Prediction and Measurement of the local extinction coefficient in sprays for 3D simulation/experiment data comparison
AbstractIn the recent years, large progresses in laser imaging techniques have allowed to extract spatially resolved 2D and 3D quantitative spray information even in optically dense situations. The main breakthrough of these techniques is the possibility of suppressing unwanted effects from multiple light scattering using Structured Illumination. Thanks to this new feature, effects due to light extinction can also be corrected allowing the measurement of the local extinction coefficient. These quantitative information which is available even in challenging conditions, where Phase Doppler does not work anymore, can be used for data comparison between experiment and simulation. The local extinction coefficient is particularly valuable for the description of the droplet field, defined as the âspray regionâ, as it contains information related to both droplets size and concentration. In this article we detail, then, the procedure enabling the modelers to obtain numerically this local extinction coefficient over the full 3D spray system. Following this procedure, results can now be adequately compared between simulation and experiment. The proposed comparison approach can better guide model adjustments in situation where the initial droplet size distribution is unknown or approximated and presents a step towards future validations of spray simulations, especially those based on Lagrangian Particle Tracking. The approach is exemplified here for the case of a Diesel-type spray. The results reveal at which specific spray locations discrepancies occur, and highlight the sensitivity of the initial droplet size distribution on the resulting extinction coefficient
Sensitivity of VOF simulations of the liquid jet breakup to physical and numerical parameters
AbstractIn this paper the characteristics of the primary breakup of a liquid jet is analyzed numerically. We applied the Volumes of Fluids (VOF) approach utilizing the Direction Averaged Curvature (DAC) model, to estimate the interface curvature, and the Direction Averaged Normal (DAN) model, to propagate the interface. While being used for the first time to predict liquid atomization, this methodology showed a high accuracy. The influence of varying the fluid properties, namely liquid-gas density and viscosity ratio, and injection conditions is discussed related to the required grid resolution. Resulting droplet sizes are compared to distributions obtained through the One-Dimensional Turbulence (ODT) model
Opposite prognostic roles of HIF1alpha and HIF2alpha expressions in bone metastatic clear cell renal cell cancer
BACKGROUND: Prognostic markers of bone metastatic clear cell renal cell cancer (ccRCC) are poorly established. We tested prognostic value of HIF1alpha/HIF2alpha and their selected target genes in primary tumors and corresponding bone metastases. RESULTS: Expression of HIF2alpha was lower in mRCC both at mRNA and protein levels (p/mRNA/=0.011, p/protein/=0.001) while HIF1alpha was similar to nmRCC. At the protein level, CAIX, GAPDH and GLUT1 were increased in mRCC. In all primary RCCs, low HIF2alpha and high HIF1alpha as well as CAIX, GAPDH and GLUT1 expressions correlated with adverse prognosis, while VEGFR2 and EPOR gene expressions were associated with favorable prognosis. Multivariate analysis confirmed high HIF2alpha protein expression as an independent risk factor. Prognostic validation of HIFs, LDH, EPOR and VEGFR2 in RNA-Seq data confirmed higher HIF1alpha gene expression in primary RCC as an adverse (p=0.07), whereas higher HIF2alpha and VEGFR2 expressions as favorable prognostic factors. HIF1alpha/HIF2alpha-index (HIF-index) proved to be an independent prognostic factor in both the discovery and the TCGA cohort. PATIENTS AND METHODS: Expressions of HIF1alpha and HIF2alpha as well as their 7 target genes were analysed on the mRNA and protein level in 59 non-metastatic ccRCCs (nmRCC), 40 bone metastatic primary ccRCCs (mRCC) and 55 corresponding bone metastases. Results were validated in 399 ccRCCs from the TCGA project. CONCLUSIONS: We identified HIF2alpha protein as an independent marker of the metastatic potential of ccRCC, however, unlike HIF1alpha, increased HIF2alpha expression is a favorable prognostic factor. The HIF-index incorporated these two markers into a strong prognostic biomarker of ccRCC
Genome Wide Transcriptome Analysis of Dendritic Cells Identifies Genes with Altered Expression in Psoriasis
Activation of dendritic cells by different pathogens induces the secretion of proinflammatory mediators resulting in
local inflammation. Importantly, innate immunity must be properly controlled, as its continuous activation leads to the
development of chronic inflammatory diseases such as psoriasis. Lipopolysaccharide (LPS) or peptidoglycan (PGN)
induced tolerance, a phenomenon of transient unresponsiveness of cells to repeated or prolonged stimulation,
proved valuable model for the study of chronic inflammation. Thus, the aim of this study was the identification of the
transcriptional diversity of primary human immature dendritic cells (iDCs) upon PGN induced tolerance. Using SAGESeq
approach, a tag-based transcriptome sequencing method, we investigated gene expression changes of primary
human iDCs upon stimulation or restimulation with Staphylococcus aureus derived PGN, a widely used TLR2 ligand.
