    The Stellar and Gas Kinematics of Several Irregular Galaxies

    We present long-slit spectra of three irregular galaxies from which we determinethe stellar kinematics in two of the galaxies (NGC 1156 and NGC 4449) and ionized-gas kinematics in all three (including NGC 2366). We compare this to the optical morphology and to the HI kinematics of the galaxies. In the ionized gas, we see a linear velocity gradient in all three galaxies. In NGC 1156 we also detect a weak linear velocity gradient in the stars of (5+/-1/sin i) km/s/kpc to a radius of 1.6 kpc. The stars and gas are rotating about the same axis, but this is different from the major axis of the stellar bar which dominates the optical light of the galaxy. In NGC 4449 we do not detect organized rotation of the stars and place an upper limit of (3/sin i) km/s/kpc to a radius of 1.2 kpc. For NGC 4449, which has signs of a past interaction with another galaxy, we develop a model to fit the observed kinematics of the stars and gas. In this model the stellar component is in a rotating disk seen nearly face-on while the gas is in a tilted disk with orbits whose planes precess in the gravitational potential. This model reproduces the apparent counter-rotation of the inner gas of the galaxy.     The stellar velocity dispersion in the inner 1.3 disk scale-lengths of the irregular galaxy NGC 4449

    We present measurements of the stellar velocity dispersion in the inner 1 arcmin radius (1.3 disk scale-lengths) of the irregular galaxy NGC 4449 determined from long-slit absorption-line spectra. The average observed dispersion is 29 +/-2 km/s, the same as predicted from NGC 4449's luminosity. No significant rotation in the stars is detected. If we assume a maximum rotation speed of the stars from the model determined from the gas kinematics of Hunter et al. (2002), the ratio V_max/sigma_z measured globally is 3. This ratio is comparable to values measured in spiral galaxies, and implies that the stellar disk in NGC 4449 is kinematically relatively cold. The intrinsic minor-to-major axis ratio (b/a)_0 is predicted to be in the range 0.3-0.6, similar to values derived from the distribution of observed b/a of Im galaxies. However, V/sigma_z measured locally is 0.5-1.1, and so the circular velocity of NGC 4449 is comparable or less than the velocity of the stars within the central 1.3 disk scale-lengths of the galaxy.Comment: To be published in ApJ, Nov 200

    Late Quaternary slip rates across the central Tien Shan, Kyrgyzstan, central Asia

    Slip rates across active faults and folds show that late Quaternary faulting is distributed across the central Tien Shan, not concentrated at its margins. Nearly every intermontane basin contains Neogene and Quaternary syntectonic strata deformed by Holocene north‐south shortening on thrust or reverse faults. In a region that spans two thirds of the north‐south width of the central Tien Shan, slip rates on eight faults in five basins range from ∌0.1 to ∌3 mm/yr. Fault slip rates are derived from faulted and folded river terraces and from trenches. Radiocarbon, optically stimulated luminescence, and thermoluminescence ages limit ages of terraces and aid in their regional correlation. Monte Carlo simulations that sample from normally distributed and discrete probability distributions for each variable in the slip rate calculations generate most likely slip rate values and 95% confidence limits. Faults in basins appear to merge at relatively shallow depths with crustal‐scale ramps that underlie mountain ranges composed of pre‐Cenozoic rocks. The sum and overall pattern of late Quaternary rates of shortening are similar to current rates of north‐south shortening measured using Global Positioning System geodesy. This similarity suggests that deformation is concentrated along major fault zones near range‐basin margins. Such faults, separated by rigid blocks, accommodate most of the shortening in the upper crust

    Evidence for prehistoric coseismic folding along the Tsaotun segment of the Chelungpu fault near Nan-Tou, Taiwan

    Taiwan\u27s 1999 Mw 7.6 earthquake generated over 85 km surface rupture along the Chelungpu thrust fault. Paleoseismic studies at the Shi‐Jia site near Nantou city, reveal folding as the predominant form of deformation. Stratigraphic relations across the 1999 fold scarp show the style and degree of deformation caused by the penultimate event is similar to observed 1999 deformation. A boring transect across the fold scarp provides additional evidence of an earlier earthquake. Investigations at the Shi‐Jia site revealed three prehistoric events; accelerator mass spectrometry (AMS) radiocarbon ages indicate that the penultimate earthquake occurred between 1160 and 1440 A.D. Paleoseismic studies north of the Shi‐Jia site reveal much younger penultimate earthquakes, suggesting a 1999‐type event may not be characteristic along the Tsaotun segment of the Chelungpu fault

