    Mitigation Training in Preparation of Face-to-face Learning in Schools under Health Reviews during Pandemic

    The Covid-19 pandemic has hit the whole world, including Indonesia. This pandemic has greatly impacted all sectors, including education. Online learning process has many obstacles and brings serious impacts. As a result, the policy of implementing face-to-face learning during pandemic was issued. Mitigation training is very important. This training attempts to discuss face-to-face learning at schools and obtain representative opinions regarding the preparation and consideration based on health reviews in relevant with the latest research and recommendations. This online training was held with the participants involving teachers and parents (n=219) from Elementary and Junior High School of Al-azhar SummareconSerpong, and has been shared on YouTube platform. Several opinions were obtained from parents who disagree with face-to-face learning’s idea. It also increased the knowledge regarding mitigation of face-to-face learning at schools. The education aspect is indeed something that must become a priority, but without compromising the student’s health aspect.Pandemic covid-19 telah melanda dunia termasuk Indonesia. Pandemic ini sangat berdampak keseluruh sector, termasuk Pendidikan. menurut hasil evaluasi oleh kementrian Pendidikan dan kebudayaan Indonesia, Proses pembelajaran daring yang berlangsung selama pandemi mengalami banyak hambatan dan memberikan dampak serius. Sehingga muncul kebijakan pemberlakuan sekolah tatap muka disaat pandemic. Pelatihan mitigasi untuk mempersiapkannya pun menjadi hal yang sangat penting. pelatihan ini bertujuan untuk membahas rencana tatap muka disekolah serta memperoleh pendapat yang representative perihal persiapan dan menjelakan tinjauan kesehatan berdasarkan riset dan rekomendasi terbaru. Pelatihan ini diadakan secara online dengan peserta guru, orang tua (n=219) dari Sekolah dasar dan sekolah menengah pertama al azhar summarecon serpong tangerang selatan dan telah di share di platform youtube. Dari pelatihan tersebut di dapat beberapa pendapat orang tua yang belum menyetujui adanya pembelajaran tatap muka, serta penambahan pemahaman sekolah dan orang tua terkait mitigasi sekolah tatap muka di sekolah. Aspek Pendidikan memang hal yang harus menjadi prioritas namun tanpa mengesampingkan aspek kesehatan bagi anak-anak

    Sedative Effects of Intraperitoneal Diazepam in Mice

    Objectives: To determine the effectiveness of Diazepam in comparison with Phenobarbital. Methods: Twenty-seven male Swiss Webster mice were used and randomly divided into three groups of negative control (NS), positive control (Phenobarbital), and diazepam group. Two tests were performed on these group: the traction Test and the Fireplace Test. Pupillary diameter was also observed. Results: A significant difference based on the Kruskal - Wallis statistical test was observed between the positive control and the diazepam group (0.05). Conclusion: Diazepam has a better sedative effect than Phenobarbital. The sedative effect produced by Diazepam is stronger, with faster onset and longer half-life than the Phenobarbital the positive control. However, different test methods and comparisons should be sought to support this conclusion

    The Effect of Increased Glucose Induction on GSH Levels in Insulin Gaussia Luciferase (iGL) Cells Derived from Rat Pancreatic Beta Cells

    Background: Prolonged hyperglycaemia can make the pancreatic beta cells work harder and cause fatigue. When this happens, it can trigger oxidative stress reactions, which can produce free radical compounds that can damage pancreatic beta cells. The body compensates by activating protective mechanisms such as the production of antioxidant compounds to reduce the levels of free radicals in the cells. One such compound is glutathione (GSH). Insulin Gaussia Luciferase (iGL) cells are a cell line derived from rat pancreatic β (beta) cells. These cells can be used as a model of oxidative stress in hyperglycaemia to measure GSH levels and there are no studies using iGL cells to measure GSH levels. Therefore, in this study, the iGL cells are used as the object of research.Objective: To investigate the effect of GSH levels on glucose toxicity condition through in vitro experiments on iGL cells.Methods: The study used 5 different glucose concentrations of 11, 16.5, 22, 33, and 44 mM with the addition of iGL cell growth medium exposed for 7 days. We measured the amount of intracellular GSH using a colourimetric method at a wavelength of 405 nm. The analysis used in this study was a one-way ANOVA test. Differences between groups were tested using SPSS.Results: The results of this study showed that there was an increasing trend in total GSH levels on the third and seventh day.Conclusion: there was a daily decrease in GSH/cell in the iGL sample cells exposed to different concentrations of glucose for 7 days. This can be due to an increase in the oxidative stress reactions in the cells, which can lead to a decrease in the levels of antioxidants

    Rev-erb agonist improves adverse cardiac remodeling and survival in myocardial infarction through an anti-inflammatory mechanism

    Rev-erb α, known as nuclear receptor 1D1 (NR1D1), regulates circadian rhythm, modulates glucose and lipid metabolism, and inflammatory response. However, little is known about the effect of Rev-erb agonist on the progression of myocardial infarction (MI) and heart failure. To investigate it, wild-type male mice underwent sham-operation or permanent ligation of the left anterior descending coronary artery to create MI model. Rev-erb agonist SR9009 (100 mg/kg/day) or vehicle was intraperitoneally administered. Echocardiography was performed to evaluate cardiac function 1 week after surgery. The gene and protein expression levels in the left ventricles (LVs) were determined with real-time PCR, western blotting, and immunofluorescence. Moreover, immune cell infiltration into the LVs was analyzed by flow cytometry. Survival rate and reduced LV function were significantly improved by the treatment with SR9009 after MI. The expression level and plasma concentration of brain natriuretic peptide were significantly lower in MI mice treated with SR9009 (MI+SR) than in MI mice treated with vehicle (MI+V). Moreover, the mRNA expression levels of inflammatory-related molecules such as Il6, Mcp1, Ly6g, Cd11b, matrix metallopeptidase (Mmp)9, and the protein expression levels of phosphorylated NF-κB p65, phosphorylated ERK, and phosphorylated p38 were also significantly lower in MI+SR than in MI+V. Immunofluorescence intensity for MMP-9 was enhanced in the LVs, but was less so in MI+SR than in MI+V. Furthermore, infiltrations of neutrophils and proinflammatory macrophages in the LVs were dramatically increased in MI+V and were significantly suppressed in MI+SR. Rev-erb agonist SR9009 treatment inhibited post-MI mortality and improved cardiac function through modulating inflammation and remodeling process
