The article presents the results of the study of the emotional perception of clients conducted among Russian social workers who help children. The research is based on the concept of the role of emotional laborin professional activities. At the same time, we proceed from the understanding that the content and volume of emotional labordepends on the type of clients of social workers: its specificity when working with children will not be the same as when workingwith adults. The results of the research show that social workers who help children have difficultiesin regulating emotions when interacting with clients, and therefore in assessing children's actions and behavior. The results of the study showthat the effectiveness of emotional laborof social workers helping children depends on the level of knowledge of the age characteristics of children and the pedagogical mechanisms of communicating with them.We conclude that we should pay more attention to the role of emotional laborin everyday social work with childrenpublishersversionPeer reviewe

Health saving strategy as a factor of "society wellness"

The article deals with the problem of reducing the motor activity of a modern man, especially a young man, in connection with scientific and technological progress. The results of the presented study confirm these fears: inactive lifestyle is becoming the norm. The importance of promoting the health saving strategy as one of the factors of successful social and economic development of the society is justified.publishersversionPeer reviewe

This article is devoted to a technique of estimation of students’ training results of the higher education in the conditions of the competence-based approach implementation. The formation stages of an assessment of training results on the example of the discipline “Information science” according to an educational program 270800.62 “Construction” including: detection of the created competences’ list, determination of their structure; detection of planned results of training and indexes of their estimation; determination of the appropriate methods and means of estimation. Compliances between results of training and components of a subject matter for the purpose of criteria determination of the level estimation of the acquired competences are revealed. Such approach will allow to provide the accuracy of estimation of the received competences at the expense of increase in amount of the criteria corresponding to a certain level of proficiency, and also due to the implementation of the modern training techniques and new approaches to an assessment of training results taking into account the competence-based approach which will allow to provide the accuracy of the estimation procedure directed on personal achievements of students.

Magnet for the Needle in Haystack: “Crystal Structure First” Fragment Hits Unlock Active Chemical Matter Using Targeted Exploration of Vast Chemical Spaces

Fragment-based drug discovery (FBDD) has successfully led to approved therapeutics for challenging and “undruggable” targets. In the context of FBDD, we introduce a novel, multidisciplinary method to identify active molecules from purchasable chemical space. Starting from four small-molecule fragment complexes of protein kinase A (PKA), a template-based docking screen using Enamine’s multibillion REAL Space was performed. A total of 93 molecules out of 106 selected compounds were successfully synthesized. Forty compounds were active in at least one validation assay with the most active follow-up having a 13,500-fold gain in affinity. Crystal structures for six of the most promising binders were rapidly obtained, verifying the binding mode. The overall success rate for this novel fragment-to-hit approach was 40%, accomplished in only 9 weeks. The results challenge the established fragment prescreening paradigm since the standard industrial filters for fragment hit identification in a thermal shift assay would have missed the initial fragments

A Multi-Pronged Approach Targeting SARS-CoV-2 Proteins Using Ultra-Large Virtual Screening

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), previously known as 2019 novel coronavirus (2019-nCoV), has spread rapidly across the globe, creating an unparalleled global health burden and spurring a deepening economic crisis. As of July 7th, 2020, almost seven months into the outbreak, there are no approved vaccines and few treatments available. Developing drugs that target multiple points in the viral life cycle could serve as a strategy to tackle the current as well as future coronavirus pandemics. Here we leverage the power of our recently developed in silico screening platform, VirtualFlow, to identify inhibitors that target SARS-CoV-2. VirtualFlow is able to efficiently harness the power of computing clusters and cloud-based computing platforms to carry out ultra-large scale virtual screens. In this unprecedented structure-based multi-target virtual screening campaign, we have used VirtualFlow to screen an average of approximately 1 billion molecules against each of 40 different target sites on 17 different potential viral and host targets in the cloud. In addition to targeting the active sites of viral enzymes, we also target critical auxiliary sites such as functionally important protein-protein interaction interfaces. This multi-target approach not only increases the likelihood of finding a potent inhibitor, but could also help identify a collection of anti-coronavirus drugs that would retain efficacy in the face of viral mutation. Drugs belonging to different regimen classes could be combined to develop possible combination therapies, and top hits that bind at highly conserved sites would be potential candidates for further development as coronavirus drugs. Here, we present the top 200 in silico hits for each target site. While in-house experimental validation of some of these compounds is currently underway, we want to make this array of potential inhibitor candidates available to researchers worldwide in consideration of the pressing need for fast-tracked drug development.</p
