    DIAGNOSIS OF GROUPS OF ELECTRO-RADIO COMPONENTS ACCORDING TO THE TRANSIENT RESPONSES FOR THE DETECTION OF DEFECTS ON THE FUNCTIONAL CELLS

    Objectives. A method for diagnosing electronic components with minimally complicated control and measuring instrumentation and the ability to obtain information from internal control points of operating units is developed. Methods. The developed method for control of groups of electronic components presupposes the passage of a controllable influence through a few control points, connected via several electronic components. In addition, each electronic component contributes its share to the transformation of the parameter output, so the discrepancy of its allowable values for the identification of defective electronic component are required to assess the implication of the expected value of the probable defect of each electronic component in the schema with the aim of isolating it. The faulty component in the electronic circuit is identified by the minimum estimated variance, by which the magnitude of the defect will match the expected value of the electronic component parameter. Results. As a result of the control no more than ten groups of electrical components on functional cell matrices can be assessed as having a defective element by comparing the calculated variance values for each electrical component and the corresponding electrical component parameter itself. Conclusion. Diagnosing groups of electric components on transient characteristics for the detection of defects in the functional cell matrices enables efficiency control operations improvement and troubleshooting in the search for modern electronic equipment reliability

    Quarantine nematode species and pathotypes potentially dangerous for domestic potato production: populations diversity and the genetics of potato resistance

    The review considers quarantine species and nematode pathotypes potentially dangerous for domestic potato production. Potatoes are affected by more than 30 types of parasitic nematodes, but the review focuses on the most harmful representatives of genera that cause great damage to potato production: Globodera, Ditylenchus, Nacob bus and Meloidogyne. Phytopathological and molecular methods of identification of species and pathotypes and the main achievements in studying the population variability of parasitic potato nematodes were analyzed. It was shown that due to the peculiarities of the life cycle of nematodes and lability of their genomes, the genetic variability of these organisms is very high, which creates a threat of forming new pathogenic genotypes of the parasites. The information about the intra- and interpopulation variability of nematodes is important for studying the ways of introduction and distribution of separate species, as well as for searching for the correlations of molecular markers with the pathotype. Phylogenetic studies based on modern data on genetic variability of populations have allowed to reveal species complexes in Globodera pallida (Stone) Behrens and Nacobbus aberrans (Thorne) Thorne & Allen (sensu lato), including cryptic species. The main components of successful protection preventing a wide distribution of parasitic nematodes are quarantine measures, agricultural techniques, biological methods of protection and cultivation of resistant cultivars. Special attention in the review is paid to the breeding of potato cultivars with durable resistance to various nematode pathotypes, because the cultivation of such varieties is the most ecologically safe and economically advantageous way to prevent epiphytoties. Currently, significant progress has been made in the genetic protection of potato cultivars, especially against cyst-forming nematodes. The review provides data on sources of potato resistance to parasitic nematodes identified in collections of wild and cultivated species. Data on identified R-gens and QTL of resistance that have been introduced into breeding varieties using different methods and approaches are analyzed. The literature data on the study of structural and functional organization of genes for resistance to potato cyst nematodes are given. The results of molecular research on revealing the polymorphisms of loci involved in the control of resistance to cyst and gall nematodes, the development of molecular markers of certain genes and their use in marker-assisted selection for developing of new resistant cultivars, including those with group resistance, are considered

    Potato resistance to quarantine diseases

    The casual agent of potato wart Synchytrium endobioticum (Schilb.) Perc. and potato golden nematode (PGN) Globodera rostochiensis (Wollenweber) Behrens are the quarantine species causing the most widespread and destructive diseases of potato in the Russian Federation and other countries of the world. The potato pale nematode Globodera pallida (Stone) Behrens is not found in Russia, although in the European Union it is found everywhere. The review provides information on the harmfulness of S. endobioticum and PGN. To date, 43 pathotypes of S. endobioticum and 5 pathotypes of PGN have been revealed in the world. In the Russian Federation, only the first (D1) pathotype of potato wart and pathotype Ro1 of PGN have been found. Modern sets of differentials for S. endobioticum and PGN and methods of pathotype composition determination, including efforts to develop molecular markers (SSR) to determine the race of S. endobioticum, are presented. Data on the resistance of commercial potato cultivars to these quarantine diseases and methods for resistance determination are reviewed. Modern data on the genetics of potato resistance to S. endobioticum, G. rostochiensis and G. pallida, including mapping and cloning of R-genes, are presented. Available literature data on molecular markers of R-genes for marker assisted selection and the evaluation of their effectiveness are presented. The use of multiplex systems allows the presence of several genes for resistance to one or more pathogens to be analyzed at once. Mechanisms of potato quantitative resistance to S. endobioticum and PGN and adaptation processes in pathogens populations resulting in overcoming resistance of host are discussed. Cultivation of cultivars poorly susceptible to PGN can stimulate the adaptive variability of the pathogen and induce virulent pathotypes for 2–3 pathogen generations

    Интродукционное изучение гиностеммы пятилистной (Gynostemma pentaphyllum (Thunb.) Makino) в условиях Западного Предкавказья

