    Modulation of Phenol Oxidation in Cofacial Dyads

    The presentation of two phenols on a xanthene backbone is akin to the tyrosine dyad (Y[subscript 730] and Y[subscript 731]) of ribonucleotide reductase. X-ray crystallography reveals that the two phenol moieties are cofacially disposed at 4.35 Å. Cyclic voltammetry reveals that phenol oxidation is modulated within the dyad, which exhibits a splitting of one-electron waves with the second oxidation of the phenol dyad occurring at larger positive potential than that of a typical phenol. In contrast, a single phenol appended to a xanthene exhibits a two-electron process, consistent with reported oxidation pathways of phenols in acetonitrile.     Inactivation of pollen and other effects of genome-plastome incompatibility in Oenothera

    A series of strains of the homozygous species Oenothera grandiflora (characterized by the genome BB and plastome III) were combined with plastome IV from O. parviflora (BC-IV) by means of appropriate crosses. An incompatibility between genome B and plastome IV is expressed in the haplo- and diplophase: (1) B-IV pollen, though normally developed, is largely inactive. The extent of the inactivation varies between different strains and shows a seasonal fluctuation as determined by seed set in outcrossing and selfing experiments. (2) In most of the strains lethality of BB-IV embryos is the rule, leading to empty seeds. This can be ameliorated by including another plastome in the zygotes and developing embryos on account of the biparental plastid transmission in Oenothera. It can best be demonstrated in crosses with a seed parent having normal green plastids of plastome IV and mutated chlorophyll deficient plastids from a different plastome in the pollen parent, leading to variegated progeny as well as a remainder of empty seeds. (3) In about one-half of the strains the BB-IV plants exhibit a temporary bleaching of the virescens type. The incompatibily between genome B and plastome IV does not support the earlier assumption that plastome IV is the ancestor of plastomes II, III, and V. Instead, a precursor plastome is postulated from which plastomes II, III, and IV are descended.     The class III ribonucleotide reductase from Neisseria bacilliformis can utilize thioredoxin as a reductant

    The class III anaerobic ribonucleotide reductases (RNRs) studied to date couple the reduction of ribonucleotides to deoxynucleotides with the oxidation of formate to CO[subscript 2]. Here we report the cloning and heterologous expression of the Neisseria bacilliformis class III RNR and show that it can catalyze nucleotide reduction using the ubiquitous thioredoxin/thioredoxin reductase/NADPH system. We present a structural model based on a crystal structure of the homologous Thermotoga maritima class III RNR, showing its architecture and the position of conserved residues in the active site. Phylogenetic studies suggest that this form of class III RNR is present in bacteria and archaea that carry out diverse types of anaerobic metabolism.Singapore.     Detection of carpal tunnel syndrome by infrared thermography

    Abstract The aim of this study is to approach the possibilities of stimulated infrared thermography to the carpal tunnel syndrome diagnosis. Eleven patients were studied. Six of them were affected by the carpal tunnel syndrome. The five others were healthy. The study consists in exciting the patients' hands and in analyzing their thermal response. The infrared thermograms obtained show that the hands infected by the carpal tunnel syndrome give a different thermal response

    Estimating non-response bias in family studies: application to mental health and lifestyle.

    Non-response to mailed surveys reduces the effective sample size and may introduce bias. Non-response has been studied by (1) comparison to available data in population based registers, (2) directly contacting non-respondents by telephone or single-item reply cards, and (3) longitudinal repetition of the survey. The goal of this paper was to propose an additional method to study non-response bias: when the variable of interest has a familial component, data from respondents can be used as proxy for the data from their non-responding family members. This approach was used with data on smoking, alcohol consumption, physical activity, coffee- and tea-use, education, body mass index, religion, burnout, life events, personality and mental health in large number of siblings and DZ twins registered with the Netherlands Twin Register. In addition, for smoking behavior, we also used the second strategy by sending a reply card. Results show that scores of members from less cooperative families or incomplete twin pairs tended to be more unfavorable than the scores from highly cooperative families or complete twin pairs. For example, family members from less cooperative families cycled less often and scored higher on anxious depression and neuroticism. For smoking, both the results of the reply card and the results of the additional method suggested a higher percentage smokers among the non-respondents but this was only significant with reply card method. In general, differences between highly/less cooperative families and complete/incomplete DZ twins were small. Results suggest that, even for studies with moderate response rates, data collected on health, personality and lifestyle are relatively unbiased

    The Familial Clustering of Age at Menarche in Extended Twin Families

    The timing of puberty is complex, possibly involving many genetic factors that may interact with environmental influences. Familial resemblance for age at menarche was studied in a sample of 4,995 female twins, 1,296 sisters, 2,946 mothers and 635 female spouses of male twins. They had indicated their age at menarche as part of a larger longitudinal survey. We assessed assortative mating for age at menarche, gene–environment interaction effects and estimated the heritability of individual differences in pubertal timing. There was significant evidence of gene–environment interaction, accounting for 1.5% of the variance. There was no indication of consistent mate assortment on age at menarche. Individual differences in age at menarche are highly heritable, with additive genetic factors explaining at least 70% of the true variation. An additional 1.5% of the variation can be explained by a genotype–environment interaction effect where environmental factors are more important in individuals genetically predisposed for late menarche

    Biophysical Characterization of Fluorotyrosine Probes Site-Specifically Incorporated into Enzymes:

    Fluorinated tyrosines (F[subscript n]Y's, n = 2 and 3) have been site-specifically incorporated into E. coli class Ia ribonucleotide reductase (RNR) using the recently evolved M. jannaschii Y-tRNA synthetase/tRNA pair. Class Ia RNRs require four redox active Y's, a stable Y radical (Y·) in the β subunit (position 122 in E. coli), and three transiently oxidized Y's (356 in β and 731 and 730 in α) to initiate the radical-dependent nucleotide reduction process. F[subscript n]Y (3,5; 2,3; 2,3,5; and 2,3,6) incorporation in place of Y₁₂₂-β and the X-ray structures of each resulting β with a diferric cluster are reported and compared with wt-β2 crystallized under the same conditions. The essential diferric-F[subscript n]Y· cofactor is self-assembled from apo F[subscript n]Y-β2, Fe ²⁺, and O₂ to produce ∼1 Y·/β2 and ∼3 Fe ³⁺ /β2. The F[subscript n]Y· are stable and active in nucleotide reduction with activities that vary from 5% to 85% that of wt-β2. Each F[subscript n] Y·-β2 has been characterized by 9 and 130 GHz electron paramagnetic resonance and high-field electron nuclear double resonance spectroscopies. The hyperfine interactions associated with the 19 F nucleus provide unique signatures of each F[subscript n]Y· that are readily distinguishable from unlabeled Y·'s. The variability of the abiotic F[subscript n]Y pK a 's (6.4 to 7.8) and reduction potentials (-30 to +130 mV relative to Y at pH 7.5) provide probes of enzymatic reactions proposed to involve Y·'s in catalysis and to investigate the importance and identity of hopping Y·'s within redox active proteins proposed to protect them from uncoupled radical chemistry.National Institutes of Health (U.S.) (Grant GM29595)National Science Foundation (U.S.) (Grant 0645960
