    Comparative transcriptome analysis reveals resistance-related genes and pathways in Musa acuminata banana 'Guijiao 9' in response to Fusarium wilt.

    Fusarium wilt caused by Fusarium oxysporum f. sp. cubense (Foc), is one of the most devastating diseases in bananas resulting in significant loss of Cavendish bananas production worldwide. Here we show the agronomic traits and the resistance of 'Guijiao 9' in the field trials from 2012 to 2017. And then we dissect and compare the transcriptome response from these two cultivars (cv. 'Guijiao 9' and cv. Williams) in an attempt to understand the molecular basis that contribute to the enhanced Foc tropical race 4 (Foc-TR4) resistance. 'Guijiao 9' is a Cavendish cultivar with strong resistance to Foc-TR4, which was reflected in a lower disease severity and incidence in glasshouse and field trails, when compared to the susceptible cultivar Williams. Gene expression profiles of 'Guijiao 9' and Williams were captured by performing RNA-Seq analysis on 16 biological samples collected over a six day period post inoculation with Foc-TR4. Transcriptional reprogramming in response to Foc-TR4 was detected in both genotypes but the response was more drastic in 'Guijiao 9' than in Williams. Specific genes involved in plant-pathogen interaction and defense signaling including MAPK, calcium, salicylic acid, jasmonic acid and ethylene pathways were analyzed and compared between 'Guijiao 9' and Williams. Genes associated with defense-related metabolites synthesis such as NB-LRR proteins, calmodulin-binding protein and phenylpropanoids biosynthesis genes were significantly up-regulated in 'Guijiao 9' resistant to Foc-TR4 infection. Taken together, this study highlights the important roles of plant hormone regulation and defense gene activation in mediating resistance in 'Guijiao 9'

    Molecular detection of Cylindrocarpon destructans in infected Chinese ginseng roots and soil

    Ginseng (<i>Panax ginseng</i> C.A. Meyer) is one of the most important medicinal plants in China, but its yields are often reduced by a variety of root pathogens. The root rot of ginseng is a destructive soil-borne disease caused by Cylindrocarpon destructans (teleomorph: Neonectria radicicola). A species-specific polymerase chain reaction (PCR) assay was developed for rapid detection of C. destructans in diseased ginseng roots and artificially inoculated soil. One pair of specific primers was designed from comparisons of nucleotide sequences of the nuclear ribosomal internal transcribed spacer (ITS) regions of 22 fungal isolates from northeast of China. Under stringent PCR conditions, primers CD-F and CD-R amplified only a 450 bp fragment from C. destructans DNA but not from other pathogens or negative control. The sensitivity of detection was 1 pg genomic DNA per 25 ìl PCR reaction volume, and C. destructans could be specifically detected with CD-F/CD-R from infected ginseng roots and soil. The approach outlined here could be further utilized as a rapid and reliable tool for the diagnosis and monitoring of the root rot of ginseng.Key words: Panax ginseng, Cylindrocarpon destructans, internal transcribed spacer (ITS), polymerase chain reaction (PCR), molecular detection

    A Slot Is Not Built in One Utterance: Spoken Language Dialogs with Sub-Slots

    A slot value might be provided segment by segment over multiple-turn interactions in a dialog, especially for some important information such as phone numbers and names. It is a common phenomenon in daily life, but little attention has been paid to it in previous work. To fill the gap, this paper defines a new task named Sub-Slot based Task-Oriented Dialog (SSTOD) and builds a Chinese dialog dataset SSD for boosting research on SSTOD. The dataset includes a total of 40K dialogs and 500K utterances from four different domains: Chinese names, phone numbers, ID numbers and license plate numbers. The data is well annotated with sub-slot values, slot values, dialog states and actions. We find some new linguistic phenomena and interactive manners in SSTOD which raise critical challenges of building dialog agents for the task. We test three state-of-the-art dialog models on SSTOD and find they cannot handle the task well on any of the four domains. We also investigate an improved model by involving slot knowledge in a plug-in manner. More work should be done to meet the new challenges raised from SSTOD which widely exists in real-life applications.     Curcumin Delays Retinal Degeneration by Regulating Microglia Activation in the Retina of rd1 Mice