    Relevance. Due to the fact that gynostemma is an endemic plant, providing the pharmaceutical industry with domestic raw materials is not possible, this problem can be solved only through the introduction of this species into the culture.Material and methods. Studies on the introduction of Gynostemma pentaphyllum (Thunb.) Makino in the experimental fields of medicinal crop rotation of the North Caucasus branch of FGBNU VILAR, located in the zone of Western Ciscaucasia, were carried out to study the biomorphological and environmental features of growth and development of the plant, the most optimal conditions for growing and determination of economically valuable indicators. Propagation was carried out through seedlings from seeds received under the contract from South Korea, which were planted in the open ground in early May in areas with direct sunlight and under the canopy of trees (shade). Gynostemma pentaphyllum is a climbing liana with a creeping stem, so it was grown as a trellised crop.Results. It has been established that gynostemma under Krasnodar Territory conditions passes all stages of phenological development, starting from the first year of vegetation, grows and bears fruit every year. The duration of the growing season from 229 to 238 days was determined. Harvesting was carried out in the phase of budding – beginning of flowering. The most optimal condition for growing this crop is shading compared to the option of direct sunlight, where there was a significant increase in growth and development of gynostemma, which contributed to a significant increase in yield. On the rhizome of gynostemma II and III years of vegetation new additional shoots are formed, the number of which is significantly higher in the variant with shading. The conducted introduction studies indicate the possibility of cultivation of Gynostemma pentaphyllum (Thunb.) Makino in the zone of the Western Caucasus.Актуальность. В связи с тем, что гиностемма – эндемическое растение, обеспечение фармацевтической промышленности отечественным сырьем не представляется возможным, решить эту проблему можно только за счет введения данного вида в культуру.Материал и методика исследований. Проведены исследования по интродукции гиностеммы пятилистной (Gynostemma pentaphyllum (Thunb.) Makino) на опытных полях лекарственного севооборота Северо-Кавказского филиала ФГБНУ ВИЛАР, расположенных в зоне Западного Предкавказья, с целью изучения биоморфологических и экологических особенностей роста и развития растения, выявления наиболее оптимальных условий выращивания и определения хозяйственно ценных показателей. Размножение проводили через рассаду из семян, полученных по договору из Южной Кореи, которая высаживалась в открытый грунт в начале мая на участках с прямым солнечным освещением и под пологом деревьев (затенение). Гиностемма пятилистная – это вьющаяся лиана с ползучим стеблем, поэтому она выращивалась как шпалерная культура.Результаты. Установлено, что гиностемма в условиях Краснодарского края проходит все стадии фенологического развития, начиная с первого года вегетации, ежегодно растет и плодоносит. Определена продолжительность вегетационного периода от 229 до 238 суток. Уборку урожая проводили в фазу бутонизации - начала цветения. Наиболее оптимальным условием для выращивания данной культуры является затенение в сравнении с вариантом прямого солнечного освещения, где наблюдалось значительное усиление роста и развития гиностеммы, что способствовало значительному приросту урожайности. На корневище гиностеммы II и III года вегетации образуются новые дополнительные побеги, количество которых значительно выше в варианте при затенении. Проведенные интродукционные исследования свидетельствуют о возможности культивирования гиностеммы пятилистной в зоне Западного Предкавказья

    LKB1/KRAS mutant lung cancers constitute a genetic subset of NSCLC with increased sensitivity to MAPK and mTOR signalling inhibition

    LKB1/STK11 is a multitasking tumour suppressor kinase. Germline inactivating mutations of the gene are responsible for the Peutz-Jeghers hereditary cancer syndrome. It is also somatically inactivated in approximately 30% of non-small-cell lung cancer (NSCLC). Here, we report that LKB1/KRAS mutant NSCLC cell lines are sensitive to the MEK inhibitor CI-1040 shown by a dose-dependent reduction in proliferation rate, whereas LKB1 and KRAS mutations alone do not confer similar sensitivity. We show that this subset of NSCLC is also sensitised to the mTOR inhibitor rapamycin. Importantly, the data suggest that LKB1/KRAS mutant NSCLCs are a genetically and functionally distinct subset and further suggest that this subset of lung cancers might afford an opportunity for exploitation of anti-MAPK/mTOR-targeted therapies

    A statistical framework for assessing pharmacological responses and biomarkers using uncertainty estimates

    High-throughput testing of drugs across molecular-characterised cell lines can identify candidate treatments and discover biomarkers. However, the cells’ response to a drug is typically quantified by a summary statistic from a best-fit dose-response curve, whilst neglecting the uncertainty of the curve fit and the potential variability in the raw readouts. Here, we model the experimental variance using Gaussian Processes, and subsequently, leverage uncertainty estimates to identify associated biomarkers with a new Bayesian framework. Applied to in vitro screening data on 265 compounds across 1074 cancer cell lines, our models identified 24 clinically established drug-response biomarkers, and provided evidence for six novel biomarkers by accounting for association with low uncertainty. We validated our uncertainty estimates with an additional drug screen of 26 drugs, 10 cell lines with 8 to 9 replicates. Our method is applicable to any dose-response data without replicates, and improves biomarker discovery for precision medicine

    Epidemiological Features of Enterovirus Infection during Flood on the Territory of Jewish Autonomous Region

    Long-term manifestations of epidemiological process of enterovirus infection in the Jewish Autonomous Region were similar to those in the Khabarovsk Territory, though with a lower intensity. After establishment of emergency situation regime, in view of the rainfall flooding in 2013, enterovirus morbidity rate had been increasing within three weeks duration, then started to fall rapidly. Clustered cases of enterovirus infection were not registered. Viral serous meningitis ratio was insignificant as minor forms of the disease prevailed; coxsackie virus A-9 and echovirus-6 dominated. During the flood period isolated from samples of patients with minor forms of enteroviral disease were three enterovirus strains, type 71, sub-genotype C4a, possessing a high degree of genetic similarity to the Chinese ones, 2010-2011. All in all impact of the natural disaster on the epidemiological situation on enteroviral infection in the Jewish Autonomous Region turned to be insignificant
