    A Textured Silicon Calorimetric Light Detector

    We apply the standard photovoltaic technique of texturing to reduce the reflectivity of silicon cryogenic calorimetric light detectors. In the case of photons with random incidence angles, absorption is compatible with the increase in surface area. For the geometrically thin detectors studied, energy resolution from athermal phonons, dominated by position dependence, is proportional to the surface-to-volume ratio. With the CaWO4 scintillating crystal used as light source, the time constants of the calorimeter should be adapted to the relatively slow light-emission times.Comment: Submitted to Journal of Applied Physic

    Kinetic theory for non-equilibrium stationary states in long-range interacting systems

    We study long-range interacting systems perturbed by external stochastic forces. Unlike the case of short-range systems, where stochastic forces usually act locally on each particle, here we consider perturbations by external stochastic fields. The system reaches stationary states where external forces balance dissipation on average. These states do not respect detailed balance and support non-vanishing fluxes of conserved quantities. We generalize the kinetic theory of isolated long-range systems to describe the dynamics of this non-equilibrium problem. The kinetic equation that we obtain applies to plasmas, self-gravitating systems, and to a broad class of other systems. Our theoretical results hold for homogeneous states, but may also be generalized to apply to inhomogeneous states. We obtain an excellent agreement between our theoretical predictions and numerical simulations.     Loss of Melanoregulin (MREG) Enhances Cathepsin-D Secretion by the Retinal Pigment Epithelium

    Cathepsin-D (Cat-D) is a major proteolytic enzyme in phagocytic cells. In the retinal pigment epithelium (RPE), it is responsible for the daily degradation of photoreceptor outer segments (POSs) to maintain retinal homeostasis. Melanoregulin (MREG)-mediated loss of phagocytic capacity has been linked to diminished intracellular Cat-D activity. Here, we demonstrate that loss of MREG enhances the secretion of intermediate Cat-D (48 kDa), resulting in a net enhancement of extracellular Cat-D activity. These results suggest that MREG is required to maintain Cat-D homeostasis in the RPE and likely plays a protective role in retinal health. In this regard, in the Mreg dsu/dsu mouse, we observe increased basal laminin. Loss of the Mreg dsu allele is not lethal and therefore leads to slow age-dependent changes in the RPE. Thus, we propose that this model will allow us to study potential dysregulatory functions of Cat-D in retinal disease

    The CRESST Experiment: Recent Results and Prospects

    The CRESST experiment seeks hypothetical WIMP particles that could account for the bulk of dark matter in the Universe. The detectors are cryogenic calorimeters in which WIMPs would scatter elastically on nuclei, releasing phonons. The first phase of the experiment has successfully deployed several 262 g sapphire devices in the Gran Sasso underground laboratories. A main source of background has been identified as microscopic mechanical fracturing of the crystals, and has been eliminated, improving the background rate by up to three orders of magnitude at low energies, leaving a rate close to one count per day per kg and per keV above 10 keV recoil energy. This background now appears to be dominated by radioactivity, and future CRESST scintillating calorimeters which simultaneously measure light and phonons will allow rejection of a great part of it.Comment: To appear in the proceedings of the CAPP2000 Conference, Verbier, Switzerland, July, 2000 (eds J.     Cusp energetic particle events: Implications for a major acceleration region of the magnetosphere

    The Charge and Mass Magnetospheric Ion Composition Experiment (CAMMICE) on board the Polar spacecraft observed 75 energetic particle events in 1996 while the satellite was at apogee. All of these events were associated with a decrease in the magnitude of the local magnetic field measured by the Magnetic Field Experiment (MFE) on Polar. These new events showed several unusual features: (1) They were detected in the dayside polar cusp near the apogee of Polar with about 79% of the total events in the afternoonside and 21% in the morningside; (2) an individual event could last for hours; (3) the measured helium ion had energies up to and many times in excess of 2.4 MeV; (4) the intensity of 1–200 KeV/e helium was anticorrelated with the magnitude of the local geomagnetic field but correlated with the turbulent magnetic energy density; (5) the events were associated with an enhancement of the low-frequency magnetic noise, the spectrum of which typically extends from a few hertz to a few hundreds of hertz as measured by the Plasma Wave Instrument (PWI) on Polar; and (6) a seasonal variation was found for the occurrence rate of the events with a maximum in September. These characterized a new phenomenon which we are calling cusp energetic particle (CEP) events. The observed high charge state of helium and oxygen ions in the CEP events indicates a solar source for these particles. Furthermore, the measured 0.52–1.15 MeV helium flux was proportional to the difference between the maximum and the minimum magnetic field in the event. A possible explanation is that the energetic helium ions are energized from lower energy helium by a local acceleration mechanism associated with the high-altitude dayside cusp. These observations represent a potential discovery of a major acceleration region of the magnetosphere

