    Pubertal timing in girls and depression: A systematic review

    Background: Because the incidence of depression increases after puberty, it is possible that pubertal timing in girls influences the onset of depression. Our objective was to assess the effect of early and late puberty in girls on the incidence of depression. Methods: We systematically searched relevant databases for controlled studies that assessed the impact of pubertal timing in girls on the incidence of depression or depressive symptoms. The last search was completed in August 2013. Two authors selected the studies, extracted the data, and assessed the quality of the evidence. Meta-analyses of the adjusted and unadjusted results were calculated using random effects. Results: Four cohort studies were included (n=8055 participants). Early puberty significantly increased the risk of new cases of depression in the unadjusted meta-analysis (RR=1.33; CI 95%: 1.02, 1.73) but not in the adjusted estimate of two of the included studies (RR=1.48; CI 95%: 0.69, 2.28). For late puberty, no significant associations were found (unadjusted RR=1.28; CI 95%: 0.87, 1.88). Two studies assessed the effect of early puberty on depressive symptoms and found positive associations. The quality of the available evidence was rated as very low. Limitations: The polled results had wide confidence intervals, and the available evidence was of very low quality. Conclusions: The available evidence supports little confidence regarding the impact of pubertal timing on the onset of depression in girls but suggests that early puberty in girls may increase the risk of depression. Further higher quality studies are needed to clarify the association between pubertal timing and the incidence of depression in girls and women

    Molecular diversity and the fate of biochemical fractions of eucalypt tissues in soil

    The molecular diversity of the source substrate has been regarded as a significant controller of the proportion of plant material that is either mineralized or incorporated into soil organic matter (SOM). However, quantitative parameters to express substrate molecular diversity remain elusive. In this research, we fractionated leaves, twigs, bark, and root tissues of 13C-enriched eucalypt seedlings into hot water extractables (HWE), total solvent (acetone) extractables (TSE), a cellulosic fraction (CF), and the acid unhydrolyzable residue (AUR). We used 13C NMR spectroscopy to obtain a molecular diversity index (MDI) based on the relative abundance of carbohydrate, protein, lignin, lipid, and carbonyl functional groups within the biochemical fractions. Subsequently, we obtained artificial plant organs containing fixed proportions (25%) of their respective biochemical fractions to be incubated with soil material obtained from a Haplic Ferralsol for 200-days, under controlled temperature (25 ± 1 ◦C) and moisture adjusted to 70–80% of the soil water holding capacity. Our experimental design was a randomized complete block design, arranged according to a factorial scheme including 4 plant organs, 4 biochemical fractions, and 3 blocks as replicates. During the incubation, we assessed the evolution of CO2 from the microcosms after 1, 2, 3, 4, 7, 10, 13, 21, 28, 38, 45, 70, 80, 92, 112, 148, 178 and 200 days from the start of the incubation. After the incubation, soil subsamples were submitted to a density fractionation to separate the light fraction of SOM (LFOM) i.e., with density <1.8 g cm 3. The heavy fraction remaining was submitted to wetsieving yielding the sand-sized SOM (SSOM) and the mineral-associated SOM (MAOM), with particle-size greater and smaller than 53 μm, respectively. We found that HWE and AUR exhibited comparatively higher MDIs than the TSE and CF. During the incubation, HWE and CF were the primary sources of 13C-CO2 from all plant organs and after 92 days, the respiration of the TSE of bark and roots increased. Otherwise, the AUR contributed the least for the release of 13C-CO2. There were no significant relationships between the MDI and the amount of 13C transferred into the LFOM or SSOM. Otherwise, the transfer of 13C into the MAOM increased as a linear-quadratic function of MDI, which in turn was negatively correlated with the total 13C-CO2 loss. Overall, the MDI exerted a stronger control on the 13C-labeled MAOM than on 13C-CO2 emissions, highlighting the need to improve our ability to distinguish and quantify direct plant inputs from those of microbial origin entering soil C pools.publishedVersio

    Color Trails Test: a Brazilian normative sample

    The present study aims to evaluate psychometric data on the population of Brazil using of the Color Trails Test (CTT). The instrument was applied to 1,942 subjects who came from the north (1.4%), northeast (10.4%), midwest (7.7%), southeast (65.9%), and south (13.9%) regions of Brazil. Ages varied from 18 to 86 years (M = 40.03 years; DP = 16.02), with 386 males (19.9%) and 1,556 females (80.1%). Regarding schooling, 57 (3%) of participants were illiterate, 1.240 (64%) had primary education and 645 (33%) had a high school and/or superior diploma. The length of schooling ranged from 0 to 14 (M = 7.81, SD = 3.51) years. Good accuracy ratings were obtained using Spearman's coefficient on the main measures of the CTT, including time of execution form 1 (0.76) and form 2 (0.82). Regional variations showed that the northeast region had the worst results on the instrument. Males had better performance on the proposed task, with significant differences between age groups and the worst results for ages greater than 60 years. For both sexes, an increase in score was noticed with increasing age, and better performance was found with increasing levels of education. The present results suggest that the CTT can identify attentional disturbances with significant variations in the time of execution

