Enzymatic activity and immunoreactivity of Aca s 4, an alpha-amylase allergen from the storage mite Acarus siro

<p>Abstract</p> <p>Background</p> <p>Enzymatic allergens of storage mites that contaminate stored food products are poorly characterized. We describe biochemical and immunological properties of the native alpha-amylase allergen Aca s 4 from <it>Acarus siro</it>, a medically important storage mite.</p> <p>Results</p> <p><it>A. siro </it>produced a high level of alpha-amylase activity attributed to Aca s 4. This enzyme was purified and identified by protein sequencing and LC-MS/MS analysis. Aca s 4 showed a distinct inhibition pattern and an unusual alpha-amylolytic activity with low sensitivity to activation by chloride ions. Homology modeling of Aca s 4 revealed a structural change in the chloride-binding site that may account for this activation pattern. Aca s 4 was recognized by IgE from house dust mite-sensitive patients, and potential epitopes for cross-reactivity with house dust mite group 4 allergens were found.</p> <p>Conclusions</p> <p>We present the first protein-level characterization of a group 4 allergen from storage mites. Due to its high production and IgE reactivity, Aca s 4 is potentially relevant to allergic hypersensitivity.</p

Central calcifying epithelial odontogenic tumour in the posterior maxilla: a case report

The calcifying epithelial odontogenic tumour (CEOT), or Pindborg tumour, is a rare, benign odontogenic tumour. CEOT is usually asymptomatic and an incidental radiological finding, often presenting as a mandibular radiolucency with flecks of calcific material. We report an unusual case of CEOT in the left posterior maxilla of a 46-year-old male that was associated with an unerupted tooth. The tumour in this case caused non-specific sinus symptoms and appeared radiographically similar to an odontoma or ossifying fibroma due to its dense calcific contents. Diagnosis was confirmed histologically following surgical removal of the lesion, which showed classic CEOT histomorphology. We report this case to highlight the unusual clinico-radiologic presentation and illustrate the diagnostic difficulties that can occur with radiolucent and/or radiopaque lesions in the jaws

V první části se zabývám problematikou zhoršené kvality uhlí, která nastává s postupným přesouváním těžby do nově otevřené lokality lomu Libouš II.-sever. Dále zde zmiňuji nutnost zlepšit proces míchání uhlí zařazením dalšího homogenizačního stroje umístěného na VKS. V další části mé práce zpracovávám technickou specifikaci modernizace technologie jednotlivých dílčích celků. V neposlední řadě popisuje návrh odvodnění pláně VKS.This work summarizes the knowledge and experience I have received in SD-1. Strojírenská, a. s. company, during the realization and modernization of technological mechanisms in DNT. In the first part of this thesis, I consider the issues surrounding the deteriorating quality of coal, which is the direct result of the progressive shift in coal mines to the new-opened location of the quarry Libouš II.-sever. Subsequently, I mention the necessity of improving the process of coal stirring by adding an extra homogenization machine placed on VKS. In the next section of my work I analyze the technical specifications of the technology of the individual partial wholes. V této diplomové práci se zabývám institutem nabytí od neoprávněného, který je prolomením římskoprávní zásady nemo plus iuris ad alium tranferre potest quam ipse habet, v českém překladu, že nikdo nemůže na jiného převést víc práv, než sám má. Nabytí od neoprávněného tak zasahuje do ústavně zaručeného práva vlastnit majetek a je proto důležité, aby jej právní úprava dostatečně reflektovala, kdy ne vždy tomu tak bylo. V diplomové práci se zabývám především vývojem tohoto institutu, zásadami, které jsou s tímto institutem spojeny a zejména analýzou a popisem současné právní úpravy. Práce je rozdělena do čtyř kapitol, kterým předchází úvod. Kapitoly se dále člení na podkapitoly, které mají podrobněji rozebrat dané téma. Práce je zakončena závěrem. V první kapitole se zabývám obecnou problematikou věcných práv. Dále zde rozebírám institut vlastnického práva, jeho ústavní východiska, definici i způsoby jakým je možné vlastnictví nabývat. Druhá kapitola je koncipována jako popsání historického vývoje nabytí od neoprávněného. Rozebírám nejdříve období římského práva, zejména pak kladu důraz na zásadu nemo plus iuris. Dále pokračují přes vývoj v germánském právu, až ke kodifikacím v 19. století. Zde se zabývám třemi velkými kodifikacemi ABGB, Code Civil a BGB. Cathepsin L from the hard tick Ixodes ricinus

Ticks are globally important parasites involved in transmission of a wide variety of infectious agents. The most common tick species found in Europe is the hard tick Ixodes ricinus, which transmits bacterium Borrelia burgdorferi (a causative agent of Lyme disease) or tick-borne encephalitis virus. Cathepsin proteases are important in the process of digestion of blood proteins in the tick gut. This work is focused on cathepsin L, an important digestive cysteine protease of ticks. Recombinant I. ricinus cathepsin L was expressed in Pichia pastoris and separated from the culture medium by chromatographic purification. N-terminal protein sequencing and labeling by activity-based probe Green-DCG-04 were used for characterization of purified cathepsin L. Substrate and inhibitor specificity were analyzed using peptide substrates and inhibitors. This analysis showed that Z-FR-AMC is a suitable substrate with pH optimum 3.5, and that Z-FF-DMK is an efficient inhibitor. It was demonstrated that cathepsin L cleaves protein substrates in strongly acidic environment (pH 3.5-4.5). Cathepsin L-like proteolytic activity was demonstrated in salivary gland extract and in saliva of the I. ricinus tick. The Opportunities of Modernization for the Engines of the Bucket Wheel Excavator KU 800.20

Import 04/07/2011Tato práce shrnuje poznatky a zkušenosti které jsem získal ve firmě SD-1.Strojírenská a.s. při realizaci některých uváděných modernizací rypadla KU 800.20. V první části popisuji kolesové rypadlo KU800.20 před provedenou modernizací a dále se zaobírám problematikou nasazení rypadla v lokalitě Dolů Nástup Tušimice. V další kapitole je popsána výměna špičky kolesového výložníku a uvádím technická data nového kolesa s pohonem. V dalších částech mé práce popisuji technické inovace pohonů základních pohybů stroje, tzn. výsuv a zdvih kolesového výložníku, otoč horní stavby a analyzuji problematiku převodovek pohonů jednotlivých pasů stroje. Na závěr mé práce uvádím ekonomické a technické přínosy, které by tyto modernizační změny, měly proti původnímu provedení kolesového rypadla přinést.This work summarizes knowledge and experiences which I got in the company SD-1. Strojírenská, a. s. during the realization of some of the mentioned modernization of excavator KU.800.20. In the first part of my work I am describing bucket wheel excavator before the modernization and I am also writing about problems of using the excavator in the area of mines Nástup Tušimice. The exchange of the tip of bucket-wheel jib is described in the next chapter. I will also mention technical information about new paddlewheel drive. In the next parts of my work I am describing technical innovations of drives of basic machine motion which means extension and lifting of bucket wheel jib and upper construction turn and I am also analysing the problems of transmission drives of particular strips of machine.