    A Synoptic X-ray Study of M31 with the Chandra-HRC

    We have obtained 17 epochs of Chandra High Resolution Camera (HRC) snapshot images, each covering most of the M31 disk. The data cover a total baseline of 2.5 years and contain a mean effective exposure of 17 ks. We have measured the mean fluxes and long-term lightcurves for 166 objects detected in these data. At least 25% of the sources show significant variability. The cumulative luminosity function (CLF) of the disk sources is well-fit by a power-law with a slope comparable to those observed in typical elliptical galaxies. The CLF of the bulge is a broken power law similar to measurements made by previous surveys. We note several sources in the southwestern disk with L_X > 10^{37} erg/s . We cross-correlate all of our sources with published optical and radio catalogs, as well as new optical data, finding counterpart candidates for 55 sources. In addition, 17 sources are likely X-ray transients. We analyze follow-up HST WFPC2 data of two X-ray transients, finding F336W (U-band equivalent) counterparts. In both cases, the counterparts are variable. In one case, the optical counterpart is transient with F336W = 22.3 +/- 0.1 mag. The X-ray and optical properties of this object are consistent with a ~10 solar mass black hole X-ray nova with an orbital period of ~20 days. In the other case, the optical counterpart varies between F336W = 20.82 +/- 0.06 mag and F336W = 21.11 +/- 0.02 mag. Ground-based and HST observations show this object is bright (V = 18.8 +/- 0.1) and slightly extended. Finally, the frequency of bright X-ray transients in the M31 bulge suggests that the ratio of neutron star to black hole primaries in low-mass X-ray binaries (NS/BH) is ~1.Comment: 68 pages (27 text), 8 tables, 16 figures, 1 appendix, accepted by ApJ; accepted version contains reorganized text, new tables and figures, and updated result

    Aggregatibacter Actinomycetemcomitans Leukotoxin Cytotoxicity Occurs Through Bilayer Destabilization

    The Gram-negative bacterium, Aggregatibacter actinomycetemcomitans, is a common inhabitant of the human upper aerodigestive tract. The organism produces an RTX (Repeats in ToXin) toxin (LtxA) that kills human white blood cells. LtxA is believed to be a membrane-damaging toxin, but details of the cell surface interaction for this and several other RTX toxins have yet to be elucidated. Initial morphological studies suggested that LtxA was bending the target cell membrane. Because the ability of a membrane to bend is a function of its lipid composition, we assessed the proficiency of LtxA to release of a fluorescent dye from a panel of liposomes composed of various lipids. Liposomes composed of lipids that form nonlamellar phases were susceptible to LtxA-induced damage while liposomes composed of lipids that do not form non-bilayer structures were not. Differential scanning calorimetry demonstrated that the toxin decreased the temperature at which the lipid transitions from a bilayer to a nonlamellar phase, while 31P nuclear magnetic resonance studies showed that the LtxA-induced transition from a bilayer to an inverted hexagonal phase occurs through the formation of an isotropic intermediate phase. These results indicate that LtxA cytotoxicity occurs through a process of membrane destabilization. © 2012 Blackwell Publishing Ltd

    Revision of previous Fontan connections to total extracardiac cavopulmonary anastomosis: A multicenter experience

