    The Effect of Bacterial Endotoxin Upon the Morphology of the Tectorial Membrane and Stereocilia in the Guinea Pig Cochlea

    Endotoxin of E coli was microperfused into scala tympani or injected into the cerebrospinal fluid in anaesthetised pigmented guinea pigs. The effects of endotoxin on the cochlea were studied using electrophysiological techniques and scanning electron microscopy. We found a drop in the amplitude of the cochlear microphonics and compound action potentials 2 to 2.5 hours after injection. There were also changes in the morphology of stereocilia and the tectorial membrane. The stereocilia lost their rigidity and the tectorial membrane appeared swollen. These effects were less severe in animals which were pretreated with dexamethasone

    Learning to Fix Build Errors with Graph2Diff Neural Networks

    Professional software developers spend a significant amount of time fixing builds, but this has received little attention as a problem in automatic program repair. We present a new deep learning architecture, called Graph2Diff, for automatically localizing and fixing build errors. We represent source code, build configuration files, and compiler diagnostic messages as a graph, and then use a Graph Neural Network model to predict a diff. A diff specifies how to modify the code’s abstract syntax tree, represented in the neural network as a sequence of tokens and of pointers to code locations. Our network is an instance of a more general abstraction which we call Graph2Tocopo, which is potentially useful in any development tool for predicting source code changes. We evaluate the model on a dataset of over 500k real build errors and their resolutions from professional developers. Compared to the approach of DeepDelta [23], our approach tackles the harder task of predicting a more precise diff but still achieves over double the accuracy

    Bipotential Adult Liver Progenitors Are Derived from Chronically Injured Mature Hepatocytes

    SummaryAdult liver progenitor cells are biliary-like epithelial cells that emerge only under injury conditions in the periportal region of the liver. They exhibit phenotypes of both hepatocytes and bile ducts. However, their origin and their significance to injury repair remain unclear. Here, we used a chimeric lineage tracing system to demonstrate that hepatocytes contribute to the progenitor pool. RNA-sequencing, ultrastructural analysis, and in vitro progenitor assays revealed that hepatocyte-derived progenitors were distinct from their biliary-derived counterparts. In vivo lineage tracing and serial transplantation assays showed that hepatocyte-derived proliferative ducts retained a memory of their origin and differentiated back into hepatocytes upon cessation of injury. Similarly, human hepatocytes in chimeric mice also gave rise to biliary progenitors in vivo. We conclude that human and mouse hepatocytes can undergo reversible ductal metaplasia in response to injury, expand as ducts, and subsequently contribute to restoration of the hepatocyte mass

    A comparative analysis of samples of external memorials from burial grounds in Britain, Ireland and New England reveals a widespread pattern of change in monument style and content, and exponential growth in the number of permanent memorials from the 18th century onwards. Although manifested in regionally distinctive styles on which most academic attention has so far been directed, the expansion reflects global changes in social relationships and concepts of memory and the body. An archaeological perspective reveals the importance of external memorials in articulating these changing attitudes in a world of increasing material consumption

    The prognostic value of four interleukin-1 gene polymorphisms in caucasian women with breast cancer – a multicenter study

    <p>Abstract</p> <p>Background</p> <p>The proinflammatory cytokine interleukin-1 (IL-1) is known to play an important role in the carcinogenesis of breast cancer. Although IL-1 gene polymorphisms were reported to be associated with increased risk of breast cancer, their influence on survival of Caucasian breast cancer patients remains to be shown.</p> <p>Methods</p> <p>We studied the influence of four common gene polymorphisms (<it>IL1A </it>-889C/T, <it>IL1B </it>-511C/T, <it>IL1B </it>+3953E1/E2, and <it>IL1RN </it>long/2) of the IL-1 family on survival in 262 Caucasian patients with breast cancer by univariate and multivariate survival analysis. The combined effect of the four gene polymorphisms on overall survival was studied by haplotype analysis.</p> <p>Results</p> <p>In the present study 38 cases of cancer related death and a median time of follow-up (range) of 55.3 (0.4–175.8) months was observed. <it>IL1RN </it>2/2 (homozygous mutant) gene polymorphism was associated with shortened disease free and overall survival in a univariate (p = 0.001 and p = 0.01, respectively) and multivariate analysis (p = 0.002, Odds Ratio [95% Confidence Interval] = 3.6 [1.6–8.0] and p = 0.05, Odds Ratio = 3.0 [1.1–9.3], respectively). Presence of the homozygous mutant genotype of the <it>IL1A </it>-889 and <it>IL1B </it>+3953 gene polymorphism was associated with overall survival in the univariate (p = 0.004 and p = 0.002, respectively), but not in the multivariate analysis. No association was observed between all possible haplotype combinations and overall survival.</p> <p>Conclusion</p> <p>Carriage of the mutant alleles of <it>IL1RN </it>was independently associated with shortened disease free and overall survival rates in Caucasian patients with breast cancer.</p
