UNDERSTANDING THE GRAY: AGING WOMEN IN VICTORIAN CULTURE AND FICTION
My dissertation, Understanding the Gray:Aging Women in Victorian Culture and Fiction, explores the cultural construction of aging for middle-class Victorian women and how aging was experienced and then depicted within novels. Chiefly, I work from midcentury to the end of the century in order to understand the experience of aging and ways women were ascribed age due to their position in society as spinsters, mothers, and progressive women. I explore how the age of fictional women reflects and contributes to critical debates concerning how Victorian women were expected to behave. Debates over separate spheres, how women were perceived in British society, and how women’s rights changed during the 19th century highlight how aging affected women and how they were treated throughout the century. Victorian fiction illustrates the ways women achieved different roles in society and how age and the perception of age affected their ability to do so. Understanding how aging was experienced, understood, and ascribed to Victorian women who fought in various ways for new terms of citizenship and mobility helps us begin to trace how we treat and respond to aging in women today. The first chapter outlines the social status of unmarried women and spinsters, considering how age affected women’s ability to lead professional lives in Charlotte Brontë’s Villette (1853). The second chapter, on George Eliot’s Felix Holt: The Radical, explores older motherhood through Mrs Transome and illustrates how the novel seeks to teach younger women of the pitfalls of unequal marriages. Broadband Phase-Noise Suppression in a Yb-Fiber Frequency Comb
We report a simple technique to suppress high frequency phase noise of a
Yb-based fiber optical frequency comb using an active intensity noise servo.
Out-of-loop measurements of the phase noise using an optical heterodyne beat
with a continuous wave (cw) laser show suppression of phase noise by \geq7 dB
out to Fourier frequencies of 100 kHz with a unity-gain crossing of -700 kHz.
These results are enabled by the strong correlation between the intensity and
phase noise of the laser. Detailed measurements of intensity and phase noise
spectra, as well as transfer functions, reveal that the dominant phase and
intensity noise contribution above -100 kHz is due to amplified spontaneous
emission (ASE) or other quantum noise sources.Comment: 4 pages, 3 figure
Quantitative Characterization of the T Cell Receptor Repertoire of Naive and Memory subsets Using an Integrated experimental and Computational Pipeline Which Is Robust, economical, and Versatile
The T cell receptor (TCR) repertoire can provide a personalized biomarker for infectious and non-infectious diseases. We describe a protocol for amplifying, sequencing, and analyzing TCRs which is robust, sensitive, and versatile. The key experimental step is ligation of a single-stranded oligonucleotide to the 3′ end of the TCR cDNA. This allows amplification of all possible rearrangements using a single set of primers per locus. It also introduces a unique molecular identifier to label each starting cDNA molecule. This molecular identifier is used to correct for sequence errors and for effects of differential PCR amplification efficiency, thus producing more accurate measures of the true TCR frequency within the sample. This integrated experimental and computational pipeline is applied to the analysis of human memory and naive subpopulations, and results in consistent measures of diversity and inequality. After error correction, the distribution of TCR sequence abundance in all subpopulations followed a power law over a wide range of values. The power law exponent differed between naïve and memory populations, but was consistent between individuals. The integrated experimental and analysis pipeline we describe is appropriate to studies of T cell responses in a broad range of physiological and pathological contexts
International energy agency ocean energy systems task 10 wave energy converter modeling verification and validation
This is the first joint reference paper for the Ocean
Energy Systems (OES) Task 10 Wave Energy Converter modeling
verification and validation group. The group is established
under the OES Energy Technology Network program under the
International Energy Agency. OES was founded in 2001 and
Task 10 was proposed by Bob Thresher (National Renewable
Energy Laboratory) in 2015 and approved by the OES Executive
Committee EXCO in 2016. The kickoff workshop took place in
September 2016, wherein the initial baseline task was defined.