Based on the expression pattern of the altered genes, we identified non-tolerizeable and tolerizeable genes. Gene
Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (Kegg) analysis showed marked enrichment of
immune-, cell cycle- and apoptosis related genes. In parallel to the marked induction of proinflammatory mediators,
negative feedback regulators of innate immunity, such as TNFAIP3, TNFAIP8, Tyro3 and Mer are markedly
downregulated in tolerant cells. We also demonstrate, that the expression pattern of TNFAIP3 and TNFAIP8 is
altered in both lesional, and non-lesional skin of psoriatic patients. Finally, we show that pretreatment of immature
dendritic cells with anti-TNF-α inhibits the expression of IL-6 and CCL1 in tolerant iDCs and partially releases the
suppression of TNFAIP8. Our findings suggest that after PGN stimulation/restimulation the host cell utilizes different
mechanisms in order to maintain critical balance between inflammation and tolerance. Importantly, the transcriptome
sequencing of stimulated/restimulated iDCs identified numerous genes with altered expression to date not associated
with role in chronic inflammation, underlying the relevance of our in vitro model for further characterization of IFNprimed
iDCs
Science with a small two-band UV-photometry mission II: Observations of stars and stellar systems
We outline the impact of a small two-band UV-photometry satellite mission on
the field of stellar physics, magnetospheres of stars, binaries, stellar
clusters, interstellar matter, and exoplanets. On specific examples of
different types of stars and stellar systems, we discuss particular
requirements for such satellite missions in terms of specific mission
parameters such as bandpass, precision, cadence, and mission duration. We show
that such a mission may provide crucial data not only for hot stars that emit
most of their light in UV, but also for cool stars, where UV traces their
activity. This is important, for instance, for exoplanetary studies, because
the level of stellar activity influences habitability. While the main asset of
the two-band UV mission rests in time-domain astronomy, an example of open
clusters proves that such a mission would be important also for the study of
stellar populations. Properties of the interstellar dust are best explored when
combining optical and IR information with observations in UV. It is well known
that dust absorbs UV radiation efficiently. Consequently, we outline how such a
UV mission can be used to detect eclipses of sufficiently hot stars by various
dusty objects and study disks, rings, clouds, disintegrating exoplanets or
exoasteroids. Furthermore, UV radiation can be used to study the cooling of
neutron stars providing information about the extreme states of matter in the
interiors of neutron stars and used for mapping heated spots on their surfaces.Comment: Submitted to Space Science Review
Quick Ultra-VIolet Kilonova surveyor (QUVIK)
We present a near-UV space telescope on a ~70kg micro-satellite with a
moderately fast repointing capability and a near real-time alert communication
system that has been proposed in response to a call for an ambitious Czech
national mission. The mission, which has recently been approved for Phase 0, A,
and B1 study shall measure the brightness evolution of kilonovae, resulting
from mergers of neutron stars in the near-UV band and thus it shall distinguish
between different explosion scenarios. Between the observations of transient
sources, the satellite shall perform observations of other targets of interest,

Upravljanje otporno na kvarove modularnim prekidaÄko-reluktantnim strojem nadahnuto prirodom
Fault tolerance is an obligatory feature in safety critical applications (aeronautical, aerospace, medical and military applications, power plants, etc.), where loss of life, environmental disasters, equipment destructions or unplanned downtimes must be avoided. For such applications, a novel bio-inspired motion control system is proposed. All its three components (the switched reluctance machine, the power converter and the control system) are designed to be as fault tolerant as possible. This paper describes all these three fault tolerant components: the bio-inspired control system having self-healing capabilities, the power converter with an extra leg and the fault tolerant modular machine. The theoretical expectations and simulation results are validated by means of laboratory experiments.Otpornost na kvarove je nuĆŸnost u sigurnosno kritiÄnim aplikacijama (aeronautiÄke, zrakoplovne, medicinske i vojne aplikacije, elektrane itd.), gdje je potrebno izbjeÄi smrtne sluÄajeve, prirodne nepogode, uniĆĄtenje opreme ili neplanirane prekide u radu. Za takve aplikacije, predloĆŸen je novi slijedni sustav nadahnut prirodom. Sve tri komponente (prekidaÄko-reluktantni stroj, pretvaraÄ i sustav upravljanja) su projektirani da budu ĆĄto je viĆĄe moguÄe otporni na kvarove. Ovaj rad opisuje sve tri komponente: sustav upravljanja nadahnut prirodom sa samolijeÄeÄim svojstvima, pretvaraÄ s dodatnom granom i modularni stroj otporan na kvarove.
