    Lens epithelial cell apoptosis and intracellular Ca(2+) increase in the presence of xanthurenic acid

    BACKGROUND: Xanthurenic acid is an endogenous product of tryptophan degradation by indoleamine 2,3-dioxygenase (IDO). We have previously reported that IDO is present in mammalian lenses, and xanthurenic acid is accumulated in the lenses with aging. Here, we studied the involvement of xanthurenic acid in the human lens epithelial cell physiology. METHODS: Human lens epithelial cells primary cultures were used. Control cells, and cells in the presence of xanthurenic acid grow in the dark. Western blot analysis and immunofluorescence studies were performed. RESULTS: In the presence of xanthurenic acid human lens epithelial cells undergo apoptosis-like cell death. In the control cells gelsolin stained the perinuclear region, whereas in the presence of 10 μM xanthurenic acid gelsolin is translocated to the cytoskeleton, but does not lead to cytoskeleton breakdown. In the same condition caspase-3 activation, and DNA fragmentation was observed. At low (5 to 10 μM) of xanthurenic acid concentration, the elongation of the cytoskeleton was associated with migration of mitochondria and cytochrome c release. At higher concentrations xanthurenic acid (20 μM and 40 μM) damaged mitochondria were observed in the perinuclear region, and nuclear DNA cleavage was observed. We observed an induction of calpain Lp 82 and an increase of free Ca(2+) in the cells in a xanthurenic acid concentration-dependent manner. CONCLUSIONS: The results show that xanthurenic acid accumulation in human lens epithelial cells disturbs the normal cell physiology and leads to a cascade of pathological events. Xanthurenic acid induces calpain Lp82 and caspases in the cells growing in the dark and can be involved in senile cataract development

    Pulsatile Wall Motion (PWM) Measurements after Endovascular Abdominal Aortic Aneurysm Exclusion are not Useful in the Classification of Endoleak

    AbstractThe pulsatile wall motion (PWM) of AAA is reduced after endovascular stent-graft placement. The purpose of this study was to identify whether PWM after endografting was useful in the classification of endoleak.Patients and Methods162 patients treated with EVAR underwent pre- and post-operative PWM assessment with ultrasonography. Follow-up was 1–9 years. 111 patients had well-excluded aneurysms, three patients had enlarging aneurysms without any recognizable endoleak (endotension), 16 had type I, 31 had type II and 1 had type III endoleak.ResultsThe PWM was reduced from about 1mm pre-operatively to 0.24mm post-operatively in well-excluded aneurysms. PWM remained stable during follow-up. Type I endoleak was associated with moderately reduced PWM (proximal endoleak 0.79mm and distal 0.32mm). PWM in patients with type II endoleak was higher (0.32mm) post-operatively (p=0.002) compared to well-excluded aneurysms.ConclusionPWM is permanently reduced after endografting. The smallest reduction in PWM was in patients with type II endoleaks. However, the overlap between the groups does not allow reliable identification of patients having endoleak with PWM-measurements

    In this paper corrosion resistance of experimental dental Co-Cr-X (X = Mo, Ni) alloys, high purity titanium and Ag-amalgam was studied in Ringer’s solution. On the basis of electrochemical measurements, the relevant indicator of general corrosion jcorr, which is used for ranking of materials, was determined. It was found that commercial alloy Wironit® is the most stable material followed by pure titanium, Co-Cr-Mo and Co-Cr-Ni alloys, while amalgam showed the lowest resistance in the experimental conditions of model oral cavity medium.U ovom radu ispitana je korozijska otpornost eksperimentalnih zubnih Co-Cr-X (X = Mo, Ni) slitina, titana i Ag-amalgama u Ringerovoj otopini. Na osnovi elektrokemijskih mjerenja određen je relevantni pokazatelj opće korozije jkor, koji se koristi za rangiranje materijala. Pokazalo se, da je korozijski najstabilnija komercijalna slitina Wironit®, koju slijede čisti titan, te Co-Cr-Mo i Co-Cr-Ni slitine, dok se amalgam pokazao najmanje otpornim u ispitnim uvjetima koji simuliraju medij usne šupljine

    Large-Amplitude Ultraviolet Variations in the RR Lyrae Star ROTSE-I J143753.84+345924.8

    The NASA Galaxy Evolution Explorer (GALEX) satellite has obtained simultaneous near and far ultraviolet light curves of the ROTSE-I Catalog RR Lyrae ab-type variable star J143753.84+345924.8. A series of 38 GALEX Deep Imaging Survey observations well distributed in phase within the star's 0.56432d period shows an AB=4.9mag variation in the far UV (1350-1750A) band and an AB=1.8mag variation in the near UV (1750-2750A) band, compared with only a 0.8mag variation in the broad, unfiltered ROTSE-I (4500-10000A) band. These GALEX UV observations are the first to reveal a large RR Lyrae amplitude variation at wavelengths below 1800A. We compare the GALEX and ROTSE-I observations to predictions made by recent Kurucz stellar atmosphere models. We use published physical parameters for the comparable period (0.57433d), well-observed RR Lyrae star WY Antliae to compute predicted FUV, NUV, and ROTSE-I light curves for J143753.84+345924.8. The observed light curves agree with the Kurucz predictions for [Fe/H]=-1.25 to within AB=0.2mag in the GALEX NUV and ROTSE-I bands, and within 0.5mag in the FUV. At all metallicities between solar and one hundredth solar, the Kurucz models predict 6-8mag of variation at wavelengths between 1000-1700A. Other variable stars with similar temperature variations, such as Cepheids, should also have large-amplitude FUV light curves, observable during the ongoing GALEX imaging surveys.Comment: This paper will be published as part of the Galaxy Evolution Explorer (GALEX) Astrophysical Journal Letters Special Issue.     The GALEX UV luminosity function of the cluster of galaxies Abell 1367

