    SPATIAL ECOLOGY AND HABITAT USE OF THE WESTERN MASSASAUGA (SISTRURUS TERGEMINUS) IN NEBRASKA

    We used radiotelemetry during 2004 and 2005 to investigate seasonal movements and habitat use and to improve our understanding of how land management practices were affecting Western Massasauga (Sistrurus tergeminus) populations in Nebraska. We found Western Massasaugas to brumate in crayfish burrows 100% of the time, and spring emergence occurred during March - April. The longest movements occurred during May when snakes were moving away from brumation sites to summer habitat, after which movements decreased until a second peak in August. We did not find significant differences between male and female activity area, range length, total distance moved, daily movement, or distance from spring and fall brumation sites. However the core activity range of females was significantly less than that of males. During the active season, grassland habitat was used 75.6% of the time compared to habitats of grassland-shrubs (10.7%), hydrophilic vegetation (9.1%), woodlands (3.1%), and shrublands (1.5%). Identifying variations in a species spatial ecology is important for achieving long-term conservation goals because life history events, resource availability, and land management practices may have an effect on seasonal movements and habitat use. The results of our study demonstrate the need for, and some of the necessary data for, improved land management planning in order to meet conservation needs of the Western Massasauga in Nebraska

    Subcellular Localization of the Leucine Biosynthetic Enzymes in Yeast

    When baker's yeast spheroplasts were lysed by mild osmotic shock, practically all of the isopropylmalate isomerase and the Ī²-isopropylmalate dehydrogenase was released into the 30,000 Ɨ g supernatant fraction, as was the cytosol marker enzyme, glucose-6-phosphate dehydrogenase. Ī±-Isopropylmalate synthase, however, was not detected in the initial supernatant, but could be progressively solubilized by homogenization, appearing more slowly than citrate synthase but faster than cytochrome oxidase. Of the total glutamate-Ī±-ketoisocaproate transaminase activity, approximately 20% was in the initial soluble fraction, whereas solubilization of the remainder again required homogenization of the spheroplast lysate. Results from sucrose density gradient centrifugation of a cell-free particulate fraction and comparison with marker enzymes suggested that Ī±-isopropylmalate synthase was located in the mitochondria. It thus appears that, in yeast, the first specific enzyme in the leucine biosynthetic pathway (Ī±-isopropylmalate synthase) is particulate, whereas the next two enzymes in the pathway (isopropylmalate isomerase and Ī²-isopropylmalate dehydrogenase) are ā€œsoluble,ā€ with glutamate-Ī±-ketoisocaproate transaminase activity being located in both the cytosol and particulate cell fractions

    Detection of a Substantial Molecular Gas Reservoir in a brightest cluster galaxy at z = 1.7

    We report the detection of CO(2-1) emission coincident with the brightest cluster galaxy (BCG) of the high-redshift galaxy cluster SpARCS1049+56, with the Redshift Search Receiver (RSR) on the Large Millimetre Telescope (LMT). We confirm a spectroscopic redshift for the gas of z = 1.7091+/-0.0004, which is consistent with the systemic redshift of the cluster galaxies of z = 1.709. The line is well-fit by a single component Gaussian with a RSR resolution-corrected FWHM of 569+/-63 km/s. We see no evidence for multiple velocity components in the gas, as might be expected from the multiple image components seen in near-infrared imaging with the Hubble Space Telescope. We measure the integrated flux of the line to be 3.6+/-0.3 Jy km/s and, using alpha_CO = 0.8 Msun (K km s^-1 pc^2)^-1 we estimate a total molecular gas mass of 1.1+/-0.1x10^11 Msun and a M_H2/M_star ~ 0.4. This is the largest gas reservoir detected in a BCG above z > 1 to date. Given the infrared-estimated star formation rate of 860+/-130 Msun/yr, this corresponds to a gas depletion timescale of ~0.1Gyr. We discuss several possible mechanisms for depositing such a large gas reservoir to the cluster center -- e.g., a cooling flow, a major galaxy-galaxy merger or the stripping of gas from several galaxies -- but conclude that these LMT data are not sufficient to differentiate between them.Comment: accepted for publication in ApJ Letter

    This NebGuide describes how to use a chlorophyll meter as a tool to improve nitrogen management by detecting nitrogen deficiency and determining the need for additional N fertilizer. Fertilizer nitrogen (N) is increasingly recognized as the source of nitrate contamination in much of Nebraska\u27s groundwater. Improving the efficiency of fertilizer N use reduces the amount of N that can potentially contaminate water resources. Effective management of fertilizer N is a major challenge for grain crop producers. Many factors that affect its efficiency are beyond a producer\u27s control. Weather, equipment limitations and breakdowns, and availability of labor and fertilizer during critical periods can lead to inadequate N supply to the crop. Fertilizer N is relatively inexpensive, and deficiencies can result in substantial yield reductions. Producers are inclined to manage fertilizer N to minimize the risk of deficiency, which can lead to excessive fertilizer applications. Although they understand fertilizer applied at excessive rates costs money and may lead to contamination of the environment, producers also want assurance that applying less fertilizer N will not reduce crop yields