    Centimeter to decimeter hollow concretions and voids in Gale Crater sediments, Mars

    Voids and hollow spheroids between ∌1 and 23 cm in diameter occur at several locations along the traverse of the Curiosity rover in Gale crater, Mars. These hollow spherical features are significantly different from anything observed in previous landed missions. The voids appear in dark-toned, rough-textured outcrops, most notably at Point Lake (sols 302-305) and Twin Cairns Island (sol 343). Point Lake displays both voids and cemented spheroids in close proximity; other locations show one or the other form. The spheroids have 1-4 mm thick walls and appear relatively dark-toned in all cases, some with a reddish hue. Only one hollow spheroid (Winnipesaukee, sol 653) was analyzed for composition, appearing mafic (Fe-rich), in contrast to the relatively felsic host rock. The interior surface of the spheroid appears to have a similar composition to the exterior with the possible exceptions of being more hydrated and slightly depleted in Fe and K. Origins of the spheroids as Martian tektites or volcanic bombs appear unlikely due to their hollow and relatively fragile nature and the absence of in-place clearly igneous rocks. A more likely explanation to both the voids and the hollow spheroids is reaction of reduced iron with oxidizing groundwater followed by some re-precipitation as cemented rind concretions at a chemical reaction front. Although some terrestrial concretion analogs are produced from a precursor siderite or pyrite, diagenetic minerals could also be direct precipitates for other terrestrial concretions. The Gale sediments differ from terrestrial sandstones in their high initial iron content, perhaps facilitating a higher occurrence of such diagenetic reactions

    The nedd-8 activating enzyme gene underlies genetic resistance to infectious pancreatic necrosis virus in Atlantic salmon

    Genetic resistance to infectious pancreatic necrosis virus (IPNV) in Atlantic salmon is a rare example of a trait where a single locus (QTL) explains almost all of the genetic variation. Genetic marker tests based on this QTL on salmon chromosome 26 have been widely applied in selective breeding to markedly reduce the incidence of the disease. In the current study, whole genome sequencing and functional annotation approaches were applied to characterise genes and variants in the QTL region. This was complemented by an analysis of differential expression between salmon fry of homozygous resistant and homozygous susceptible genotypes challenged with IPNV. These analyses pointed to the NEDD-8 activating enzyme 1 (nae1) gene as a putative functional candidate underlying the QTL effect. The role of nae1 in IPN resistance was further assessed via CRISPR-Cas9 knockout of the nae1 gene and chemical inhibition of the nae1 protein activity in Atlantic salmon cell lines, both of which resulted in highly significant reduction in productive IPNV replication. In contrast, CRISPR-Cas9 knockout of a candidate gene previously purported to be a cellular receptor for the virus (cdh1) did not have a major impact on productive IPNV replication. These results suggest that nae1 is the causative gene underlying the major QTL affecting resistance to IPNV in salmon, provide further evidence for the critical role of neddylation in host-pathogen interactions, and highlight the value in combining high-throughput genomics approaches with targeted genome editing to understand the genetic basis of disease resistance

    Assessment of Right Ventricular Function in the Research Setting: Knowledge Gaps and Pathways Forward. An Official American Thoracic Society Research Statement

    BACKGROUND: Right ventricular (RV) adaptation to acute and chronic pulmonary hypertensive syndromes is a significant determinant of short- and long-term outcomes. Although remarkable progress has been made in the understanding of RV function and failure since the meeting of the NIH Working Group on Cellular and Molecular Mechanisms of Right Heart Failure in 2005, significant gaps remain at many levels in the understanding of cellular and molecular mechanisms of RV responses to pressure and volume overload, in the validation of diagnostic modalities, and in the development of evidence-based therapies. METHODS: A multidisciplinary working group of 20 international experts from the American Thoracic Society Assemblies on Pulmonary Circulation and Critical Care, as well as external content experts, reviewed the literature, identified important knowledge gaps, and provided recommendations. RESULTS: This document reviews the knowledge in the field of RV failure, identifies and prioritizes the most pertinent research gaps, and provides a prioritized pathway for addressing these preclinical and clinical questions. The group identified knowledge gaps and research opportunities in three major topic areas: 1) optimizing the methodology to assess RV function in acute and chronic conditions in preclinical models, human studies, and clinical trials; 2) analyzing advanced RV hemodynamic parameters at rest and in response to exercise; and 3) deciphering the underlying molecular and pathogenic mechanisms of RV function and failure in diverse pulmonary hypertension syndromes. CONCLUSIONS: This statement provides a roadmap to further advance the state of knowledge, with the ultimate goal of developing RV-targeted therapies for patients with RV failure of any etiology

