysis (mean and standard deviation), reliability analysis, Pearson correlation analysis and multiple regression. The findings show that there are relationships between relationships with customer and supplier, ICT, material flows management, performance measurement and supply chain management (SCM)performance, except for corporate culture. Forming a good customer and supplier relationship is the main predictor of SCM performance, followed by performance measurement, material flows management and ICT. It is recommended that future study to determine additional success factors that are pertinent to firms’ current SCM strategies and directions, competitive advantages and missions. Further study is recommended also to examine a wider scope that includes more geographical data coverage, other nature of businesses and research instruments

    A novel class of microRNA-recognition elements that function only within open reading frames.

    MicroRNAs (miRNAs) are well known to target 3' untranslated regions (3' UTRs) in mRNAs, thereby silencing gene expression at the post-transcriptional level. Multiple reports have also indicated the ability of miRNAs to target protein-coding sequences (CDS); however, miRNAs have been generally believed to function through similar mechanisms regardless of the locations of their sites of action. Here, we report a class of miRNA-recognition elements (MREs) that function exclusively in CDS regions. Through functional and mechanistic characterization of these 'unusual' MREs, we demonstrate that CDS-targeted miRNAs require extensive base-pairing at the 3' side rather than the 5' seed; cause gene silencing in an Argonaute-dependent but GW182-independent manner; and repress translation by inducing transient ribosome stalling instead of mRNA destabilization. These findings reveal distinct mechanisms and functional consequences of miRNAs that target CDS versus the 3' UTR and suggest that CDS-targeted miRNAs may use a translational quality-control-related mechanism to regulate translation in mammalian cells

    ATOMIUM: A high-resolution view on the highly asymmetric wind of the AGB star pi(1)Gruis: I. First detection of a new companion and its effect on the inner wind

    The nebular circumstellar environments of cool evolved stars are known to harbour a rich morphological complexity of gaseous structures on different length scales. A large part of these density structures are thought to be brought about by the interaction of the stellar wind with a close companion. The S-type asymptotic giant branch (AGB) star π1Gruis, which has a known companion at ∼440 au and is thought to harbour a second, closer-by (< 10 au) companion, was observed with the Atacama Large Millimeter/submillimeter Array as part of the ATOMIUM Large programme. In this work, the brightest CO, SiO, and HCN molecular line transitions are analysed. The continuum map shows two maxima, separated by 0.04″ (6 au). The CO data unambiguously reveal that π1Gru’s circumstellar environment harbours an inclined, radially outflowing, equatorial density enhancement. It contains a spiral structure at an angle of ∼38 ± 3° with the line-of-sight. The HCN emission in the inner wind reveals a clockwise spiral, with a dynamical crossing time of the spiral arms consistent with a companion at a distance of 0.04″ from the AGB star, which is in agreement with the position of the secondary continuum peak. The inner wind dynamics imply a large acceleration region, consistent with a beta-law power of ∼6. The CO emission suggests that the spiral is approximately Archimedean within 5″, beyond which this trend breaks down as the succession of the spiral arms becomes less periodic. The SiO emission at scales smaller than 0.5″ exhibits signatures of gas in rotation, which is found to fit the expected behaviour of gas in the wind-companion interaction zone. An investigation of SiO maser emission reveals what could be a stream of gas accelerating from the surface of the AGB star to the companion. Using these dynamics, we have tentatively derived an upper limit on the companion mass to be ∼1.1 M⊙

    Targeting Bone Alleviates Osteoarthritis in Osteopenic Mice and Modulates Cartilage Catabolism