    Updated Clinical Classification of Pulmonary Hypertension

    In 1998, a clinical classification of pulmonary hypertension (PH) was established, categorizing PH into groups which share similar pathological and hemodynamic characteristics and therapeutic approaches. During the 5th World Symposium held in Nice, France, in 2013, the consensus was reached to maintain the general scheme of previous clinical classifications. However, modifications and updates especially for Group 1 patients (pulmonary arterial hypertension [PAH]) were proposed. The main change was to withdraw persistent pulmonary hypertension of the newborn (PPHN) from Group 1 because this entity carries more differences than similarities with other PAH subgroups. In the current classification, PPHN is now designated number 1. Pulmonary hypertension associated with chronic hemolytic anemia has been moved from Group 1 PAH to Group 5, unclear/multifactorial mechanism. In addition, it was decided to add specific items related to pediatric pulmonary hypertension in order to create a comprehensive, common classification for both adults and children. Therefore, congenital or acquired left-heart inflow/outflow obstructive lesions and congenital cardiomyopathies have been added to Group 2, and segmental pulmonary hypertension has been added to Group 5. Last, there were no changes for Groups 2, 3, and 4

    Management Effect on Rainfall Use Efficiency and Growth Rate of Spineless Cactus \u3cem\u3ecv.\u3c/em\u3e Miúda in Northeast Brazil

    Spineless cactus has a high rainfall use efficiency. Under water deficit, they open their stomata during the night to capture atmospheric carbon dioxide when the air temperature is lower and relative humidity higher (Taiz and Zeiger, 2009), losing less water to the atmosphere. The rainfall use efficiency (RUE) can be expressed as the amount of water used by the crop to produce dry matter (Han and Felker, 1997). The RUE showed by spineless cactus, a CAM plant, is three to five times greater than the one observed in C3 and C4 plants (Felker and Russel, 1988; Nobel, 1991). Therefore, spineless cacti (Nopaleae opuntia) are important forage resource in northeast Brazil. Management practices, however, can affect the RUE and growth of spineless cactus, especially organic and mineral fertilization. Thus, the objective of this research was to evaluate the effect of organic and nitrogen fertilization and harvest frequency on rainfall use efficiency and growth rate of spineless cactus cv.     Integration of miRNA and mRNA expression profles reveals microRNA-regulated networks during muscle wasting in cardiac cachexia

    Cardiac cachexia (CC) is a common complication of heart failure (HF) associated with muscle wasting and poor patient prognosis. Although different mechanisms have been proposed to explain muscle wasting during CC, its pathogenesis is still not understood. Here, we described an integrative analysis between miRNA and mRNA expression profiles of muscle wasting during CC. Global gene expression profiling identified 1,281 genes and 19 miRNAs differentially expressed in muscle wasting during CC. Several of these deregulated genes are known or putative targets of the altered miRNAs, including miR-29a-3p, miR-29b-3p, miR-210-5p, miR-214, and miR-489. Gene ontology analysis on integrative mRNA/miRNA expression profiling data revealed miRNA interactions affecting genes that regulate extra-cellular matrix (ECM) organization, proteasome protein degradation, citric acid cycle and respiratory electron transport. We further identified 11 miRNAs, including miR-29a-3p and miR-29b-3p, which target 21 transcripts encoding the collagen proteins related to ECM organization. Integrative miRNA and mRNA global expression data allowed us to identify miRNA target genes involved in skeletal muscle wasting in CC. Our functional experiments in C2C12 cells confirmed that miR-29b down-regulates collagen genes and contributes to muscle cell atrophy. Collectively, our results suggest that key ECM-associated miRNAs and their target genes may contribute to CC in HF

    Early liver function improvement following successful treatment of chronic hepatitis C in patients with decompensated cirrhosis: a real-life study