    Background/Aims: Retinitis pigmentosa (RP) is characterized by degeneration of photoreceptors, and there are currently no effective treatments for this disease. However, curcumin has shown neuroprotectant efficacy in a RP rat and swine model, and thus, may have neuroprotective effects in this disease. Methods: Immunofluorescence staining, electroretinogram recordings, and behavioral tests were used to analyze the effects of curcumin and the underlying mechanism in retinal degeneration 1 (rd1) mice. Results: The number of apoptotic cells in the retina of rd1 mice at postnatal day 14 significantly decreased with curcumin treatment and visual function was improved. The activation of microglia and secretion of chemokines and matrix metalloproteinases in the retina were inhibited by curcumin. These effects were also observed in a co-culture of BV2 microglial cells and retina-derived 661W cells. Conclusions: Curcumin delayed retinal degeneration by suppressing microglia activation in the retina of rd1 mice. Thus, it may be an effective treatment for neurodegenerative disorders such as RP

    Assessing variations in host resistance to Fusarium oxysporum f sp. cubense race 4 in Musa species, with a focus on the subtropical race 4

    Fusarium oxysporum f. sp. cubense (Foc) has severely curtailed banana production in the tropical regions of the world. The tropical race 4 (TR4) of Foc was detected in Australia in the 1990s and it is virulent to all Cavendish type banana cultivars, which represents the majority of banana production in Australia. Genetic resistance to Foc race 4 is urgently needed. To characterize sources of resistance, we have assessed the Foc resistance response of 34 Musa cultivars with plants grown under controlled settings. Amongst diploid banana cultivars carrying the AA genome, resistance is found in Musa acuminate sub-species including malaccensis 'Pahang' and burrnannica 'Calcutta4' In the polyploid group, the hybrids such as FHIA-18' and FHIA-25' are highly resistant against both Foc-TR4 and subtropical race 4 (Foc-STR4). Interestingly, FHIA-2' and 'CAM020' appear to be resistant to Foc-TR4 but susceptible to Foc-STR4, suggesting potential differences in the resistance mechanisms against the different race 4 strains. Using a GFP tagged Foc-STR4 strain challenged onto both resistant and susceptible M. a. malaccensis lines, a high inoculum dosage rapidly induced vascular wilt in the susceptible M. a. malaccensis lines at 2.5 weeks. This was associated with an accumulation of micro-conidia in the rhizome and the movement of the fungus through the xylem vessels. In contrast, the fungal movement was restrained in the rhizome of the resistant M. a. malaccensis lines and no sporulation was observed. Overall, this research suggests that the resistance response is dependent to an extent on inoculum dosage and that the plant host's response, in the rhizome, plays an important role in inhibiting the fungus from spreading to the rest of the plant. Identifying race 4 resistant accessions can help to understand mechanisms of resistance and provide banana breeders with the genetic resources to integrate resistance genes into commercial varieties

    Nutrition strategies to control post-weaning diarrhea of piglets: From the perspective of feeds

    Post-weaning diarrhea (PWD) is a globally significant threat to the swine industry. Historically, antibiotics as well as high doses of zinc oxide and copper sulfate have been commonly used to control PWD. However, the development of bacterial resistance and environmental pollution have created an interest in alternative strategies. In recent years, the research surrounding these alternative strategies and the mechanisms of piglet diarrhea has been continually updated. Mechanically, diarrhea in piglets is a result of an imbalance in intestinal fluid and electrolyte absorption and secretion. In general, enterotoxigenic Escherichia coli (ETEC) and diarrheal viruses are known to cause an imbalance in the absorption and secretion of intestinal fluids and electrolytes in piglets, resulting in diarrhea when Cl− secretion-driven fluid secretion surpasses absorptive capacity. From a perspective of feedstuffs, factors that contribute to imbalances in fluid absorption and secretion in the intestines of weaned piglets include high levels of crude protein (CP), stimulation by certain antigenic proteins, high acid-binding capacity (ABC), and contamination with deoxynivalenol (DON) in the diet. In response, efforts to reduce CP levels in diets, select feedstuffs with lower ABC values, and process feedstuffs using physical, chemical, and biological approaches are important strategies for alleviating PWD in piglets. Additionally, the diet supplementation with additives such as vitamins and natural products can also play a role in reducing the diarrhea incidence in weaned piglets. Here, we examine the mechanisms of absorption and secretion of intestinal fluids and electrolytes in piglets, summarize nutritional strategies to control PWD in piglets from the perspective of feeds, and provide new insights towards future research directions
