    Wettability conundrum: Discrepancies of soft contact lens performance in vitro and in vivo

    The recognition and appreciation of soft contact lenses as simple, efficient and aesthetically gratifying vision-correction devices is ever growing, especially among younger population. Stable thin tear film uniformly spread over corrective lens surface is essential for acute vision, and also for comfortable and safe contact lens wear. The significant efforts have been invested by the contact lens industry to develop soft lens surface that is completely wet by tear aqueous in the ocular environment. Number of the publications dedicated to the wettability properties of the soft hydrogel lenses is on the steady rise. However, the clinical results show that no unambiguous correlation emerges when lens surface wettability in vitro is judged against tear film stability evaluated in vivo. This paper assesses and compares the modern techniques used for evaluation of soft contact lens surface wettability and reports some findings regarding relations between lens surface wettability in vitro and in vivo. © 2011 EDP Sciences and Springer

    Space-efficient Feature Maps for String Alignment Kernels

    String kernels are attractive data analysis tools for analyzing string data. Among them, alignment kernels are known for their high prediction accuracies in string classifications when tested in combination with SVM in various applications. However, alignment kernels have a crucial drawback in that they scale poorly due to their quadratic computation complexity in the number of input strings, which limits large-scale applications in practice. We address this need by presenting the first approximation for string alignment kernels, which we call space-efficient feature maps for edit distance with moves (SFMEDM), by leveraging a metric embedding named edit sensitive parsing (ESP) and feature maps (FMs) of random Fourier features (RFFs) for large-scale string analyses. The original FMs for RFFs consume a huge amount of memory proportional to the dimension d of input vectors and the dimension D of output vectors, which prohibits its large-scale applications. We present novel space-efficient feature maps (SFMs) of RFFs for a space reduction from O(dD) of the original FMs to O(d) of SFMs with a theoretical guarantee with respect to concentration bounds. We experimentally test SFMEDM on its ability to learn SVM for large-scale string classifications with various massive string data, and we demonstrate the superior performance of SFMEDM with respect to prediction accuracy, scalability and computation efficiency.Comment: Full version for ICDM'19 pape

    Comparison of respondent-reported and sensor-recorded latrine utilization measures in rural Bangladesh: a cross-sectional study.

    Background: Health improvements realized through sanitation are likely achieved through high levels of facilities utilization by all household members. However, measurements of sanitation often rely on either the presence of latrines, which does not guarantee use, or respondent-reported utilization of sanitation facilities, which is prone to response bias. Overstatement of sanitation metrics limits the accuracy of program outcome measures, and has implications for the interpretation of related health impact data. Methods: We conducted a cross-sectional study of 213 households in 14 village water, sanitation and hygiene committee clusters throughout rural Bangladesh and used a combined data- and relationship-scale approach to assess agreement between respondent-reported latrine utilization and sensor-recorded measurement. Results: Four-day household-level respondent-reported defecation averaged 28 events (inter-quartile range [IQR] 20-40), while sensor-recorded defecation averaged 17 events (IQR 11-29). Comparative analyses suggest moderately high accuracy (bias correction factor=0.84), but imprecision in the data (broad scatter of data, Pearson's r=0.35) and thus only weak concordance between measures (ρc=0.29 [95% BCa CI 0.15 to 0.43]). Conclusions: Respondent-reported latrine utilization data should be interpreted with caution, as evidence suggests use is exaggerated. Coupling reported utilization data with objective measures of use may aid in the estimation of latrine use

    Destabilization of the thermohaline circulation by transient perturbations to the hydrological cycle

    We reconsider the problem of the stability of the thermohaline circulation as described by a two-dimensional Boussinesq model with mixed boundary conditions. We determine how the stability properties of the system depend on the intensity of the hydrological cycle. We define a two-dimensional parameters' space descriptive of the hydrology of the system and determine, by considering suitable quasi-static perturbations, a bounded region where multiple equilibria of the system are realized. We then focus on how the response of the system to finite-amplitude surface freshwater forcings depends on their rate of increase. We show that it is possible to define a robust separation between slow and fast regimes of forcing. Such separation is obtained by singling out an estimate of the critical growth rate for the anomalous forcing, which can be related to the characteristic advective time scale of the system.Comment: 37 pages, 8 figures, submitted to Clim.     Functional significance of the hepaCAM gene in bladder cancer