    OBJECTIVES: Despite higher rates of sustained virologic response (SVR), important concerns remain when patients with decompensated cirrhosis due to hepatitis C virus (HCV) are treated with direct-acting antiviral agents (DAA). Questions include efficacy, safety, and the magnitude of liver function improvement. Here, we aimed to evaluate HCV treatment data in this specific population in Brazil. METHODS: We included 85 patients with decompensated cirrhosis submitted to HCV therapy with DAA followed at two academic tertiary centers in the southeastern region of Brazil. RESULTS: Seventy-nine patients (92.9%) were Child-Pugh (CP) score B, and six (7.1%) were CP score C. The mean MELD score was 12.86. The most common treatment was sofosbuvir plus daclatasvir±ribavirin for 24 weeks. The overall intention-to-treat (ITT) SVR rate was 87.4% (74/85) and modified-ITT 96.1% (74/77). ITT SVR was associated with lower baseline INR values (p=0.029). Adverse events (AE) occurred in 57.9% (44/76) of patients. Serious AE were reported in 12.8% (10/78), and were related to the presence of hepatic encephalopathy (p=0.027). SVR was associated with improvement in CP (p&lt;0.0001) and MELD scores (p=0.021). Among baseline CP score B patients with SVR, 46% (29/63) regressed to CP score A. Ascites was independently associated with no improvement in liver function in patients who achieved SVR (p=0.001; OR:39.285; 95% CI:4.301-258.832). CONCLUSIONS: Patients with decompensated HCV cirrhosis showed a high SVR rate with interferon-free therapy. Early liver function improvement occurred after successful HCV eradication. However, long-term follow-up of these patients after SVR remains strongly advised. &nbsp

    Preliminarno ispitivanje antimikotskog i citotoksičnog djelovanja derivata cikloalkil[b]tiofena PLS-DA analizom

    A series of 2-[(arylidene)amino]-cycloalkyl[b]thiophene-3-carbonitriles (2a-x) was synthesized by incorporation of substituted aromatic aldehydes in Gewald adducts (1a-c). The title compounds were screened for their antifungal activity against Candida krusei and Criptococcusneoformans and for their antiproliferative activity against a panel of 3 human cancer cell lines (HT29, NCI H-292 and HEP). Forantiproliferative activity, the partial least squares (PLS) methodology was applied. Some of the prepared compounds exhibited promising antifungal and proliferative properties. The most active compounds for antifungal activity were cyclohexyl[b]thiophene derivatives, and forantiproliferative activity cycloheptyl[b]thiophene derivatives, especially 2-[(1H-indol-2-yl-methylidene)amino]-5,6,7,8-tetrahydro-4H-cyclohepta[b]thiophene-3-carbonitrile (2r), which inhibited more than 97 % growth of the three cell lines. The PLS discriminant analysis (PLS-DA)applied generated good exploratory and predictive results and showed that the descriptors having shape characteristics were strongly correlated with the biological data.Koristeći supstituirane aromatske aldehide u Gewaldovim aduktima 1a-c sintetizirani su derivati 2-[(ariliden)amino]-cikloalkil[b]tiofen-3-karbonitrila (2a-x). Ispitano je antimikotsko djelovanje tih spojeva na gljivice Candida krusei i Criptococcus neoformans te antiproliferativno djelovanje na tri humane tumorske stanične linije (HT29, NCI H-292 i HEP). Za antiproliferativno djelovanje primijenjena je metoda parcijalnih najmanjih kvadrata (PLS) koristeći softverski program Pentacle. Neki od ispitanih spojeva pokazuju obećavajuće antimikotsko i antiproliferativno djelovanje. Najjače antimikotsko djelovanje imaju cikloheksil[b]tiofen derivati, a najjače antiproliferativno djelovanje cikloheptil[b]tiofen derivati, posebice 2-[(1H-indol-2-il-metiliden)amino]-5,6,7,8-tetrahidro-4H-ciklohepta[b]tiofen-3-karbonitril (2r), koji inhibira više od 97 % rast svih triju ispitivanih staničnih linija. Primijenjena PLS diskriminirajuća analiza dala je dobre istraživačke i prognostičke rezultate i pokazala da deskriptori dobro koreliraju s biološkim rezultatima

    Clonal expansion and epigenetic reprogramming following deletion or amplification of mutant

    IDH1 mutation is the earliest genetic alteration in low-grade gliomas (LGGs), but its role in tumor recurrence is unclear. Mutant IDH1 drives overproduction of the oncometabolite d-2-hydroxyglutarate (2HG) and a CpG island (CGI) hypermethylation phenotype (G-CIMP). To investigate the role of mutant IDH1 at recurrence, we performed a longitudinal analysis of 50 IDH1 mutant LGGs. We discovered six cases with copy number alterations (CNAs) at the IDH1 locus at recurrence. Deletion or amplification of IDH1 was followed by clonal expansion and recurrence at a higher grade. Successful cultures derived from IDH1 mutant, but not IDH1 wild type, gliomas systematically deleted IDH1 in vitro and in vivo, further suggestive of selection against the heterozygous mutant state as tumors progress. Tumors and cultures with IDH1 CNA had decreased 2HG, maintenance of G-CIMP, and DNA methylation reprogramming outside CGI. Thus, while IDH1 mutation initiates gliomagenesis, in some patients mutant IDH1 and 2HG are not required for later clonal expansions