Experience from similar offshore wind validation/verification
projects (OC3-OC5 conducted within the International Energy
Agency Wind Task 30) [1], [2] showed that a simple test
case would help the initial cooperation to present results in
a comparable way. A heaving sphere was chosen as the first
test case. The team of project participants simulated different
numerical experiments, such as heave decay tests and regular
and irregular wave cases. The simulation results are presented

Speciated measurements of semivolatile and intermediate volatility organic compounds (S/IVOCs) in a pine forest during BEACHON-RoMBAS 2011
Understanding organic composition of gases and particles is essential to identifying sources and atmospheric processing leading to organic aerosols (OA), but atmospheric chemical complexity and the analytical techniques available often limit such analysis. Here we present speciated measurements of semivolatile and intermediate volatility organic compounds (S/IVOCs) using a novel dual-use instrument (SV-TAG-AMS) deployed at Manitou Forest, CO, during the Bio-hydro-atmosphere interactions of Energy, Aerosols, Carbon, H_2O, Organics & Nitrogen – Rocky Mountain Biogenic Aerosol Study (BEACHON-RoMBAS) 2011 campaign. This instrument provides on-line speciation of ambient organic compounds with 2 h time resolution. The species in this volatility range are complex in composition, but their chemical identities reveal potential sources. Observed compounds of biogenic origin include sesquiterpenes with molecular formula C_(15)H_(24) (e.g., β-caryophyllene and longifolene), which were most abundant at night. A variety of other biogenic compounds were observed, including sesquiterpenoids with molecular formula C_(15)H_(22), abietatriene and other terpenoid compounds. Many of these compounds have been identified in essential oils and branch enclosure studies but were observed in ambient air for the first time in our study. Semivolatile polycyclic aromatic hydrocarbons (PAHs) and alkanes were observed with highest concentrations during the day and the dependence on temperature suggests the role of an evaporative source. Using statistical analysis by positive matrix factorization (PMF), we classify observed S/IVOCs by their likely sources and processes, and characterize them based on chemical composition. The total mass concentration of elutable S/IVOCs was estimated to be on the order of 0.7 µg m^(−3) and their volatility distributions are estimated for modeling aerosol formation chemistry
Worldwide data sets constrain the water vapor uptake coefficient in cloud formation
Cloud droplet formation depends on the condensation of water vapor on ambient aerosols, the rate of which is strongly affected by the kinetics of water uptake as expressed by the condensation (or mass accommodation) coefficient, α_c. Estimates of α_c for droplet growth from activation of ambient particles vary considerably and represent a critical source of uncertainty in estimates of global cloud droplet distributions and the aerosol indirect forcing of climate. We present an analysis of 10 globally relevant data sets of cloud condensation nuclei to constrain the value of αc for ambient aerosol. We find that rapid activation kinetics (α_c > 0.1) is uniformly prevalent. This finding resolves a long-standing issue in cloud physics, as the uncertainty in water vapor accommodation on droplets is considerably less than previously thought
A hepatoprotective Lindera obtusiloba extract suppresses growth and attenuates insulin like growth factor-1 receptor signaling and NF-kappaB activity in human liver cancer cell lines
<p>Abstract</p> <p>Background</p> <p>In traditional Chinese and Korean medicine, an aqueous extract derived from wood and bark of the Japanese spice bush <it>Lindera obtusiloba </it>(<it>L.obtusiloba</it>) is applied to treat inflammations and chronic liver diseases including hepatocellular carcinoma. We previously demonstrated anti-fibrotic effects of <it>L.obtusiloba </it>extract in hepatic stellate cells. Thus, we here consequently examine anti-neoplastic effects of <it>L.obtusiloba </it>extract on human hepatocellular carcinoma (HCC) cell lines and the signaling pathways involved.</p> <p>Methods</p> <p>Four human HCC cell lines representing diverse stages of differentiation were treated with <it>L.obtusiloba </it>extract, standardized according to its known suppressive effects on proliferation and TGF-β-expression. Beside measurement of proliferation, invasion and apoptosis, effects on signal transduction and NF-κB-activity were determined.</p> <p>Results</p> <p><it>L.obtusiloba </it>extract inhibited proliferation and induced apoptosis in all HCC cell lines and provoked a reduced basal and IGF-1-induced activation of the IGF-1R signaling cascade and a reduced transcriptional NF-κB-activity, particularly in the poorly differentiated SK-Hep1 cells. Pointing to anti-angiogenic effects, <it>L.obtusiloba </it>extract attenuated the basal and IGF-1-induced expression of hypoxia inducible factor-1α, vascular endothelial growth factor, peroxisome proliferator-activated receptor-γ, cyclooxygenase-2 and inducible nitric oxide synthase.</p> <p>Conclusions</p> <p>The traditional application of the extract is confirmed by our experimental data. Due to its potential to inhibit critical receptor tyrosine kinases involved in HCC progression via the IGF-1 signaling pathway and NF-κB, the standardized <it>L.obtusiloba </it>extract should be further analysed for its active compounds and explored as (complementary) treatment option for HCC.</p