    Crack opening behavior in ceramic matrix composites

    The evolution of matrix cracks in a meltā€infiltrated SiC/SiC ceramic matrix composite (CMC) under uniaxial tension was examined using scanning electron microscopy (SEM) combined with digital image correlation (DIC) and manual crack opening displacement (COD) measurements. CMC modeling and life prediction strongly depend a thorough understanding of when matrix cracks occur, the extent of cracking for given conditions (timeā€temperatureā€environmentā€stress), and the interactions of matrix cracks with fibers and interfaces. In this work, strain relaxation due to matrix cracking, the relationship between CODs and applied stress, and damage evolution at stresses below the proportional limit were assessed. Direct experimental observation of strain relaxation adjacent to regions of matrix cracking is presented and discussed. Additionally, crack openings were found to increase linearly with increasing applied stress, and no crack was found to pass fully through the gage crossā€section. This calls into question the modeling assumption of throughā€cracks for all loading conditions and fiber architectures, which can obscure oxidation mechanisms that are active in realistic cracking conditions.     Increasing subsequences and the hard-to-soft edge transition in matrix ensembles

    Our interest is in the cumulative probabilities Pr(L(t) \le l) for the maximum length of increasing subsequences in Poissonized ensembles of random permutations, random fixed point free involutions and reversed random fixed point free involutions. It is shown that these probabilities are equal to the hard edge gap probability for matrix ensembles with unitary, orthogonal and symplectic symmetry respectively. The gap probabilities can be written as a sum over correlations for certain determinantal point processes. From these expressions a proof can be given that the limiting form of Pr(L(t) \le l) in the three cases is equal to the soft edge gap probability for matrix ensembles with unitary, orthogonal and symplectic symmetry respectively, thereby reclaiming theorems due to Baik-Deift-Johansson and Baik-Rains.Comment: LaTeX, 19 page

    Fosmidomycin Decreases Membrane Hopanoids and Potentiates the Effects of Colistin on Burkholderia multivorans Clinical Isolates

    Burkholderia cepacia complex (Bcc) pulmonary infections in people living with cystic fibrosis (CF) are difficult to treat because of the extreme intrinsic resistance of most isolates to a broad range of antimicrobials. Fosmidomycin is an antibacterial and antiparasitic agent that disrupts the isoprenoid biosynthesis pathway, a precursor to hopanoid biosynthesis. Hopanoids are involved in membrane stability and contribute to polymyxin resistance in Bcc bacteria. Checkerboard MIC assays determined that although isolates of the Bcc species B. multivorans were highly resistant to treatment with fosmidomycin or colistin (polymyxin E), antimicrobial synergy was observed in certain isolates when the antimicrobials were used in combination. Treatment with fosmidomycin decreased the MIC of colistin for isolates as much as 64-fold to as low as 8 Ī¼g/ml, a concentration achievable with colistin inhalation therapy. A liquid chromatography-tandem mass spectrometry technique was developed for the accurate quantitative determination of underivatized hopanoids in total lipid extracts, and bacteriohopanetetrol cyclitol ether (BHT-CE) was found to be the dominant hopanoid made by B. multivorans. The amount of BHT-CE made was significantly reduced upon fosmidomycin treatment of the bacteria. Uptake assays with 1-N-phenylnaphthylamine were used to determine that dual treatment with fosmidomycin and colistin increases membrane permeability, while binding assays with boron-dipyrromethene-conjugated polymyxin B illustrated that the addition of fosmidomycin had no impact on polymyxin binding. This work indicates that pharmacological suppression of membrane hopanoids with fosmidomycin treatment can increase the susceptibility of certain clinical B. multivorans isolates to colistin, an agent currently in use to treat pulmonary infections in CF patients

    A Comparison Study of Eleven Static Heuristics for Mapping a Class of Independent Tasks onto Ileterogeneous Distributed Computing Systems

    ABSTRACT Il\u27lixed-machine heterogeneous computing (HC) environments utilize a distributed suite of different high-performance machines, interconnected with high-speed links to perform different computationally intensive applications that have diverse comput ational requirements. HC environments are well suited to meet thl: computational dell-tands of large, diverse groups of tasks. The problem of mapping (defined as matching and scheduling) these tasks onto the machines of a distributed HC environment has been shown, in general, to be NP-complete, requiring the development of heuristic techniques. Selecting the best heuristic to use in a given enviroi~menth, owever, remains a difficult problem, because comparisons are often clouded by different underlying assumptions in the original studies of each heuristic. There~fore; a collection of eleven heuristics from the literature has been selected: a,dapted, in~plementeda, nd anaiyzed under one set of common assumptions. It is assumed that the heuristics derive a, mapping statically (i.e., off-line). It is also assumed that a meta-task (i.e., a set of independent, non-communicating tasks) is being mapped, and that the goal is to minimize the total execution time of the metla-task. The eleven heuristics examined are Opportunistic Load Balancing, Minimum Execution Time, MininLlum Clompletion Time, Min-min, hllax-min, Duplex? Genetic i-Ilgorithm, Simulated Annealing, Genetic Simulat.ed .Annealing, Tabu, and Ax. This study provides one even basis for comparisor] and insights into circumstances where one technique will out perform another. The evaluation procedure is specified, the heuristics are defined, and then comparison results are discussed. It is shown that for the ca.ses studied here, the relat,ively simple Min-min heuristic performs well in comparison to the other techniques
