    Malignant Gliomas: A Case Study

    Malignant gliomas, of grade III and grade IV malignancy, are incurable neoplasms that arise from cells with several well-characterized genetic profile abnormalities that cause uncontrollable growth and infiltration in the brain. Presenting symptoms of both generalized and focal neurological abnormalities are induced by increased intracranial pressure and focal neuronal dysfunction, respectively. On average, patients experience 3 months or less of clinical history before receiving diagnosis based on multifactorial comparison of clinical and pathological presentation of the tumor. Following diagnosis, maximal safe resection and adjuvant radiotherapy and concurrent chemotherapy typically ensues with subsequent management chemotherapy regimens. Despite aggressive treatment approaches, progression or recurrence is highly typical based on 5-yr survival rates of 5.1% and 27.9% of grade IV glioblastoma multiforme (GBM) and grade III anaplastic astrocytoma (AA), respectively, the two most common malignant gliomas. Severely progressive clinical and functional deterioration in the terminal stage of care may warrant cessation of curative care replaced with maximal palliative care. Brain tumor patients experience the burden of terminal illness as other cancer patients do, but with added neurological-specific impairments that reduce quality of life. Possible causes of death include herniation, tumor progression, and systemic illness, but can be potentially multifactorial. The following manuscript characterizes the pathological mechanisms of oncogenesis and growth, followed by a comprehensive review of the clinical care for brain tumor patients from symptom onset to cause of death. To aid in the clinical applicability of these concepts, a case study of a single patient “WL”, who received a diagnosis of grade III anaplastic astrocytoma following 3 months of visual deterioration, will prompt the clinical review by illustration of disease course and treatment

    Conservation and concealment in SpeciesBanking.com, USA: an analysis of neoliberal performance in the species offsetting industry

    Market-based strategies are promoted as neoliberal governance solutions to environmental problems, from local to global scales. Tradable mitigation schemes are proliferating. These include species banking, which enables payments for the purchase of species credits awarded to conserved areas to offset development impacts on protected species elsewhere. An analysis of species banks in the USA through a survey of data from the website www.SpeciesBanking.com (established as a ‘clearing house’ for species banking information) was complemented by questionnaire material from USA bank managers. The number of USA species banks has increased rapidly, bank area ownership and management is consolidated in a small number of organizations, and public information on species credit price is limited. In interrogating the case material, the roles of specific economic policies associated with neoliberalism are considered, focusing on the extension of privatization, de- and re-regulation and marketization into the arena of environmental conservation, and commodification processes as manifested in species banking. Problematic ecological and distributive ‘concealments’ in species banking include the ‘development-led’ nature of conservation banking, tendencies towards net biodiversity loss, and an emphasis on supporting conservation-related wealth accumulation by larger landowners and investors

    Online communication and dating relationships: Effects of decreasing online communication on feelings of closeness and relationship satisfaction

    The purpose of this study was to examine the effects of online communication on feelings of closeness and relationship satisfaction using an experimental design and to test whether these effects varied based on participants’ attitudes about online communication. Individuals in dating relationships were randomly assigned to two 48-hr conditions: communication as usual or refraining from online communication. Participants who reported that online communication was important for their dating relationships reported lower satisfaction and closeness after decreasing their online communication; there were no between-group differences among participants who reported relatively low importance. Thus, it appears that online communication has a positive effect on relationship satisfaction and feelings of closeness, but only for those who consider online communication to be important

    Metabolic reprogramming by viruses in the sunlit and dark ocean

    BACKGROUND:Marine ecosystem function is largely determined by matter and energy transformations mediated by microbial community interaction networks. Viral infection modulates network properties through mortality, gene transfer and metabolic reprogramming.RESULTS:Here we explore the nature and extent of viral metabolic reprogramming throughout the Pacific Ocean depth continuum. We describe 35 marine viral gene families with potential to reprogram metabolic flux through central metabolic pathways recovered from Pacific Ocean waters. Four of these families have been previously reported but 31 are novel. These known and new carbon pathway auxiliary metabolic genes were recovered from a total of 22 viral metagenomes in which viral auxiliary metabolic genes were differentiated from low-level cellular DNA inputs based on small subunit ribosomal RNA gene content, taxonomy, fragment recruitment and genomic context information. Auxiliary metabolic gene distribution patterns reveal that marine viruses target overlapping, but relatively distinct pathways in sunlit and dark ocean waters to redirect host carbon flux towards energy production and viral genome replication under low nutrient, niche-differentiated conditions throughout the depth continuum.CONCLUSIONS:Given half of ocean microbes are infected by viruses at any given time, these findings of broad viral metabolic reprogramming suggest the need for renewed consideration of viruses in global ocean carbon models.This item is part of the UA Faculty Publications collection. In surface ocean waters, viruses are known to act as gene vectors among susceptible hosts. Some of these genes may have metabolic functions and are thus termed auxiliary metabolic genes (AMGs). AMGs introduced to new hosts by viruses can enhance viral replication and/or potentially affect biogeochemical cycles by modulating key microbial pathways. Here we identify 748 viral populations that cluster into 94 genera along a vertical geochemical gradient in the Cariaco Basin, a permanently stratified and euxinic ocean basin. The viral communities in this ODWC appear to be relatively novel as 80 of these viral genera contained no reference viral sequences, likely due to the isolation and unique features of this system. We identify viral elements that encode AMGs implicated in distinctive processes, such as sulfur cycling, acetate fermentation, signal transduction, [Fe–S] formation, and N-glycosylation. These AMG-encoding viruses include two putative Mu-like viruses, and viral-like regions that may constitute degraded prophages that have been modified by transposable elements.     Interrogating marine virus-host interactions and elemental transfer with BONCAT and nanoSIMS-based methods

    While the collective impact of marine viruses has become more apparent over the last decade, a deeper understanding of virus-host dynamics and the role of viruses in nutrient cycling would benefit from direct observations at the single-virus level. We describe two new complementary approaches - stable isotope probing coupled with nanoscale secondary ion mass spectrometry (nanoSIMS) and fluorescence-based biorthogonal non-canonical amino acid tagging (BONCAT) - for studying the activity and biogeochemical influence of marine viruses. These tools were developed and tested using several ecologically relevant model systems (Emiliania huxleyi/EhV207, Synechococcus sp. WH8101/Syn1, and Escherichia coli/T7). By resolving carbon and nitrogen enrichment in viral particles, we demonstrate the power of nanoSIMS tracer experiments in obtaining quantitative estimates for the total number of viruses produced directly from a particular production pathway (by isotopically labeling host substrates). Additionally, we show through laboratory experiments and a pilot field study that BONCAT can be used to directly quantify viral production (via epifluorescence microscopy) with minor sample manipulation and no dependency on conversion factors. This technique can also be used to detect newly synthesized viral proteins. Together these tools will help fill critical gaps in our understanding of the biogeochemical impact of viruses in the ocean
