    Cyclosporine augments hepatic regenerative response in rats

    A number of mechanisms participate in the hepatic injury that occurs during and following liver transplantation. A normal allograft regenerative response is probably essential for a successful transplant outcome. In this study, the effect of cyclosporine, a potent immunosuppressant used routinely after liver transplantation, on the regenerative response of the liver after partial hepatectomy was investigated. Male Wistar rats were pretreated for one week with either cyclosporine or the olive oil vehicle and were subjected to either a two-thirds partial hepatectomy or a sham operation. Animals were sacrificed at various times postoperatively and the remnant livers were weighed to determine the liver weight to body weight ratio, two biochemical measures of a regenerative response (cytosolic ornithine decarboxylase activity and thymidine kinase activity), and the hepatic content of estrogen and androgen receptors, as the content of these receptors has been shown to modulate, at least in part, the subsequent hepatic regenerative response. The preoperative hepatic cytosol content of ornithine decarboxylase, thymidine kinase, and estrogen receptor was significantly greater (P<0.05) in rats pretreated with cyclosporine than in those treated with the vehicle alone. A significant increase in ornithine decarboxylase and thymidine kinase activities occurred after partial hepatectomy in both the cyclosporine-pretreated and vehicle-pretreated animals. The absolute levels for each parameter were also greater in the cyclosporine-treated animals than in the vehicle-treated controls at 24 hr after partial hepatectomy (P<0.05). The pattern of change in the hepatic cytosolic content of estrogen and androgen receptors in both groups of animals was comparable with those described previously for regenerating liver. These data suggest that cyclosporine may predispose the liver to respond to either a regenerative signal or perceived need and thereby fortuitously enhance liver graft performance after successful surgical implantation. © 1989 Plenum Publishing Corporation

    Overtones of the Si−H Stretching−Bending Polyad in SiHD<sub>3</sub>: Internal Coordinate Force Field, ab initio Dipole Moment Surfaces, and Band Intensities

    Overtones of the Si−H stretching−bending polyad of the SiHD3 molecule are studied using an internal coordinate force field model. The potential parameters are optimized by fitting to the experimental band centers. The Fermi resonance between the Si−H stretching and bending motions is insignificant due to cancellation of the contributions from kinetic and potential terms. This suggests a slow redistribution of vibrational energy between these two degrees of freedom and induces local mode character of respective vibrations. Band intensities are calculated by using ab initio one- and three-dimensional dipole moment surfaces (DMS). These agree reasonably well with the observations. The successful reproduction of relative intensities between the (n1 − 1)ʋ1 + 2ʋ5 stretching−bending combination bands and the n1ʋ1 stretching bands establishes the importance of the bending motion in the multidimensional DMS for intensity investigations

    Cryostratigraphy, sedimentology, and the late Quaternary evolution of the Zackenberg River delta, northeast Greenland

    The Zackenberg River delta is located in northeast Greenland (74°30′ N, 20°30′ E) at the outlet of the Zackenberg fjord valley. The fjord-valley fill consists of a series of terraced deltaic deposits (ca. 2 km2) formed during relative sea-level (RSL) fall. We investigated the deposits using sedimentological and cryostratigraphic techniques together with optically stimulated luminescence (OSL) dating. We identify four facies associations in sections (4 to 22 m in height) exposed along the modern Zackenberg River and coast. Facies associations relate to (I) overriding glaciers, (II) retreating glaciers and quiescent glaciomarine conditions, (III) delta progradation in a fjord valley, and (IV) fluvial activity and niveo-aeolian processes. Pore, layered, and suspended cryofacies are identified in two 20 m deep ice-bonded sediment cores. The cryofacies distribution, together with low overall ground-ice content, indicates that permafrost is predominately epigenetic in these deposits. Fourteen OSL ages constrain the deposition of the cored deposits to between approximately 13 and 11 ka, immediately following deglaciation. The timing of permafrost aggradation was closely related to delta progradation and began following the subaerial exposure of the delta plain (ca. 11 ka). Our results reveal information concerning the interplay between deglaciation, RSL change, sedimentation, permafrost aggradation, and the timing of these events. These findings have implications for the timing and mode of permafrost aggradation in other fjord valleys in northeast Greenland

    The chaotic behavior of the black hole system GRS 1915+105

    A modified non-linear time series analysis technique, which computes the correlation dimension D2D_2, is used to analyze the X-ray light curves of the black hole system GRS 1915+105 in all twelve temporal classes. For four of these temporal classes D2D_2 saturates to ≈4−5\approx 4-5 which indicates that the underlying dynamical mechanism is a low dimensional chaotic system. Of the other eight classes, three show stochastic behavior while five show deviation from randomness. The light curves for four classes which depict chaotic behavior have the smallest ratio of the expected Poisson noise to the variability (<0.05 < 0.05) while those for the three classes which depict stochastic behavior is the highest (>0.2 > 0.2). This suggests that the temporal behavior of the black hole system is governed by a low dimensional chaotic system, whose nature is detectable only when the Poisson fluctuations are much smaller than the variability.Comment: Accepted for publication in Astrophysical Journa

    The effect of different types of hepatic injury on the estrogen and androgen receptor activity of liver

    Mammalian liver contains receptors for both estrogens and androgens. Hepatic regeneration after partial hepatectomy in male rats is associated with a loss of certain male-specific hepatic characteristics. In this study we investigated the effects of lesser forms of hepatic injury on the levels of estrogen and androgen receptor activity in the liver. Adult male rats were subjected to portacaval shunt, partial portal vein ligation, hepatic artery ligation, or two-thirds partial hepatectomy. Another group of animals was treated with cyclosporine. At the time of sacrifice the livers were removed and used to determine the estrogen and androgen receptor activity in the hepatic cytosol. A significant reduction (p < 0.05) in the hepatic cytosolic androgen receptor activity and a slight increase in the estrogen receptor activity occurred following total portosystemic shunting. Partial ligation of the portal vein, which produces a lesser degree of portosystemic shunting, had no effect on the levels of the estrogen and androgen receptor activity present within hepatic cytosol. Cyclosporine-treated animals had significantly greater (p < 0.01) levels of estrogen receptor activity in the hepatic cytosol compared to vehicle-treated control animals. Levels of estrogen and androgen receptor activity within the hepatic cytosol remained unchanged after ligation of the hepatic artery. The reduction in the cytosolic estrogen and androgen receptor activity in the liver after partial hepatectomy was confirmed. In summary, certain types of hepatic injury are associated with profound changes in the estrogen and androgen receptor content within the liver. © 1989 Informa UK Ltd All rights reserved: reproduction in whole or part not permitted