    We present the GALEX NUV (2310 A) and FUV (1530 A) galaxy luminosity functions of the nearby cluster of galaxies A1367 in the magnitude range -20.3< M_AB < -13.3. The luminosity functions are consistent with previous (~ 2 mag shallower) estimates based on the FOCA and FAUST experiments, but display a steeper faint-end slope than the GALEX luminosity function for local field galaxies. Using spectro-photometric optical data we select out star-forming systems from quiescent galaxies and study their separate contributions to the cluster luminosity function. We find that the UV luminosity function of cluster star-forming galaxies is consistent with the field. The difference between the cluster and field LF is entirely due to the contribution at low luminosities (M_AB >-16 mag) of non star-forming, early-type galaxies that are significantly over dense in clusters.Comment: 4 pages, 4 figures, 1 table.     GALEX UV Spectroscopy and Deep Imaging of LIRGs in the ELAIS S1 field

    The ELAIS S1 field was observed by GALEX in both its Wide Spectroscopic and Deep Imaging Survey modes. This field was previously observed by the Infrared Space Observatory and we made use of the catalogue of multi-wavelength data published by the ELAIS consortium to select galaxies common to the two samples. Among the 959 objects with GALEX spectroscopy, 88 are present in the ELAIS catalog and 19 are galaxies with an optical spectroscopic redshift. The distribution of redshifts covers the range 0<z<1.60<z<1.6. The selected galaxies have bolometric IR luminosities 10<Log(LIR)<1310<Log(L_{IR})<13 (deduced from the 15μm15 \mu m flux using ISOCAM) which means that we cover a wide range of galaxies from normal to Ultra Luminous IR Galaxies. The mean (σ\sigma) UV luminosity (not corrected for extinction) amounts to Log(λ.L1530)=9.8(0.6)Log(\lambda.L_{1530}) = 9.8 (0.6) L_\sun for the low-z (z0.35z \le 0.35) sample. The UV slope β\beta (assuming fλλβf_\lambda \propto \lambda^\beta) correlates with the GALEX FUV-NUV color if the sample is restricted to galaxies below z<0.1z < 0.1. Taking advantage of the UV and IR data, we estimate the dust attenuation from the IR/UV ratio and compare it to the UV slope β\beta. We find that it is not possible to uniquely estimate the dust attenuation from β\beta for our sample of galaxies. These galaxies are highly extinguished with a median value AFUV=2.7±0.8A_{FUV} = 2.7 \pm 0.8. Once the dust correction applied, the UV- and IR-based SFRs correlate. For the closest galaxy with the best quality spectrum, we see a feature consistent with being produced by a bump near 220nm in the attenuation curve.Comment: This paper has been published as part of the GALEX ApJL Special Issue (ApJ 619, L63

    Lessons learned from the introduction of autonomous monitoring to the EUVE science operations center

    The University of California at Berkeley's (UCB) Center for Extreme Ultraviolet Astrophysics (CEA), in conjunction with NASA's Ames Research Center (ARC), has implemented an autonomous monitoring system in the Extreme Ultraviolet Explorer (EUVE) science operations center (ESOC). The implementation was driven by a need to reduce operations costs and has allowed the ESOC to move from continuous, three-shift, human-tended monitoring of the science payload to a one-shift operation in which the off shifts are monitored by an autonomous anomaly detection system. This system includes Eworks, an artificial intelligence (AI) payload telemetry monitoring package based on RTworks, and Epage, an automatic paging system to notify ESOC personnel of detected anomalies. In this age of shrinking NASA budgets, the lessons learned on the EUVE project are useful to other NASA missions looking for ways to reduce their operations budgets. The process of knowledge capture, from the payload controllers for implementation in an expert system, is directly applicable to any mission considering a transition to autonomous monitoring in their control center. The collaboration with ARC demonstrates how a project with limited programming resources can expand the breadth of its goals without incurring the high cost of hiring additional, dedicated programmers. This dispersal of expertise across NASA centers allows future missions to easily access experts for collaborative efforts of their own. Even the criterion used to choose an expert system has widespread impacts on the implementation, including the completion time and the final cost. In this paper we discuss, from inception to completion, the areas where our experiences in moving from three shifts to one shift may offer insights for other NASA missions

    Fault Induced Document Retrieval Officer (FIDO) is a computer program that reduces the need for a large and costly team of engineers and/or technicians to monitor the state of a spacecraft and associated ground systems and respond to anomalies. FIDO includes artificial-intelligence components that imitate the reasoning of human experts with reference to a knowledge base of rules that represent failure modes and to a database of engineering documentation. These components act together to give an unskilled operator instantaneous expert assistance and access to information that can enable resolution of most anomalies, without the need for highly paid experts. FIDO provides a system state summary (a configurable engineering summary) and documentation for diagnosis of a potentially failing component that might have caused a given error message or anomaly. FIDO also enables high-level browsing of documentation by use of an interface indexed to the particular error message. The collection of available documents includes information on operations and associated procedures, engineering problem reports, documentation of components, and engineering drawings. FIDO also affords a capability for combining information on the state of ground systems with detailed, hierarchically-organized, hypertext- enabled documentation