    Subchondral bone modifications occur early in the development of osteoarthritis (OA). The level of bone resorption might impact cartilage remodeling. We therefore assessed the in vivo and in vitro effects of targeting bone resorption in OA and cartilage metabolism.OA was induced by meniscectomy (MNX) in ovariectomized osteopenic mice (OP) treated with estradiol (E2), pamidronate (PAM), or phosphate buffered saline (PBS) for 6 weeks. We assessed the subchondral bone and cartilage structure and the expression of cartilage matrix proteases. To assess the involvement of bone soluble factors in cartilage metabolism, supernatant of human bone explants pre-treated with E2 or PAM were transferred to cartilage explants to assess proteoglycan release and aggrecan cleavage. OPG/RANKL mRNA expression was assessed in bone explants by real-time quantitative PCR. The role of osteoprotegerin (OPG) in the bone-cartilage crosstalk was tested using an OPG neutralizing antibody.Bone mineral density of OP mice and osteoclast number were restored by E2 and PAM (p<0.05). In OP mice, E2 and PAM decreased ADAMTS-4 and -5 expression, while only PAM markedly reduced OA compared to PBS (2.0±0.63 vs 5.2±0.95; p<0.05). OPG/RANKL mRNA was increased in human bone explants treated with both drugs (2.2-3.7-fold). Moreover, supernatants from bone explants cultured with E2 or PAM reduced aggrecan cleavage and cartilage proteoglycan release (73±8.0% and 80±22% of control, respectively, p<0.05). This effect was reversed with osteoprotegerin blockade.The inhibition of bone resorption by pamidronate in osteopenic mice alleviates the histological OA score with a reduction in the expression of aggrecanases. Bone soluble factors, such as osteoprotegerin, impact the cartilage response to catabolic factors. This study further highlights the importance of subchondral bone in the regulation of joint cartilage damage in OA

    Identification of Clinically Relevant Protein Targets in Prostate Cancer with 2D-DIGE Coupled Mass Spectrometry and Systems Biology Network Platform

    Prostate cancer (PCa) is the most common type of cancer found in men and among the leading causes of cancer death in the western world. In the present study, we compared the individual protein expression patterns from histologically characterized PCa and the surrounding benign tissue obtained by manual micro dissection using highly sensitive two-dimensional differential gel electrophoresis (2D-DIGE) coupled with mass spectrometry. Proteomic data revealed 118 protein spots to be differentially expressed in cancer (n = 24) compared to benign (n = 21) prostate tissue. These spots were analysed by MALDI-TOF-MS/MS and 79 different proteins were identified. Using principal component analysis we could clearly separate tumor and normal tissue and two distinct tumor groups based on the protein expression pattern. By using a systems biology approach, we could map many of these proteins both into major pathways involved in PCa progression as well as into a group of potential diagnostic and/or prognostic markers. Due to complexity of the highly interconnected shortest pathway network, the functional sub networks revealed some of the potential candidate biomarker proteins for further validation. By using a systems biology approach, our study revealed novel proteins and molecular networks with altered expression in PCa. Further functional validation of individual proteins is ongoing and might provide new insights in PCa progression potentially leading to the design of novel diagnostic and therapeutic strategies

    Osteoprotegerin in Exosome-Like Vesicles from Human Cultured Tubular Cells and Urine

    Urinary exosomes have been proposed as potential diagnostic tools. TNF superfamily cytokines and receptors may be present in exosomes and are expressed by proximal tubular cells. We have now studied the expression of selected TNF superfamily proteins in exosome-like vesicles from cultured human proximal tubular cells and human urine and have identified additional proteins in these vesicles by LC-MS/MS proteomics. Human proximal tubular cells constitutively released exosome-like vesicles that did not contain the TNF superfamily cytokines TRAIL or TWEAK. However, exosome-like vesicles contained osteoprotegerin (OPG), a TNF receptor superfamily protein, as assessed by Western blot, ELISA or selected reaction monitoring by nLC-(QQQ)MS/MS. Twenty-one additional proteins were identified in tubular cell exosomelike vesicles, including one (vitamin D binding protein) that had not been previously reported in exosome-like vesicles. Twelve were extracellular matrix proteins, including the basement membrane proteins type IV collagen, nidogen-1, agrin and fibulin-1. Urine from chronic kidney disease patients contained a higher amount of exosomal protein and exosomal OPG than urine from healthy volunteers. Specifically OPG was increased in autosomal dominant polycystic kidney disease urinary exosome-like vesicles and expressed by cystic epithelium in vivo.     Adaptive Radial Basis Function Methods for Pricing Options Under Jump-Diffusion Models

    The aim of this paper is to show that option prices in jump-diffusion models can be computed using meshless methods based on radial basis function (RBF) interpolation instead of traditional mesh-based methods like finite differences or finite elements. The RBF technique is demonstrated by solving the partial integro-differential equation for American and European options on non-dividend-paying stocks in the Merton jump-diffusion model, using the inverse multiquadric radial basis function. The method can in principle be extended to Lévy-models. Moreover, an adaptive method is proposed to tackle the accuracy problem caused by a singularity in the initial condition so that the accuracy in option pricing in particular for small time to maturity can be improved
