    Penilaian Indikator K3l Pada Penggunaan Tower Crane

    Tower crane merupakan alat berat yang sangat bermanfaat dan mampu menggantikan peran manusia karena fungsinya untuk mengangkut dan memindahkan material baik vertikal maupun horizontal. Salah satu aspek yang sangat penting dan perlu diperhatikan dalam menggunakan tower crane adalah Keselamatan dan Kesehatan Kerja dan Lingkungan (K3L). Banyak peraturan yang mengatur tentang penggunaan tower crane khususnya mengenai K3L. Model pengukuran tingkat keselamatan dalam penggunaan tower crane yang telah dihasilkan (Kwan dan Anggrawan, 2014) merupakan aplikasi yang sangat membantu dalam penggunaan tower crane. Namun, model pengukuran tersebut perlu disertai indikator karena tanpa indikator apabila digunakan, model ini akan menghasilkan nilai akhir yang bias karena sifatnya yang subjektif. Untuk itu penelitian ini bertujuan untuk memperoleh apa saja indikator Keselamatan dan Kesehatan dan Lingkungan (K3L) pada penggunaan tower crane. Data dari penelitian ini diperoleh dari pengisian kuesioner dari para responden yang akan diolah untuk mendapatkan bobot dari masing-masing indikator. Hasil dari penelitian ini adalah indikator penilaian Keselamatan dan Kesehatan Kerja dan Lingkungan pada penggunaan tower crane yang akan disertakan ke dalam model pengukuran. Dari penelitian ini diperoleh indikator penelitian yang memiliki pengaruh yang berbeda terhadap tingkat K3L pada penggunaan tower crane

    Antiplasmodial activity of chloroquine analogs against chloroquine-resistant parasites, docking studies and mechanisms of drug action

Belo Horizonte, MG, BrazilBackground: Given the threat of resistance of human malaria parasites, including to artemisinin derivatives, new agents are needed. Chloroquine (CQ) has been the most widely used anti-malarial, and new analogs (CQAns) presenting alkynes and side chain variations with high antiplasmodial activity were evaluated. Methods: Six diaminealkyne and diaminedialkyne CQAns were evaluated against CQ-resistant (CQ-R) (W2) and CQ-sensitive (CQ-S) (3D7) Plasmodium falciparum parasites in culture. Drug cytotoxicity to a human hepatoma cell line (HepG2) evaluated, allowed to calculate the drug selectivity index (SI), a ratio of drug toxicity to activity in vitro. The CQAns were re-evaluated against CQ-resistant and -sensitive P. berghei parasites in mice using the suppressive test. Docking studies with the CQAns and the human (HssLDH) or plasmodial lactate dehydrogenase (PfLDH) enzymes, and, a β-haematin formation assay were performed using a lipid as a catalyst to promote crystallization in vitro. Results: All tested CQAns were highly active against CQ-R P. falciparum parasites, exhibiting half-maximal inhibitory concentration (IC50) values below 1 μΜ. CQAn33 and CQAn37 had the highest SIs. Docking studies revealed the best conformation of CQAn33 inside the binding pocket of PfLDH; specificity between the residues involved in H-bonds of the PfLDH with CQAn37. CQAn33 and CQAn37 were also shown to be weak inhibitors of PfLDH. CQAn33 and CQAn37 inhibited β-haematin formation with either a similar or a 2-fold higher IC50 value, respectively, compared with CQ. CQAn37 was active in mice with P. berghei, reducing parasitaemia by 100%. CQAn33, -39 and -45 also inhibited CQ-resistant P. berghei parasites in mice, whereas high doses of CQ were inactive. Conclusions: The presence of an alkyne group and the size of the side chain affected anti-P. falciparum activityin vitro. Docking studies suggested a mechanism of action other than PfLDH inhibition. The β-haematin assay suggested the presence of an additional mechanism of action of CQAn33 and CQAn37. Tests with CQAn34, CQAn37, CQAn39 and CQAn45 confirmed previous results against P. berghei malaria in mice, and CQAn33, 39 and 45 were active against CQ-resistant parasites, but CQAn28 and CQAn34 were not. The result likely reflects structure-activity relationships related to the resistant phenotype

    Simulation and Training of Gynaecological Skills

    In order to offer our patients, the "state of the art" treatment in gynaecology, we need a structured teaching program for trainees concerning the gynaecological skills. In recent years, training and education in endoscopic surgery has been critically reviewed. Clinicians, both surgeons as gynaecologists who perform endoscopic surgery without proper training of the specific psychomotor skills, are at higher risk to increased patient morbidity and mortality. The traditional apprentice-tutor model is no longer valid for developing all skills necessary in gynaecological surgery, particularly in endoscopy. Endoscopic training should happen at both the theoretical and the practical skill level. The acquisition of the correct knowledge regarding general laparoscopy, hysteroscopy and standard level procedures is as important as learning the necessary psychomotor skills to successfully perform endoscopic manipulations. Training in the operating room can only start when it is proven that knowledge and skills are present. To learn and train total abdominal hysterectomy by laparotomy there are inexpensive simple models that can be used, which are easy to reproduce. The development, construction, cost, and utility of a low-cost and anatomically representative vaginal hysterectomy simulator also has been described.     Dual Targeting of PDGFRα and FGFR1 Displays Synergistic Efficacy in Malignant Rhabdoid Tumors