    Weekly vs. Every-3-Week Paclitaxel and Carboplatin for Ovarian Cancer

    BACKGROUND A dose-dense weekly schedule of paclitaxel (resulting in a greater frequency of drug delivery) plus carboplatin every 3 weeks or the addition of bevacizumab to paclitaxel and carboplatin administered every 3 weeks has shown efficacy in ovarian cancer. We proposed to determine whether dose-dense weekly paclitaxel and carboplatin would prolong progression-free survival as compared with paclitaxel and carboplatin administered every 3 weeks among patients receiving and those not receiving bevacizumab. METHODS We prospectively stratified patients according to whether they elected to receive bevacizumab and then randomly assigned them to receive either paclitaxel, administered intravenously at a dose of 175 mg per square meter of body-surface area every 3 weeks, plus carboplatin (dose equivalent to an area under the curve [AUC] of 6) for six cycles or paclitaxel, administered weekly at a dose of 80 mg per square meter, plus carboplatin (AUC, 6) for six cycles. The primary end point was progression-free survival. RESULTS A total of 692 patients were enrolled, 84% of whom opted to receive bevacizumab. In the intention-to-treat analysis, weekly paclitaxel was not associated with longer progression-free survival than paclitaxel administered every 3 weeks (14.7 months and 14.0 months, respectively; hazard ratio for disease progression or death, 0.89; 95% confidence interval [CI], 0.74 to 1.06; P=0.18). Among patients who did not receive bevacizumab, weekly paclitaxel was associated with progression-free survival that was 3.9 months longer than that observed with paclitaxel administered every 3 weeks (14.2 vs. 10.3 months; hazard ratio, 0.62; 95% CI, 0.40 to 0.95; P=0.03). However, among patients who received bevacizumab, weekly paclitaxel did not significantly prolong progression-free survival, as compared with paclitaxel administered every 3 weeks (14.9 months and 14.7 months, respectively; hazard ratio, 0.99; 95% CI, 0.83 to 1.20; P=0.60). A test for interaction that assessed homogeneity of the treatment effect showed a significant difference between treatment with bevacizumab and without bevacizumab (P=0.047). Patients who received weekly paclitaxel had a higher rate of grade 3 or 4 anemia than did those who received paclitaxel every 3 weeks (36% vs. 16%), as well as a higher rate of grade 2 to 4 sensory neuropathy (26% vs. 18%); however, they had a lower rate of grade 3 or 4 neutropenia (72% vs. 83%). CONCLUSIONS Overall, weekly paclitaxel, as compared with paclitaxel administered every 3 weeks, did not prolong progression-free survival among patients with ovarian cancer

    Phase II trial of debulking surgery and photodynamic therapy for disseminated intraperitoneal tumors

    Background: Photodynamic therapy (PDT) combines photosensitizer drug, oxygen, and laser light to kill tumor cells on surfaces. This is the initial report of our phase II trial, designed to evaluate the effectiveness of surgical debulking and PDT in carcinomatosis and sarcomatosis. Methods: Fifty-six patients were enrolled between April 1997 and January 2000. Patients were given Photofrin (2.5 mg/kg) intravenously 2 days before tumor-debulking surgery. Laser light was delivered to all peritoneal surfaces. Patients were followed with CT scans and laparoscopy to evaluate responses to treatment. Results: Forty-two patients were adequately debulked at surgery; these comprise the treatment group. There were 14 GI malignancies, 12 ovarian cancers and 15 sarcomas. Actuarial median survival was 21 months. Median time to recurrence was 3 months (range, 1-21 months). The most common serious toxicities were anemia (38%), liver function test (LFT) abnormalities (26%), and gastrointestinal toxicities(19%), and one patient died. Conclusions: Photofrin PDT for carcinomatosis has been successfully administered to 42 patients, with acceptable toxicity. The median survival of 21 months exceeds our expectations; however, the relative contribution of surgical resection versus PDT is unknown. Deficiencies in photosensitizer delivery, tissue oxygenation, or laser light distribution leading to recurrences may be addressed through the future use of new photosensitizers
