The Phenotypic Analysis of Lactobacillus plantarum shsp Mutants Reveals a Potential Role for hsp1 in Cryotolerance

Small heat shock proteins (sHSPs) are ubiquitous, low molecular weight (MW) proteins that share a conserved alpha-crystallin domain. sHSPs oligomers exhibit chaperon-like activities by interacting with unfolded substrates, thereby preventing their aggregation and precipitation. Unlike most lactobacilli, which have single shsp genes, three different sHSP-encoding genes, i.e., hsp1, hsp2, and hsp3, were previously identified in the probiotic Lactobacillus plantarum WCFS1. Early studies, including the characterization of the knock out (KO) mutant for hsp2, indicated a different organization and transcriptional regulation of these genes and suggested that the three L. plantarum sHSPs might accomplish different tasks in stress response. To unravel the role of sHSPs, KO mutants of hsp1 and hsp3 were generated using a Cre-lox based system. Mutation of either genes resulted in impaired growth capacity under normal conditions, heat-stress and stresses typically found during host interactions and food technological process. However, survival to heat shock and the level of thermal stabilization of cytoplasmic proteins were similar between mutants and parental strain. Transcriptional analysis revealed that in the mutant genetic backgrounds there is an upregulated basal expression of the un-mutated mate hsps and other stress-related genes, which may compensate for the loss of HSP function, hence possibly accounting for the lack of a remarkable susceptibility to heat challenge. HSP3 seemed relevant for the induction of thermotolerance, while HSP1 was required for improved cryotolerance. Cell surface properties and plasma membrane fluidity were investigated to ascertain the possible membrane association of sHSP. Intriguingly, the loss of hsp1 was associated to a lower level of maximal membrane fluidity upon heat stress. A role for HSP1 in controlling and improving membrane fluidity is suggested which may pertains its cryoprotective function