    © 2016 The Author(s) Subunits of the SWI/SNF chromatin remodeling complex are mutated in a significant proportion of human cancers. Malignant rhabdoid tumors (MRTs) are lethal pediatric cancers characterized by a deficiency in the SWI/SNF subunit SMARCB1. Here, we employ an integrated molecular profiling and chemical biology approach to demonstrate that the receptor tyrosine kinases (RTKs) PDGFRα and FGFR1 are coactivated in MRT cells and that dual blockade of these receptors has synergistic efficacy. Inhibitor combinations targeting both receptors and the dual inhibitor ponatinib suppress the AKT and ERK1/2 pathways leading to apoptosis. MRT cells that have acquired resistance to the PDGFRα inhibitor pazopanib are susceptible to FGFR inhibitors. We show that PDGFRα levels are regulated by SMARCB1 expression, and assessment of clinical specimens documents the expression of both PDGFRα and FGFR1 in rhabdoid tumor patients.     Mesophase evolution in heat-treated solid petroleum pitches

    A combination of analytical techniques was used to follow the mesophase evolution of petroleum pitches in the solid phase. The samples were characterized by matrix assisted laser desorption and time of flight analysis by mass spectrometry (MALDI-TOF-MS), high field 13C nuclear magnetic resonance (13C NMR), low field ¹H NMR, electron spin resonance (EPR), polarized optical microscopy and thermogravimetric analysis (TGA). MALDI-TOF-MS revealed an oligomeric distribution of molecular weights in the pitch: monomeric (m/z 200 to 400), dimeric (m/z 400 to 650), trimeric (m/z 650 to 950) and tetrameric (m/z 950 to 1600), an essential factor for the production of carbon fibers, among other advanced composite of carbon. Relaxation time (T1) studies permitted to observe two large areas: the aromatics and the mesophases. These results also showed that the heating time increases the mesophasic area. Also through direct and non-destructive 13C NMR measurements, we obtained the content of aromatic and aliphatic carbons, as well as the degree of protonation of these carbons. Optical microscopy with polarized light showed the interfaces between the anisotropic and isotropic regions as well as the process of coalescence of mesophase. The results obtained by the other techniques corroborate the MALDI-TOF-MS and NMR results.Uma combinação de técnicas analíticas foi utilizada para acompanhar a evolução da mesofase em pixes de petróleo em fase sólida. As amostras foram caracterizadas por dessorção a laser assistida por matriz, seguida de espectrometria de massas por tempo de vôo (MALDI-TOF-MS), ressonância magnética nuclear de 13C (13C NMR) de alto campo, ¹H NMR de baixo campo, ressonância eletrônica de spin (EPR), microscopia ótica polarizada e análise termogravimétrica (TGA). A análise por MALDI-TOF-MS revelou uma distribuição oligomérica de pesos moleculares no pixe: monomérica (m/z 200 a 400), dimérica (m/z 400 a 650), trimérica (m/z 650 a 950) e tetramérica (m/z 950 a 1600), fator essential para a produção de fibras de carbono, bem como outros compósitos de carbono. Estudos de tempo de relaxação (T1) permitiram a observação de duas grandes áreas: a dos aromáticos e a de mesofase. Estes resultados também mostraram que o tempo de aquecimento aumenta a área de mesofase. Através de medidas não destrutivas de 13C NMR, foram obtidos os conteúdos de carbonos aromáticos e alifáticos, e também o grau de protonação desses carbonos. A microscopia ótica com luz polarizada mostrou as interfaces entre as regiões anisotrópicas e isotrópicas, bem como o processo de coalescência da mesofase.     Super-resolution architecture of mammalian centriole distal appendages reveals distinct blade and matrix functional components

Distal appendages (DAPs) are nanoscale, pinwheel-like structures protruding from the distal end of the centriole that mediate membrane docking during ciliogenesis, marking the cilia base around the ciliary gate. Here we determine a super-resolved multiplex of 16 centriole-distal-end components. Surprisingly, rather than pinwheels, intact DAPs exhibit a cone-shaped architecture with components filling the space between each pinwheel blade, a new structural element we term the distal appendage matrix (DAM). Specifically, CEP83, CEP89, SCLT1, and CEP164 form the backbone of pinwheel blades, with CEP83 confined at the root and CEP164 extending to the tip near the membrane-docking site. By contrast, FBF1 marks the distal end of the DAM near the ciliary membrane. Strikingly, unlike CEP164, which is essential for ciliogenesis, FBF1 is required for ciliary gating of transmembrane proteins, revealing DAPs as an essential component of the ciliary gate.     Development of targeted therapy for ovarian cancer mediated by a plasmid expressing diphtheria toxin under the control of H19 regulatory sequences

    <p>Abstract</p> <p>Background</p> <p>Ovarian cancer ascites fluid (OCAF), contains malignant cells, is usually present in women with an advanced stage disease and currently has no effective therapy. Hence, we developed a new therapy strategy to target the expression of diphtheria toxin gene under the control of H19 regulatory sequences in ovarian tumor cells. H19 RNA is present at high levels in human cancer tissues (including ovarian cancer), while existing at a nearly undetectable level in the surrounding normal tissue.</p> <p>Methods</p> <p>H19 gene expression was tested in cells from OCAF by the in-situ hybridization technique (ISH) using an H19 RNA probe. The therapeutic potential of the toxin vector DTA-H19 was tested in ovarian carcinoma cell lines and in a heterotopic animal model for ovarian cancer.</p> <p>Results</p> <p>H19 RNA was detected in 90% of patients with OCAF as determined by ISH. Intratumoral injection of DTA-H19 into ectopically developed tumors caused 40% inhibition of tumor growth.</p> <p>Conclusion</p> <p>These observations may be the first step towards a major breakthrough in the treatment of human OCAF, while the effect in solid tumors required further investigation. It should enable us to identify likely non-responders in advance, and to treat patients who are resistant to all known therapies, thereby avoiding treatment failure.</p
