So the developed model will integrate the patients' medical records with their daily measurements of physiological parameters (including the last checkup records) by making use of ARM processor and RS 232 etc. This can be used as a diagnosis reference for physicians, reducing the time required to modify prescriptions and enabling the nursing staff to fully understand the patients' physiological conditions. By using video conferencing, healthcare practitioners and patients can reduce the costs associated with regular office visits. To provide remote patient monitoring in which electronic devices will transmit patient health information to doctor’s computer and android mobile, enabling them to monitor the patient's condition without being physically present near to the patient's bed

    Fissure in ano presents with features like severe pain in anus during and after defecation, constipation, stools streaked with blood. This is challenge to treat due to its nature of recurrence. The condition demands innovative techniques for its management. Many techniques are tried in its management, each by no means better than the other. The condition can be correlated with Parikartika according to Ayurveda. Ayurvedic treatment is beneficial in such cases which includes conservative management like Deepana, Pachana, Vatanulomana and Basti Karma and local application of Madhura-Sheetha-Snighdha Dravyas, Taila Poorana, Lepa and Pichudharana. A case study of patient having features of Parikartika was selected from OPD and Karpoora Ghrita Pichu was done for 7 days. Assessment was done on Pain, per rectal bleeding, constipation, sphincter tone and burning sensation. Significant improvement was observed after treatment

    The role of BRCA1 in breast cancer metastasis is a less explored area that might have importance in increased aggressiveness of BRCA1 defective triple negative cancers. The possible influence of BRCA1 on apico basal polarity and ezrin, radixin and meosin (ERM) proteins are discussed in this review as a reason for cell metastasis. This might help in developing antimetastatic drugs that could help for better prognosis in BRCA1 defective breast cancers

    Cytotoxicity of naphthoquinones and their capacity to generate reactive oxygen species is quenched when conjugated with gold nanoparticles

    Several reports have demonstrated the anticancer activities of plumbagin, a naphthoquinone derivative isolated from plants belonging to Plumbaginaceae family. However, to the best of our knowledge, there are no reports which describe gold nanoconjugation with plumbagin, even though plumbagin is considered to be a promising therapeutic agent. In this report, we demonstrate the fabrication and characterization of gold nanoparticles conjugated with plumbagin (AuPB) that can reduce the toxicity of the latter, and their capacity for cellular localization and generation of reactive oxygen species. The anticancer activity and ability of plumbagin to produce reactive oxygen species was studied and compared with that of bromoderivatives of 1,4 naphthoquinones such as 2-bromo-1,4-naphthoquinone (2-BNQ) and 2,3-dibromo-1, 4-naphthoquinone (2,3-DBNQ) and their gold nanoconjugates. Plumbagin and bromoderivatives of 1,4 naphthoquinones in the form of gold nanoconjugates showed reduced cytotoxicity and apoptosis compared with the pristine compounds, ie, plumbagin, 2-BNQ, and 2,3-DBNQ. Interestingly, we observed that the gold nanoparticles could quench the reactive oxygen species-generating capacity of plumbagin, 2-BNQ, and 2,3-BNQ, which is one of the main mechanisms of action of the naphthoquinones. Therefore, it can be concluded that conjugation with gold nanoparticles can reduce the cytotoxicity of these compounds

    Antistress and antimicrobial studies of biphenyl chalcone derivatives

    This work reports the synthesis, characterization of new (2E)-1-(biphenyl-4-yl)-3-aryl)-prop-2-en-1-one derivatives (C1-C10). The chalcone derivatives were evaluated for in vitro antibacterial, antifungal, antioxidant activities. The compounds exhibited moderate to good antimicrobial activity at MIC 10-40 μg/mL. The compounds were docked at the active site of the methionyl-tRNA synthetase (metRS) (PDB ID: 1A8H) to predict their putative interactions. The compounds further screened for antioxidant property, amid C5, C7 and C8 exhibited good DPPH radical scavenging activity. Compound C5 was selected for antistress studies against gamma radiation induced oxidative stress markers in E .coli K 12. The C5 pretreatment and irradiated bacteria sample showed modulatory action of stress enzymes SOD and CAT to near basal level and significant demulating effect on the level of TBARS.Keywords: Biphenyl chalcone derivatives, radioprotection, antioxidant activity, antimicrobial activity, molecular dockin

    APOPTOSIS INDUCTION OF CENTELLA ASIATICA ON HUMAN BREAST CANCER CELLS

    The present study evaluated the ability of methanolic extract of Centella asiatica (Linn) Urban (Umbelliferae) to induce apoptosis in different cancer cell lines. MCF-7 cells emerged as the most sensitive cell line for in vitro growth inhibitory activity. C. asiatica extract induced apoptosis in MCF-7 cells as indicated by nuclear condensation, increased annexin staining, loss of mitochondrial membrane potential and induction of DNA breaks identified by TUNEL reactivity. It is possible that the use of C. asiatica extract as a component in herbal medicines could be justifiable