The partial position paper on the topic of "Housing and settlement development" is related to the parallel position papers of the working team of the ARL state working group NRW. Its task is to contribute to the presentation of future prospects for rural areas in North Rhine-Westphalia, to discuss problems and to make recommendations for stimulating and implementing measures. The demographic development in North Rhine-Westphalia is characterised by strong re-regional differences between growth and shrinkage. The quantitative and qualitative changes caused by population and household development have different regional effects on future housing needs and demands. The finite nature of land resources makes it necessary to focus more on internal development. As a result of a critical analysis of the challenges, the future prospects of rural areas as residential and settlement areas in NRW are based on five main themes. They relate to expanded and modified offers of small housing units for different demand groups, the strengthening of holistic and strategic planning approaches in rural municipalities, active land resource management in municipal and regional housing and settlement development, the use of municipal control instruments and regional and inter-municipal cooperation. The above-mentioned topics are presented and explained in five core-requirements. Es steht im Zusammenhang mit den parallel erarbeiteten Positionspapieren der Arbeitsgruppe der ARL-Landesarbeitsgemeinschaft NRW und trägt mit diesen zusammen dazu bei, Zukunftsperspektiven ländlicher Räume in Nordrhein-Westfalen vorzustellen. In Nordrhein-Westfalen tragen die ländlichen Räume - ungeachtet großer regionaler Differenziertheit - maßgeblich zur wirtschaftlichen Leistungskraft des Landes bei. In addition to climate change, major challenges for economic development and for the transformation of rural working environments arise from demographic change, an increasing land use competition, digitization and structural change in agriculture. The corona pandemic is currently setting additional accents, the consequences which are not yet fully foreseeable. As a result of a critical examination of the challenges, the future prospects of rural areas as economic regions in NRW are based on five main issues. They relate to a new understanding of growth, the securing and retention of skilled labour as a central factor of economic stability and development capability. Furthermore they relate to options for action to reduce the problem of land availability and competition, to changed, regionally adapted concepts of economic development and, last but not least, to a forward-looking, approach to an increasingly differentiated agricultural sector and a changing role of agriculture in society. The partial position paper on the topic of "Public Service Provision" is related to the parallel position papers of the working team of the ARL state working group NRW. Its task is to contribute to presenting the future prospects of rural areas in North Rhine-Westphalia, to discuss problems and to make recommendations for stimulating and implementing measures. The present position paper explicitly addresses the areas of fire brigades and fire protection as well as health and care in the rural areas of North Rhine-Westphalia. Both areas are facing great challenges, among other things due to demographic change, but are important for everyday life and the future of rural areas. The above-mentioned topics are presented and discussed in five core-requirements. Each of them is followed by address-oriented recommendations for action. Decisive challenges for both areas are, among others, the lack of skilled labor and migration, especially of younger population groups. Furthermore there are a decline in volunteers, and outdated organizational structures. The consequences of these developments require very different approaches in rural areas in order to ensure the provision of services of general interest. Above all, mobile concepts, digitization and the promotion of voluntary work are central approaches for action. The combination of family and work is nowadays an important starting point for the solution of many problems, so that, among other items, the role of women in this context must be reconsidered and promoted with regard to voluntary work. In times of the corona pandemic, digitalization in particular offers a great opportunity. Together with four parallel thematically focussed sub-position papers, the framework position paper presented here sets the presentation framework in which the results of the working group "Future perspectives of rural areas in NRW" of the ARL, LAG NRW are presented. The four main topics are "Economy and Work", "Housing and Settlement Development", "Civic Engagement and Volunteering" and "Services of General Interest, Focus: Fire Department/Fire Protection and Health/Care". The framework position paper provides introductory information on the task at hand. It is based on the structural-functional definition of "Types of rural areas" by the Thuenen Institute (Federal Research Institute for Rural Areas, Forests and Fisheries) and describes the working group's self-conception in "dealing" with rural areas. In their great heterogeneity and with regard to their development perspectives, these are seen less as problem areas than as regions of opportunities. A future-oriented approach is linked to this perspective. The framework position paper explains the derivation of the focused priority topics, which are processed in greater detail and on the basis of core-requirements and recommendations for action in the sub-position papers, and condenses the results obtained there in the form of four summarising core claims. The challenges and opportunities for the development of rural areas in NRW on which the framework position paper is based have been derived from current specific rural problems, but above all also from generally valid megatrends which will shape the future and do not only apply to rural areas. These include demographic change, digital transformation, socio-economic and cultural changes in society and work, and changing lifestyles, as well as climate change. Furthermore, the paper emphasises the role of urban development qualities and regional building culture as qualitative location factors not to be neglected in the context of local and regional development. In terms of the heterogenity of rural areas and their initial situations, sustainability means taking up the influences outlined above and developing and providing tailor-made strategies for steering and programmatic approaches to supporting rural development paths. The partial position paper on the topic of "Civic Engagement and Volunteering" is related to the parallel position papers of the working team of the ARL state working group NRW. Its task is to contribute to the presentation of future prospects for rural areas in North Rhine-Westphalia, to discuss problems and to make recommendations for stimulating and implementing measures. This paper focuses on the importance of civic engagement for the creation of the equivalence of living conditions, the provision of services of general interest in rural areas and as a driver of social innovation. Civic engagement is a pillar of rural development and must be understood as a driving force of regional economy, especially in rural areas. But engagement is changing. Associations, initiatives, municipalities and regions must respond to this if they take civic engagement and voluntary work seriously as a key force in shaping the future of rural areas. As a result of critically examining the challenges, the authors equate civic engagement in its core thesis with the cognition, appreciation and significance and their need for human and financial resources with that of local economic development (agencies). Four further thesis and concrete recommendations for action are derived from this basic understanding. They call for the creation of local/regional models and strategies for civic commitment in the sense of a clear "must have" for processes and projects on the future of rural areas in NRW. They refer to new concepts of life which are becoming more and more differentiated, to a culture of commitment and to a changing role of commitment in society. They point out options for networking activities, funding opportunities, support structures and their professionalization and plead for consistent investment in minds, formats and inspiring and impulse-giving places. Digitization and digital transformation are seen as a challenge, but above all as an opportunity for future engagement. The above topics are presented and explained in five core-requirements. Aus diesem Grundverständnis heraus werden vier weitere Kernforderungen und konkrete Handlungsempfehlungen abgeleitet. Erfahrungen, Herausforderungen und Lösungsansätze aus der Extraktion pseudonymer Daten für das Projekt INDEED

Background: In Germany there is currently no health reporting on cross-sectoral care patterns in the context of an emergency department care treatment. The INDEED project (Utilization and trans-sectoral patterns of care for patients admitted to emergency departments in Germany) collects routine data from 16 emergency departments, which are later merged with outpatient billing data from 2014 to 2017 on an individual level. Aim: The methodological challenges in planning of the internal merging of routine clinical and administrative data from emergency departments in Germany up to the final data extraction are presented together with possible solution approaches. Methods: Data were selected in an iterative process according to the research questions, medical relevance, and assumed data availability. After a preparatory phase to clarify formalities (including data protection, ethics), review test data and correct if necessary, the encrypted and pseudonymous data extraction was performed. Results: Data from the 16 cooperating emergency departments came mostly from the emergency department and hospital information systems. There was considerable heterogeneity in the data. Not all variables were available in every emergency department because, for example, they were not standardized and digitally available or the extraction effort was judged to be too high. Conclusion: Relevant data from emergency departments are stored in different structures and in several IT systems. Thus, the creation of a harmonized data set requires considerable resources on the part of the hospital as well as the data processing unit. This needs to be generously calculated for future projects
