    The influence of the fungal pathogen, Zoophthora phytonomi, on larvae of the alfalfa weevil, Hypera postica, was studied in three alfalfa fields in Illinois. Disease epizootics occurred in all three fields and disease onset was ob- served within a fairly narrow range of degree day accumulations. At the height of each epizootic, percentages of infected larvae were between 80 and 100%, and the fungus contributed to the collapse of the weevil population in each field. Percent parasitism by the larval parasitoids, Bathyplectes cur­culionis and B. anurus, was lower in our fields than is common in mid-season alfalfa weevil populations and was sometimes correlated negatively with Zoophthora phytonomi infection levels, strongly implying negative interfer- ence between the parasitoids and the pathogen.     A New Microsporidium in Alfalfa Weevil Populations: Distribution and Characterization

    A microsporidium species, not previously reported, was found infecting field populations of the alfalfa weevil, Hypera postica, in Illinois. The pathogen is widely distributed thoughout the state. Percent infection ranged from 1 % to 50% at different collection locations.     Chirally symmetric quark description of low energy \pi-\pi scattering

    Weinberg's theorem for \pi-\pi scattering, including the Adler zero at threshold in the chiral limit, is analytically proved for microscopic quark models that preserve chiral symmetry. Implementing Ward-Takahashi identities, the isospin 0 and 2 scattering lengths are derived in exact agreement with Weinberg's low energy results. Our proof applies to alternative quark formulations including the Hamiltonian and Euclidean space Dyson-Schwinger approaches. Finally, the threshold \pi-\pi scattering amplitudes are calculated using the Dyson-Schwinger equations in the rainbow-ladder truncation, confirming the formal derivation.Comment: 10 pages, 7 figures, Revtex

    Catching the flu: Syndromic surveillance, algorithmic governmentality and global health security

    How do algorithms shape the imaginary and practice of security? Does their proliferation point to a shift in the political rationality of security? If so, what is the nature and extent of that shift? This article explores these questions in relation to global health security. Prompted by an epidemic of new infectious disease outbreaks – from HIV, SARS and pandemic flu, through to MERS and Ebola – many governments are making health security an integral part of their national security strategies. Algorithms are central to these developments because they underpin a number of nextgeneration syndromic surveillance systems now routinely used by governments and international organizations to rapidly detect new outbreaks globally. This article traces the origins, design and evolution of three such internet-based surveillance systems: 1) the Program for Monitoring Emerging Diseases, 2) the Global Public Health Intelligence Network, and 3) HealthMap. The article shows how the successive introduction of those three syndromic surveillance systems has propelled algorithmic technologies into the heart of global outbreak detection. This growing recourse to algorithms for the purposes of strengthening global health security, the article argues, signals a significant shift in the underlying problem, nature, and role of knowledge in contemporary security practices

    Ionic Coulomb blockade and the determinants of selectivity in the NaChBac bacterial sodium channel

    Mutation-induced transformations of conductivity and selectivity in NaChBac bacterial channels are studied experimentally and interpreted within the framework of ionic Coulomb blockade (ICB), while also taking account of resonant quantised dehydration (QD) and site protonation. Site-directed mutagenesis and whole-cell patch-clamp experiments are used to investigate how the fixed charge Qf at the selectivity filter (SF) affects both valence selectivity and same-charge selectivity. The new ICB/QD model predicts that increasing |Qf | should lead to a shift in selectivity sequences towards larger ion sizes, in agreement with the present experiments and with earlier work. Comparison of the model with experimental data leads to the introduction of an effective charge Qf∗ at the SF, which was found to differ between Aspartate and Glutamate charged rings, and also to depend on position within the SF. It is suggested that protonation of the residues within the restricted space of the SF is important in significantly reducing the effective charge of the EEEE ring. Values of Qf∗ derived from experiments on divalent blockade agree well with expectations based on the ICB/QD model and have led to the first demonstration of ICB oscillations in Ca2+ conduction as a function of the fixed charge. Preliminary studies of the dependence of Ca2+ conduction on pH are qualitatively consistent with the predictions of the model

    The Route to Water Security for Texas: The 2015–2016 Texas Water Roadmap Forums

    Three forums were held between February 2015 and November 2016, bringing together Texas water experts from business, industry, government, academia, research, and the investment community in impartially facilitated sessions to deter-mine ways to secure Texas’ water future through accelerating growth of infrastructure, technologies, research, education, and sustainable use. Consensus emerged after the first forum that Texas is approaching a water crisis reflecting matters of supply, allocation, and quality that demands immediate action to ensure water security and equitable access to this vital resource. Participant focus rested on new technology acceleration and investment, workforce education, research underway and desired by segments of the water sector, the water-energy-food nexus, outreach and public education, data management and access, water valuation, water security, and legal and regulatory frameworks. Participants also examined funding and partnership options for development of water treatment and supply infrastructure, water rights and allocation methods, aging infrastructure, and conservation, as well as the nearly ubiquitous fragmenting and compartmentalizing of just about everything having to do with water throughout the entire water sector. The forums generated and summarized a wealth of information that can be used by any party to make progress toward the goal of building a Texas water roadmap.     Importance of the Active Site "Canopy" Residues in an O_2-Tolerant [NiFe]-Hydrogenase