    Defective Signalling of the BRCA1 Neighbouring gene, NBR2, leads to ER-? Negative tumours in breast cancer xenograft mouse models

    Majority of breast and ovarian cancer treatment modalities are based on endocrine therapies like antiestrogens which are dependent on the hormone receptor status of tumours. Although BRCA1 is a major regulator of Estrogen Receptor (ER-α), BRCA1 mutations are largely limited to hereditary cases. Here, we show the role of NBR2 (neighbour of BRCA1) in regulating ER-α. We demonstrate a positive correlation between NBR2 & ESR1 in TCGA datasets. Further, the study revealed that shRNA-mediated knockdown of NBR2 led to the downregulation of ER-α in breast cancer cells, MCF7. Finally, the downregulation of ER-α in NBR2 knockdown xenograft tumours (in female NSG mice), which showed higher invasive properties than wild type tumours was demonstrated. Thus, we concluded that in ER-α negative tumours with NBR2 deficiency, biguanides such as metformin and phenformin, which are reported to have a better efficacy under NBR2 deficient conditions, could serve as more suitable alternatives to antiestrogens

    Association of dopamine receptor polymorphisms with schizophrenia and antipsychotic response in a South Indian population

    <p>Abstract</p> <p>Background</p> <p>Alterations in the dopamine transmission and receptor density are hypothesized in the pathophysiology of schizophrenia but ethnic disparities are reported to exist in disease association and therapeutic response to psychotropic medication. Antipsychotics have higher binding affinity to D2 subtype of dopamine receptor. DRD2 Cys311, TaqIB1 and TaqIA1 variants are considered to have either reduced affinity for dopamine and hypo-dopaminergic activity.</p> <p>Methods</p> <p>We examined the role of Taq1B, Taq1D, S311C, H313H and Taq1A polymorphisms of DRD2 gene in schizophrenia and antipsychotic treatment response in 213 patients and 196 controls from a homogenous South Indian population. A more detailed genotype phenotype association analysis was carried out to understand the disease in terms of its socio-cultural factors.</p> <p>Results</p> <p>H313HTT genotype was found to be associated with schizophrenia (P = 0.004) while TaqIB1B1 genotype was significantly associated with higher psychopathology score. When treatment response was considered H313HCC, TaqIA2A2 and Taq1D1D1 had higher mean improvement scores. TaqID1D1 and H313HTT genotype were found to be significantly higher in responders than in nonresponder group. Distinct shift in the LD patterns of responder and non-responder group was observed. Certain symptoms were characteristic of our patient population. Following medication the scores and presentation of these symptoms tend to vary in the responder and non-responder groups.</p> <p>Conclusion</p> <p>Based on genotype phenotype correlations it can be suggested that certain polymorphisms can be defined for their critical functions in disease and their role in treatment response in South Indian population. The present study suggests that in addition to ethnic bias, socio-cultural factors should also be considered while evaluating genotype phenotype correlations, in association and treatment response to complex disorders like schizophrenia.</p

    Monitoring and forecasting of intensive convective precipitation with the use of the mobile meteorological radar (MMR50)

    This paper is focused on current possibilities of the measurement and predictions of intense convective precipitation through the mobile meteorological radar (MMR50). This meteorological radar equipment is part of the Information, Notification and Warning system of the Zlin Region in the Czech Republic, which consists of information and communication infrastructure for dealing with extraordinary events. The first chapter describes basic principles of radar precipitation measurement, e.g. radar estimate of rainfall intensity and radar products. The second chapter presents a methodology of measuring and predicting of intense convective precipitation using the mobile meteorological radar (MMR50), including other possibilities of precipitation forecast as NWP models, aerological, satellite, station measurements, statistics of historical situations, the risk of flash floods on the degree of soils saturation and the possibility of observation of dangerous accompanying phenomena. The last chapter deals with the verification of the principles of radar measurements and forecasts in a case study on 24th July 2015. Torrential rainfall in a combination with hail and strong wind gusts caused heavy flooding in the central part of Zlin region, which caused considerable material damage. Timely and quality information about the current and future formation and development of intense convective precipitation is essential for flood prevention measures.     The ‘peri-urban turn’:A systems thinking approach for a paradigm shift in reconceptualising urban-rural futures in the global South

    With the rapid pace of urbanization, urban sprawl has become a prevalent phenomenon, particularly in the global South, leading to the emergence of peri-urban spaces where rural-urban interfaces occur. These peri-urban areas exhibit dynamic and continuous interactions among social, economic, and environmental systems, offering valuable insights for fostering resilient futures. However, this aspect remains largely unexplored in current research due to a lack of innovative methodological approaches that effectively capture the complementarities, potentialities, and contestations inherent in the dynamics of peri-urban areas. We contend that peri-urbanisation needs to be reconceptualized as an alternative socio-spatial framework that extends the predominantly Eurocentric discourse on counterurbanisation, making it more inclusive of the emerging urban-rural transformations in the global South. By doing so, we can better understand and address the complex dynamics and challenges associated with peri-urban areas and develop strategies to foster resilience in these contexts