    AbstractBackground: Conversion to total extracardiac cavopulmonary anastomosis is an option for managing patients with dysfunction of a prior Fontan connection. Methods: Thirty-one patients (19.9 ± 8.8 years) underwent revision of a previous Fontan connection to total extracardiac cavopulmonary anastomosis at four institutions. Complications of the previous Fontan connection included atrial tachyarrhythmias (n = 20), progressive heart failure (n = 17), Fontan pathway obstruction (n = 10), effusions (n = 10), pulmonary venous obstruction by an enlarged right atrium (n = 6), protein-losing enteropathy (n = 3), right atrial thrombus (n = 2), subaortic stenosis (n = 1), atrioventricular valve regurgitation (n = 3), and Fontan baffle leak (n = 5). Conversion to an extracardiac cavopulmonary connection was performed with a nonvalved conduit from the inferior vena cava to the right pulmonary artery, with additional procedures as necessary. Results: There have been 3 deaths. Two patients died in the perioperative period of heart failure and massive effusions. The third patient died suddenly 8 months after the operation. All surviving patients were in New York Heart Association class I (n = 20) or II (n = 7), except for 1 patient who underwent heart transplantation. Early postoperative arrhythmias occurred in 10 patients: 4 required pacemakers, and medical therapy was sufficient in 6. In 15 patients, pre-revision arrhythmias were improved. Effusions resolved in all but 1 of the patients in whom they were present before revision. The condition of 2 patients with protein-losing enteropathy improved within 30 days. Conclusions: Conversion of a failing Fontan connection to extracardiac cavopulmonary connection can be achieved with low morbidity and mortality. Optimally, revision should be undertaken early in symptomatic patients before irreversible ventricular failure ensues. (J Thorac Cardiovasc Surg 2000;119:340-6

    Trigeminal neuropathy presenting secondary to SARS-CoV-2 infection

    Introduction: A 58-year-old woman presented to a multidisciplinary facial pain clinic in October 2021 complaining of a constant pain in the right side of her face since contracting coronavirus SARS-CoV-2 18 months earlier. The pain extending from the right temple down to her right cheek extraorally and including the maxillary teeth and right side of tongue intraorally. This was accompanied by anosmia, diplopia on lateral gaze, and dizziness. Methods: Clinical examination was supplemented with several neurophysiological tests to confirm the diagnosis including an MRI brain scan, quantitative sensory testing, electrophysiological blink reflex testing, corneal confocal microscopy, and pain and short-form anxiety and depression questionnaires. Results: Quantitative sensory testing showed unilateral loss of perception in thermal and mechanical sensibility and bilateral hyperalgesia indicating central sensitization. Bilateral corneal confocal microscopy showed an abnormally reduced corneal nerve fibre length on the right side. MRI, blink reflex, and masseter inhibitory testing findings were normal. Conclusion: This case study is the first case of trigeminal neuropathy related to SARS-CoV-2 infection reported in the literature. It also discusses the successful management of the patient's trigeminal neuropathic pain

    Kaposi's sarcoma‐associated herpesvirus serology in Europe and Uuganda: Multicentre study with multiple and novel assays

    AbstractA multicentre study was undertaken to define novel assays with increased inter‐assay concordance, sensitivity, specificity and predictive value for serological diagnosis of human herpesvirus type 8 (HHV‐8) infection. A total of 562 sera from European and Ugandan human immunodeficiency virus (HIV)‐infected or uninfected individuals with or without Kaposi's sarcoma (KS) and blood donors were examined under code by 18 different assays in seven European laboratories. Sera from KS patients and all non‐KS sera found positive by at least 70%, 80%, or 90% of the assays were considered "true positive." The validity of the assays was then evaluated by univariate logistic regression analysis. Two immunofluorescence assays (IFA) for detection of antibodies against HHV‐8 lytic (Rlyt) or latent (LLANA) antigens and two enzyme‐linked‐immunosorbent assays (ELISA) (M2, EK8.1) for detection of antibodies against HHV‐8 structural proteins were found to be highly concordant, specific, and sensitive, with odds ratios that indicated a high predictive value. When used together, the two IFA (Rlyt‐LLANA) showed the best combination of sensitivity (89.1%) and specificity (94.9%). The performance of these assays indicate that they may be used for the clinical management of individuals at risk of developing HHV‐8 associated tumours such as allograft recipients. J. Med.