    <p>Abstract</p> <p>Background</p> <p>The hepaCAM gene encodes a new immunoglobulin-like cell adhesion molecule, and its expression is suppressed in a variety of human cancers. Additionally, hepaCAM possesses properties often observed in tumor suppressor genes. However, the expression and biological function of hepaCAM has not been investigated in bladder cancer. Therefore we sought to examine hepaCAM expression and the relationship between its structure and function in human transitional cell carcinoma of bladder (TCCB).</p> <p>Materials and methods</p> <p>HepaCAM expression was evaluated in 28 normal and 34 TCCB bladder specimens and 2 TCCB cell lines using semi-quantitative RT-PCR. The wild-type hepaCAM and the extracellular domain-truncated mutant gene were transfected into the TCCB cell line T24, and the biological properties of both the wild-type gene and the domain-truncated mutant were then assessed.</p> <p>Results</p> <p>HepaCAM expression was down-regulated in 82% (28/34) of TCCB specimens and undetectable in the 2 TCCB cell lines tested. The localization of hepaCAM appeared to be dependent on cell density in T24 cells. In widely spread cells, hepaCAM accumulated on the perinuclear membrane and the cell surface protrusions, whereas in confluent cells, hepaCAM was predominantly localized at the sites of cell-cell contacts on the cell membrane. Functionally, hepaCAM expressed not only increased cell spreading, delayed cell detachment, enhanced wound healing and increased cell invasion; it also inhibited cell growth (P < 0.01). When the extracellular domain was deleted, the localization of hepaCAM was significantly altered, and it lost both its adhesive function and its influence on cell growth.</p> <p>Conclusions</p> <p>HepaCAM is involved in cell adhesion and growth control, and its expression is frequently silenced in TCCB. The extracellular domain of hepaCAM is essential to its physiological and biological functions.</p

    Characterising half a degree difference: a review of methods for identifying regional climate responses to global warming targets

    The Paris Agreement long term global temperature goal refers to two global warming levels: well below 2°C and 1.5°C above preindustrial. Regional climate signals at specific global warming levels, and especially the differences between 1.5°C and 2°C, are not well constrained, however. In particular, methodological challenges related to the assessment of such differences have received limited attention. This paper reviews alternative approaches for identifying regional climate signals associated with global temperature limits, and evaluates the extent to which they constitute a sound basis for impacts analysis. Four methods are outlined, including comparing data from different greenhouse gas scenarios, sub-selecting climate models based on global temperature response, pattern scaling, and extracting anomalies at the time of each global temperature increment. These methods have rarely been applied to compare 2°C with 1.5°C, but some demonstrate potential avenues for useful research. Nevertheless, there are methodological challenges associated with the use of existing climate model experiments, which are generally designed to model responses to different levels of greenhouse gas forcing, rather than to model climate responses to a specific level of forcing that targets a given level of global temperature change. Novel approaches may be required to address policy questions, in particular: to differentiate between half degree warming increments while accounting for different sources of uncertainty; to examine mechanisms of regional climate change including the potential for nonlinear responses; and to explore the relevance of time-lagged processes in the climate system and declining emissions, and the resulting sensitivity to alternative mitigation pathways

    Vocal Communications and the Maintenance of Population Specific Songs in a Contact Zone

    Bird song has been hypothesized to play a role in several important aspects of the biology of songbirds, including the generation of taxonomic diversity by speciation; however, the role that song plays in speciation within this group may be dependent upon the ability of populations to maintain population specific songs or calls in the face of gene flow and external cultural influences. Here, in an exploratory study, we construct a spatially explicit model of population movement to examine the consequences of secondary contact of populations singing distinct songs. We concentrate on two broad questions: 1) will population specific songs be maintained in a contact zone or will they be replaced by shared song, and 2) what spatial patterns in the distribution of songs may result from contact? We examine the effects of multiple factors including song-based mating preferences and movement probabilities, oblique versus paternal learning of song, and both cultural and genetic mutations. We find a variety of conditions under which population specific songs can be maintained, particularly when females have preferences for their population specific songs, and we document many distinct patterns of song distribution within the contact zone, including clines, banding, and mosaics
