    A review of data on abundance, trends in abundance, habitat use and diet of ice-breeding seals in the Southern Ocean

    The development of models of marine ecosystems in the Southern Ocean is becoming increasingly important as a means of understanding and managing impacts such as exploitation and climate change. Collating data from disparate sources, and understanding biases or uncertainties inherent in those data, are important first steps for improving ecosystem models. This review focuses on seals that breed in ice habitats of the Southern Ocean (i.e. crabeater seal, Lobodon carcinophaga; Ross seal, Ommatophoca rossii; leopard seal, Hydrurga leptonyx; and Weddell seal, Leptonychotes weddellii). Data on populations (abundance and trends in abundance), distribution and habitat use (movement, key habitat and environmental features) and foraging (diet) are summarised, and potential biases and uncertainties inherent in those data are identified and discussed. Spatial and temporal gaps in knowledge of the populations, habitats and diet of each species are also identified

    Intrabodies Binding the Proline-Rich Domains of Mutant Huntingtin Increase Its Turnover and Reduce Neurotoxicity

    Although expanded polyglutamine (polyQ) repeats are inherently toxic, causing at least nine neurodegenerative diseases, the protein context determines which neurons are affected. The polyQ expansion that causes Huntington's disease (HD) is in the first exon (HDx-1) of huntingtin (Htt). However, other parts of the protein, including the 17 N-terminal amino acids and two proline (polyP) repeat domains, regulate the toxicity of mutant Htt. The role of the P-rich domain that is flanked by the polyP domains has not been explored. Using highly specific intracellular antibodies (intrabodies), we tested various epitopes for their roles in HDx-1 toxicity, aggregation, localization, and turnover. Three domains in the P-rich region (PRR) of HDx-1 are defined by intrabodies: MW7 binds the two polyP domains, and Happ1 and Happ3, two new intrabodies, bind the unique, P-rich epitope located between the two polyP epitopes. We find that the PRR-binding intrabodies, as well as VL12.3, which binds the N-terminal 17 aa, decrease the toxicity and aggregation of HDx-1, but they do so by different mechanisms. The PRR-binding intrabodies have no effect on Htt localization, but they cause a significant increase in the turnover rate of mutant Htt, which VL12.3 does not change. In contrast, expression of VL12.3 increases nuclear Htt. We propose that the PRR of mutant Htt regulates its stability, and that compromising this pathogenic epitope by intrabody binding represents a novel therapeutic strategy for treating HD. We also note that intrabody binding represents a powerful tool for determining the function of protein epitopes in living cells

    Perturbation with Intrabodies Reveals That Calpain Cleavage Is Required for Degradation of Huntingtin Exon 1

    Background: Proteolytic processing of mutant huntingtin (mHtt), the protein that causes Huntington's disease (HD), is critical for mHtt toxicity and disease progression. mHtt contains several caspase and calpain cleavage sites that generate N-terminal fragments that are more toxic than full-length mHtt. Further processing is then required for the degradation of these fragments, which in turn, reduces toxicity. This unknown, secondary degradative process represents a promising therapeutic target for HD. Methodology/Principal Findings: We have used intrabodies, intracellularly expressed antibody fragments, to gain insight into the mechanism of mutant huntingtin exon 1 (mHDx-1) clearance. Happ1, an intrabody recognizing the proline-rich region of mHDx-1, reduces the level of soluble mHDx-1 by increasing clearance. While proteasome and macroautophagy inhibitors reduce turnover of mHDx-1, Happ1 is still able to reduce mHDx-1 under these conditions, indicating Happ1-accelerated mHDx-1 clearance does not rely on these processes. In contrast, a calpain inhibitor or an inhibitor of lysosomal pH block Happ1-mediated acceleration of mHDx-1 clearance. These results suggest that mHDx-1 is cleaved by calpain, likely followed by lysosomal degradation and this process regulates the turnover rate of mHDx-1. Sequence analysis identifies amino acid (AA) 15 as a potential calpain cleavage site. Calpain cleavage of recombinant mHDx-1 in vitro yields fragments of sizes corresponding to this prediction. Moreover, when the site is blocked by binding of another intrabody, V_L12.3, turnover of soluble mHDx-1 in living cells is blocked. Conclusions/Significance: These results indicate that calpain-mediated removal of the 15 N-terminal AAs is required for the degradation of mHDx-1, a finding that may have therapeutic implications

