Novel genetic sex markers reveal unexpected lack of, and similar susceptibility to, sex reversal in free-living common toads in both natural and anthropogenic habitats
Anthropogenic environmental changes are affecting biodiversity and microevolution worldwide. Ectothermic vertebrates are especially vulnerable because environmental changes can disrupt their sexual development and cause sex reversal, a mismatch between genetic and phenotypic sex. This can potentially lead to sex‐ratio distortion and population decline. Despite these implications, there is scarce empirical knowledge on the incidence of sex reversal in nature. Populations in anthropogenic environments may be exposed to sex‐reversing stimuli more frequently, which may lead to higher sex‐reversal rate or, alternatively, these populations may adapt to resist sex reversal. We developed PCR‐based genetic sex markers for the common toad (Bufo bufo) to assess the prevalence of sex reversal in wild populations living in natural, agricultural and urban habitats, and the susceptibility of the same populations to two ubiquitous oestrogenic pollutants in a common garden experiment. We found negligible sex‐reversal frequency in free‐living adults despite the presence of various endocrine‐disrupting pollutants in their breeding ponds. Individuals from different habitat types showed similar susceptibility to sex reversal in the laboratory: all genetic males developed female phenotype when exposed to 1 µg L(−1) 17α‐ethinylestradiol (EE2) during larval development, whereas no sex reversal occurred in response to 1 ng L(−1) EE2 and a glyphosate‐based herbicide with 3 µg L(−1) or 3 mg L(−1) glyphosate. The latter results do not support that populations in anthropogenic habitats would have either increased propensity for or higher tolerance to chemically induced sex reversal. Thus, the extremely low sex‐reversal frequency in wild toads compared to other ectothermic vertebrates studied before might indicate idiosyncratic, potentially species‐specific resistance to sex reversal

Absztrakt:
Bevezetés: A Parkinson-kór előrehaladásával jelentkező motoros,
illetve nem motoros komplikációk jelentősen rontják a Parkinson-kóros betegek
életminőségét, behatárolják a terápiás lehetőségeket. Nincs egyértelműen
meghatározva az „előrehaladott” Parkinson-kór, sem az invazív terápiák
bevezetésének optimális pillanata. Kevés irodalmi adat vonatkozik arra, hogy a
mindennapi klinikai gyakorlatban alkalmazott levodopaadagok felső határai hogyan
alakulnak előrehaladott Parkinson-kórban, illetve az egyes eszközös terápiák
elérhetősége hogyan befolyásolja ezeket. Célkitűzés: A
szubsztitúciós terápia elemzése előrehaladott Parkinson-kórban.
Módszer: Retrospektív tanulmányunkban 2011. június 1. és
2017. május 31. között minden olyan, Parkinson-kórral kezelt beteget
megvizsgáltunk, aki legalább napi 2 óra off állapotról számolt
be, dyskinesisekkel vagy ezek nélkül, és legalább napi négyszer kapott különböző
kombinációkban levodopát. Elemeztük a levodopaterápia sajátosságait az eszközös,
illetve a további konzervatív kezelésre javasolt betegcsoportoknál.
Eredmények: Előrehaladott Parkinson-kórt állapítottunk meg
311 betegnél, közülük 125-nél véltük indokoltnak valamilyen eszközös terápia
elkezdését, míg 42-nél a levodopa dózisának emelését. Az eszközös kezelésre
javasolt betegcsoport és a konzervatív kezelést folytató betegcsoport között
különbségek voltak a betegség fennállása, a levodopa átlagadagja és adagolási
frekvenciája tekintetében. A 107 levodopa-karbidopa intestinalis gélre tesztelt
betegnél az adagolási frekvencia és a levodopa-átlagadagok magasabbak voltak.
Következtetés: A vizsgált periódusban előrehaladott
Parkinson-kórban az irodalmi adatokhoz viszonyítva kisebb levodopaadagokat
találtunk, a kombináció adjuváns terápiával viszont gyakoribb. Előrehaladott
Parkinson-kórban érdemes az eszközös terápiát mérlegelni, ha a napi átlagos
levodopaadag legalább 750–1000 mg, minimum ötszöri adagolásra elosztva, és
kombinált terápia ellenére az életminőséget jelentősen csökkentő motoros
komplikációk jelentkeznek. Ezeket a betegeket erre szakosodott központokban
multidiszciplináris csapat közreműködésével kell tesztelni, és a legjobb döntést

Abstract:
Introduction: The motor and non-motor complications of
Parkinson’s disease impair the patients’ quality of life and limit therapeutical
options. There are no clear criteria for ‘advanced’ Parkinson’s disease or for
the optimal moment for invasive therapies. There is little evidence regarding
the upper limits of levodopa doses, and how these may be influenced by the
availability of device-aided therapies. Aim: To analyze
substitution therapy in patients with advanced Parkinson’s disease.
Method: In our retrospective study, we analyzed the data
from all patients with advanced Parkinson’s disease hospitalized between 1st
June 2011 and 31st May 2017, receiving combined levodopa treatment at least
4×/day, reporting a minimum of 2 hours off periods, with or
without dyskinesia. We analyzed levodopa therapy for patients who were
recommended either device-aided or conservative therapy.
Results: Out of 311 patients with advanced Parkinson’s
disease, for 125 we proposed device-aided therapies whereas in 42 patients we
increased the levodopa dose. The average levodopa doses and the administration
rate were higher for the 107 patients tested for levodopa-carbidopa intestinal
gel. Disease duration, mean levodopa doses and frequency of dosing were all
higher in patients proposed for device-aided therapies versus
patients with continued conservative treatment. Conclusion: Our
patients were on lower levodopa doses (compared to literature), but the
combinations were used more often. Device-aided therapies should be considered
in patients with severe motor complications who receive at least 750–1000 mg
levodopa daily, divided minimum 5×/day. These patients need to be tested in
specialized centers by multidisciplinary teams in order to make the best

Breeding <i>Polyommatus icarus</i> Serves as a Large-Scale and Environmentally Friendly Source of Precisely Tuned Photonic Nanoarchitectures
The colour of the butterfly wing serves as an important sexual and species-specific signal. Some species produce structural colouration by developing wing scales with photonic nanoarchitectures. These nanostructures are highly conservative, allowing only a ±10 nm peak wavelength deviation in the reflectance spectra of the blue structural colour in natural Common Blue (Polyommatus icarus) populations. They are promising templates of future artificial photonic materials and can be used in potential applications, too. In this work, we present methodology and infrastructure for breeding laboratory populations of Common Blue as a cost-effective and environmentally friendly source of nanostructures. Our technology enables the production of approximately 7500 wing samples, equivalent to 0.5–1 m2 of photonic nanoarchitecture surface within a year in a single custom-made insectarium. To ascertain the reliability of this method, we compared reflectance properties between different populations from distant geographic locations. We also provide genetic background of these populations using microsatellite genotyping. The laboratory population showed genetic erosion, but even after four generations of inbreeding, only minimal shifts in the structural colouration were observed, indicating that wild Common Blue populations may be a reliable source of raw material for photonic surfaces
