    The Multiple Altimeter Beam Experimental Lidar (MABEL), an Airborne Simulator for the ICESat-2 Mission

    The Ice, Cloud, and land Elevation Satellite-2 (ICESat-2) mission is currently under development by NASA. The primary mission of ICESat-2 will be to measure elevation changes of the Greenland and Antarctic ice sheets, document changes in sea ice thickness distribution, and derive important information about the current state of the global ice coverage. To make this important measurement, NASA is implementing a new type of satellite-based surface altimetry based on sensing of laser pulses transmitted to, and reflected from, the surface. Because the ICESat-2 measurement approach is different from that used for previous altimeter missions, a high-fidelity aircraft instrument, the Multiple Altimeter Beam Experimental Lidar (MABEL), was developed to demonstrate the measurement concept and provide verification of the ICESat-2 methodology. The MABEL instrument will serve as a prototype for the ICESat-2 mission and also provides a science tool for studies of land surface topography. This paper outlines the science objectives for the ICESat-2 mission, the current measurement concept for ICESat-2, and the instrument concept and preliminary data from MABEL

    Impact of antibody quality and anamnestic response on viremia control post-challenge in a combined Tat/Env vaccine regimen in rhesus macaques

    AbstractPreviously, priming rhesus macaques with Adenovirus type 5 host range mutant-recombinants encoding Tat and Env and boosting with Tat and Env protein in MPL-SE controlled chronic viremia by 4 logs following homologous intravenous SHIV89.6P challenge. Here we evaluated Tat, Env, and Tat/Env regimens for immunogenicity and protective efficacy using clade C Env, alum adjuvant, and a heterologous intrarectal SHIV1157ipd3N4 challenge. Despite induction of strong cellular and humoral immunity, Tat/Env group T and B-cell memory responses were not significantly enhanced over Tat- or Env-only groups. Lack of viremia control post-challenge was attributed to lower avidity Env antibodies and no anamnestic ADCC response or SHIV1157ipd3N4 neutralizing antibody development post-challenge. Poor biologic activity of the Tat immunogen may have impaired Tat immunity. In the absence of sterilizing immunity, strong anamnestic responses to heterologous virus can help control viremia. Both antibody breadth and optimal adjuvanticity are needed to elicit high-quality antibody for protective efficacy

    Whole-genome sequence data uncover widespread heterothallism in the largest group of lichen-forming fungi

    Fungal reproduction is regulated by the mating-type (MAT1) locus, which typically comprises two idiomorphic genes. The presence of one or both allelic variants at the locus determines the reproductive strategy in Fungi – homothallism vs. heterothallism. It has been hypothesized that self-fertility via homothallism is widespread in lichen-forming fungi. To test this hypothesis, we characterized the MAT1 locus of 41 genomes of lichen-forming fungi representing a wide range of growth forms and reproductive strategies in the class Lecanoromycetes, the largest group of lichen-forming fungi. Our results show the complete lack of genetic homothallism suggesting that lichens evolved from a heterothallic ancestor. We argue that this may be related to the symbiotic lifestyle of these fungi, and may be a key innovation that has contributed to the accelerated diversification rates in this fungal group

    General statistical scaling laws for stability in ecological systems

    Ecological stability refers to a family of concepts used to describe how systems of interacting species vary through time and respond to disturbances. Because observed ecological stability depends on sampling scales and environmental context, it is notoriously difficult to compare measurements across sites and systems. Here, we apply stochastic dynamical systems theory to derive general statistical scaling relationships across time, space, and ecological level of organisation for three fundamental stability aspects: resilience, resistance, and invariance. These relationships can be calibrated using random or representative samples measured at individual scales, and projected to predict average stability at other scales across a wide range of contexts. Moreover deviations between observed vs. extrapolated scaling relationships can reveal information about unobserved heterogeneity across time, space, or species. We anticipate that these methods will be useful for cross-study synthesis of stability data, extrapolating measurements to unobserved scales, and identifying underlying causes and consequences of heterogeneity

    Enhanced airway sensory nerve reactivity in non-eosinophilic asthma

    BACKGROUND: Neural mechanisms may play an important role in non-eosinophilic asthma (NEA). This study compared airway sensory nerve reactivity, using capsaicin challenge, in eosinophilic asthma (EA) and NEA and non-asthmatics. METHODS: Thirty-eight asthmatics and 19 non-asthmatics (aged 14-21 years) underwent combined hypertonic saline challenge/sputum induction, fractional exhaled nitric oxide, atopy and spirometry tests, followed by capsaicin challenge. EA and NEA were defined using a sputum eosinophil cut-point of 2.5%. Airway hyperreactivity was defined as a ≥15% drop in FEV1 during saline challenge. Sensory nerve reactivity was defined as the lowest capsaicin concentration that evoked 5 (C5) coughs. RESULTS: Non-eosinophilic asthmatics (n=20) had heightened capsaicin sensitivity (lower C5) compared with non-asthmatics (n=19) (geometric mean C5: 58.3 µM, 95% CI 24.1 to 141.5 vs 193.6 µM, 82.2 to 456.0; p<0.05). NEA tended to also have greater capsaicin sensitivity than EA, with the difference in capsaicin sensitivity between NEA and EA being of similar magnitude (58.3 µM, 24.1 to 141.5 vs 191.0 µM, 70.9 to 514.0) to that observed between NEA and non-asthmatics; however, this did not reach statistical significance (p=0.07). FEV1 was significantly reduced from baseline following capsaicin inhalation in both asthmatics and non-asthmatics but no differences were found between subgroups. No associations with capsaicin sensitivity and atopy, sputum eosinophils, blood eosinophils, asthma control or treatment were observed. CONCLUSION: NEA, but not EA, showed enhanced capsaicin sensitivity compared with non-asthmatics. Sensory nerve reactivity may therefore play an important role in the pathophysiology of NEA

    Howard S. Becker is one of the foremost sociologists of the second half of the twentieth century. Although he is perhaps best known for research on deviance and his book Outsiders, this constitutes only a very small fraction of his earliest work. This interview looks at some of the continuities and cores of his work over ?fifty years. Becker highlights how his work maintains the same core concerns, although new interests have been added over time. At the core is a concern with 'work' and 'doing things together.' Becker provides many concrete stories from the past and also raises issues about the nature of doing theory and research, how he writes and produces his studies, and the problems attached to the professionalization of sociology. His writing on art and culture can be seen as assuming a major position in his later work, but he does not identify with either postmodernism or cultural studies