    Detection of phase singularities with a Shack-Hartmann wavefront sensor

    While adaptive optical systems are able to remove moderate wavefront distortions in scintillated optical beams, phase singularities that appear in strongly scintillated beams can severely degrade the performance of such an adaptive optical system. Therefore, the detection of these phase singularities is an important aspect of strong scintillation adaptive optics. We investigate the detection of phase singularities with the aid of a Shack-Hartmann wavefront sensor and show that, in spite of some systematical deficiencies inherent to the Shack-Hartmann wavefront sensor, it can be used for the reliable detection of phase singularities, irrespective of their morphologies. We provide full analytical results, together with numerical simulations of the detection process.Comment: 23 pages, 9 figure

    T Pyxidis: The First Cataclysmic Variable with a Collimated Jet

    We present the first observational evidence for a collimated jet in a cataclysmic variable system; the recurrent nova T Pyxidis. Optical spectra show bipolar components of Hα\alpha with velocities 1400km/s\sim 1400 km/s, very similar to those observed in the supersoft X-ray sources and in SS 433. We argue that a key ingredient of the formation of jets in the supersoft X-ray sources and T Pyx (in addition to an accretion disk threaded by a vertical magnetic field), is the presence of nuclear burning on the surface of the white dwarf.Comment: 10 pages 2 figures to appear in ApJ Letter

    Jacques Rancière es un filósofo francés nacido en Argel en 1940. En su juventud, integró el equipo de intelectuales dirigidos por Louis Althusser en la Escuela Normal Superior de París. En la década de 1960, Althusser tenía gran protagonismo en el ámbito político e intelectual de Francia, e impulsó una serie de presentaciones sobre marxismo que luego confluirían en un volumen colectivo en el que Rancière colaboró (Para leer El Capital, 1965). Sin embargo, a partir de las experiencias vividas en Mayo de 1968, Rancière se distanció de Althusser a partir de entender que la distinción althusseriana entre “ciencia” e “ideología” terminaba por instituir, para Rancière, un monopolio intelectual del saber que establecía una superioridad de la ciencia por sobre los saberes prácticos de las masas (La lección de Althusser, 1974).Facultad de Humanidades y Ciencias de la Educació

    Ciencia y penitencia: Dictadura, pedagogías restrictivas y formación de pedagogos : El caso de la Universidad Nacional de La Plata

    Como es analizado habitualmente, el proceso autoritario que vivió la Argentina con la dictadura militar de 1976-1983, autodenominado «proceso de reorganización nacional» (en adelante PRN) fue el más cruento de la historia argentina. Esa dictadura fue un punto de inflexión en muchas dimensiones de lo social: fue el momento en que el terrorismo de estado adquirió su máxima expresión luego de diversas experiencias en las que se había instalado el uso ilegítimo de la fuerza estatal, fue también una expresión extrema de la violencia política, la clausura de una enorme cantidad de movimientos, fenómenos y expresiones sociales de diversa índole. Esta caracterización como punto de inflexión es también muy clara en el ámbito educacional. En ese sentido, se observó una profunda transformación en el discurso que sostuvo a las instituciones educativas durante la intervención dictatorial, por eso partiremos de entender a la dictadura como una dislocación del sistema de significaciones previas (Southwell, 2002). Desde ese punto de partida, el propósito de este capítulo será explorar brevemente el campo pedagógico universitario platense previo a la dictadura militar, y luego avanzaremos en el análisis de los aspectos de la formación de pedagogos sobre los que intervino el régimen en la UNLP para desmantelar esas expresiones pedagógicas del ’70.Facultad de Humanidades y Ciencias de la Educació