    The active site of Hyd-1, an oxygen-tolerant membrane-bound [NiFe]-hydrogenase from Escherichia coli, contains four highly conserved residues that form a “canopy” above the bimetallic center, closest to the site at which exogenous agents CO and O_2 interact, substrate H_2 binds, and a hydrido intermediate is stabilized. Genetic modification of the Hyd-1 canopy has allowed the first systematic and detailed kinetic and structural investigation of the influence of the immediate outer coordination shell on H_2 activation. The central canopy residue, arginine 509, suspends a guanidine/guanidinium side chain at close range above the open coordination site lying between the Ni and Fe atoms (N–metal distance of 4.4 Å): its replacement with lysine lowers the H_2 oxidation rate by nearly 2 orders of magnitude and markedly decreases the H_2/D_2 kinetic isotope effect. Importantly, this collapse in rate constant can now be ascribed to a very unfavorable activation entropy (easily overriding the more favorable activation enthalpy of the R509K variant). The second most important canopy residue for H_2 oxidation is aspartate 118, which forms a salt bridge to the arginine 509 headgroup: its mutation to alanine greatly decreases the H_2 oxidation efficiency, observed as a 10-fold increase in the potential-dependent Michaelis constant. Mutations of aspartate 574 (also salt-bridged to R509) to asparagine and proline 508 to alanine have much smaller effects on kinetic properties. None of the mutations significantly increase sensitivity to CO, but neutralizing the expected negative charges from D118 and D574 decreases O_2 tolerance by stabilizing the oxidized resting Ni^(III)–OH state (“Ni-B”).     Prevalence of admission plasma glucose in 'diabetes' or 'at risk' ranges in hospital emergencies with no prior diagnosis of diabetes by gender, age and ethnicity

    Aims To establish the prevalence of admission plasma glucose in 'diabetes' and 'at risk' ranges in emergency hospital admissions with no prior diagnosis of diabetes; characteristics of people with hyperglycaemia; and factors influencing glucose measurement. Methods Electronic patient records for 113 097 hospital admissions over 1 year from 2014 to 2015 included 43 201 emergencies with glucose available for 31 927 (74%) admissions, comprising 22 045 people. Data are presented for 18 965 people with no prior diagnosis of diabetes and glucose available on first attendance. Results Three quarters (14 214) were White Europeans aged 62 (43-78) years, median (IQ range); 12% (2241) South Asians 46 (32-64) years; 9% (1726) Unknown/Other ethnicities 43 (29-61) years; and 4% (784) Afro-Caribbeans 49 (33-63) years,  24 hours. Conclusions Hyperglycaemia was evident in 21% of adults admitted as an emergency; various aspects related to follow-up and initial testing, age and ethnicity need to be considered by professional bodies addressing undiagnosed diabetes in hospital admissions

    QCD based quark description of pi-pi scattering up to the sigma and rho region

    We study forward and backward pi-pi scattering within a QCD model based on the Dyson--Schwinger, Bethe--Salpeter equations truncated to the rainbow-ladder level. Our microscopic relativistic quark formulation preserves chiral symmetry and reproduces the observed scattering lengths for total isospin zero, one and two. At higher energies both scalar and vector meson resonances naturally occur in the scattering amplitudes. We also report a comparative study with phenomenological meson-exchange models and find such approaches are reasonable especially near pi-pi resonances.Comment: 9 pages, 7 figures, Revte

    Dietary Supplementation with Soluble Plantain Non-Starch Polysaccharides Inhibits Intestinal Invasion of Salmonella Typhimurium in the Chicken

    Soluble fibres (non-starch polysaccharides, NSP) from edible plants but particularly plantain banana (Musa spp.), have been shown in vitro and ex vivo to prevent various enteric pathogens from adhering to, or translocating across, the human intestinal epithelium, a property that we have termed contrabiotic. Here we report that dietary plantain fibre prevents invasion of the chicken intestinal mucosa by Salmonella. In vivo experiments were performed with chicks fed from hatch on a pellet diet containing soluble plantain NSP (0 to 200 mg/d) and orally infected with S.Typhimurium 4/74 at 8 d of age. Birds were sacrificed 3, 6 and 10 d post-infection. Bacteria were enumerated from liver, spleen and caecal contents. In vitro studies were performed using chicken caecal crypts and porcine intestinal epithelial cells infected with Salmonella enterica serovars following pre-treatment separately with soluble plantain NSP and acidic or neutral polysaccharide fractions of plantain NSP, each compared with saline vehicle. Bacterial adherence and invasion were assessed by gentamicin protection assay. In vivo dietary supplementation with plantain NSP 50 mg/d reduced invasion by S.Typhimurium, as reflected by viable bacterial counts from splenic tissue, by 98.9% (95% CI, 98.1–99.7; P<0.0001). In vitro studies confirmed that plantain NSP (5–10 mg/ml) inhibited adhesion of S.Typhimurium 4/74 to a porcine epithelial cell-line (73% mean inhibition (95% CI, 64–81); P<0.001) and to primary chick caecal crypts (82% mean inhibition (95% CI, 75–90); P<0.001). Adherence inhibition was shown to be mediated via an effect on the epithelial cells and Ussing chamber experiments with ex-vivo human ileal mucosa showed that this effect was associated with increased short circuit current but no change in electrical resistance. The inhibitory activity of plantain NSP lay mainly within the acidic/pectic (homogalacturonan-rich) component. Supplementation of chick feed with plantain NSP was well tolerated and shows promise as a simple approach for reducing invasive salmonellosis
